    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

    • The Principle of Maximum Conformality (PMC) eliminates QCD renormalization scale-setting uncertainties using fundamental renormalization group methods. The resulting scale-fixed pQCD predictions are independent of the choice of renormalization scheme and show rapid convergence. The coefficients of the scale-fixed couplings are identical to the corresponding conformal series with zero beta-function. Two all-orders methods for systematically implementing the PMC-scale setting procedure for existing high order calculations are discussed in this article. One implementation is based on the PMC-BLM correspondence (PMC-I); the other, more recent, method (PMC-II) uses the R-delta-scheme, a systematic generalization of the minimal subtraction renormalization scheme. Both approaches satisfy all of the principles of the renormalization group and lead to scale-fixed and scheme-independent predictions at each finite order. In this work, we show that PMC-I and PMC-II scale-setting methods are in practice equivalent to each other. We illustrate this equivalence for the four-loop calculations of the annihilation ratio Re+e- and the Higgs partial width Gamma(H -> b (b) over bar). Both methods lead to the same resummed ('conformal') series up to all orders. The small scale differences between the two approaches are reduced as additional renormalization group {beta(i)}-terms in the pQCD expansion are taken into account. We also show that special degeneracy relations, which underly the equivalence of the two PMC approaches and the resulting conformal features of the pQCD series, are in fact general properties of non-Abelian gauge theory.     • Proton-exchange-membrane fuel cells (PEMFCs) are attractive next-generation power sources for use in vehicles and other applications(1), with development efforts focusing on improving the catalyst system of the fuel cell. One problem is catalyst poisoning by impurity gases such as carbon monoxide (CO), which typically comprises about one per cent of hydrogen fuel(2-4). A possible solution is on-board hydrogen purification, which involves preferential oxidation of CO in hydrogen (PROX)(3-7). However, this approach is challenging(8-15) because the catalyst needs to be active and selective towards CO oxidation over a broad range of low temperatures so that CO is efficiently removed (to below 50 parts per million) during continuous PEMFC operation (at about 353 kelvin) and, in the case of automotive fuel cells, during frequent cold-start periods. Here we show that atomically dispersed iron hydroxide, selectively deposited on silica-supported platinum (Pt) nanoparticles, enables complete and 100 per cent selective CO removal through the PROX reaction over the broad temperature range of 198 to 380 kelvin. We find that the mass-specific activity of this system is about 30 times higher than that of more conventional catalysts consisting of Pt on iron oxide supports. In situ X-ray absorption fine-structure measurements reveal that most of the iron hydroxide exists as Fe-1(OH)(x) clusters anchored on the Pt nanoparticles, with density functional theory calculations indicating that Fe-1(OH)(x)-Pt single interfacial sites can readily react with CO and facilitate oxygen activation. These findings suggest that in addition to strategies that target oxide-supported precious-metal nanoparticles or isolated metal atoms, the deposition of isolated transition-metal complexes offers new ways of designing highly active metal catalysts.

  • Genome-wide identification of DNA methylation QTLs in whole blood highlights pathways for cardiovascular disease

    • Identifying methylation quantitative trait loci (meQTLs) and integrating them with disease-associated variants from genome-wide association studies (GWAS) may illuminate functional mechanisms underlying genetic variant-disease associations. Here, we perform GWAS of >415 thousand CpG methylation sites in whole blood from 4170 individuals and map 4.7 million cis- and 630 thousand trans-meQTL variants targeting >120 thousand CpGs. Independent replication is performed in 1347 participants from two studies. By linking cis-meQTL variants with GWAS results for cardiovascular disease (CVD) traits, we identify 92 putatively causal CpGs for CVD traits by Mendelian randomization analysis. Further integrating gene expression data reveals evidence of cis CpG-transcript pairs causally linked to CVD. In addition, we identify 22 trans-meQTL hotspots each targeting more than 30 CpGs and find that trans-meQTL hotspots appear to act in cis on expression of nearby transcriptional regulatory genes. Our findings provide a powerful meQTL resource and shed light on DNA methylation involvement in human diseases.

    • Adapting the processor to the target application is essential in the Internet-of-Things (IoT), and thus requires customizability in order to improve energy efficiency and scalability to provide sufficient performance. In this paper, a reconfigurable and scalable control-centric architecture is proposed, and a processor consisting of two cores and an on-chip multi-mode router is implemented. Reconfigurability is enabled by a programmable sequence mapping table (SMT) which reorganizes functional units in each cycle, thus increasing hardware utilization and reducing excessive data movement for high energy efficiency. The router facilitates both wormhole and circuit switching to construct intra- or inter-chip interconnections, providing scalable performance. Fabricated in a 65-nm process, the chip exhibits 101.4 GOPS/W energy efficiency with a die size of 3.5 mm(2). The processor carries out general-purpose processing with a code size 29% smaller than the ARM Cortex M4, and improves the performance of application-specific processing by over ten times when implementing AES and RSA using SMTs instead of general-purpose C. By utilizing the on-chip router, the processor can be interconnected up to 256 nodes, with a single link bandwidth of 1.4 Gbps.

    • This paper presents a SoC design that combines general purpose control and application-specific acceleration within a reconfigurable ASIP core for Internet-of-Things applications. Sufficient processing capability and re-configurability are provided by highly customizable data path and efficient sequence control loop. By fully utilizing the data path of proposed architecture, the processor significantly reduces >4X code size and offers superior performance compared with MSP430 and Atmega128 in FIR and Whetstone benchmarks. More than 10X speedup can be obtained in executing encryption algorithms by optimized micro-instructions without extra hardware accelerators. Fabricated in 0.18 μm CMOS, our SoC's energy efficiency beats most of the microcontrollers with a value as low as 61 μA/MHz.

    • The ability to tune magnetic orders, such as magnetic anisotropy and topological spin texture, is desired to achieve high-performance spintronic devices. A recent strategy has been to employ interfacial engineering techniques, such as the introduction of spin-correlated interfacial coupling, to tailor magnetic orders and achieve novel magnetic properties. We chose a unique polar-nonpolar LaMnO3/SrIrO3 superlattice because Mn (3d)/Ir (5d) oxides exhibit rich magnetic behaviors and strong spin-orbit coupling through the entanglement of their 3d and 5d electrons. Through magnetization and magnetotransport measurements, we found that the magnetic order is interface-dominated as the superlattice period is decreased. We were able to then effectively modify the magnetization, tilt of the ferromagnetic easy axis, and symmetry transition of the anisotropic magnetoresistance of the LaMnO3/SrIrO3 superlattice by introducing additional Mn (3d) and Ir (5d) interfaces. Further investigations using in-depth first-principles calculations and numerical simulations revealed that these magnetic behaviors could be understood by the 3d/5d electron correlation and Rashba spin-orbit coupling. The results reported here demonstrate a new route to synchronously engineer magnetic properties through the atomic stacking of different electrons, which would contribute to future applications in high-capacity storage devices and advanced computing.

    • Enzyme variants of the plant epoxide hydrolase StEH1 displaying improved stereoselectivities in the catalyzed hydrolysis of (2,3-epoxypropyl)benzene were generated by directed evolution. The evolution was driven by iterative saturation mutagenesis in combination with enzyme activity screenings where product chirality was the decisive selection criterion. Analysis of the underlying causes of the increased diol product ratios revealed two major contributing factors: increased enantioselectivity for the corresponding epoxide enantiomer(s) and, in some cases, a concomitant change in regioselectivity in the catalyzed epoxide ring-opening half-reaction. Thus, variant enzymes that catalyzed the hydrolysis of racemic (2,3-epoxypropyl)benzene into the R-diol product in an enantioconvergent manner were isolated.

    • The ultimate aim of this study is to construct polymer nanocomposite patterns by dip-pen nanolithography (DPN). Recent investigations have revealed the effect of the amount of ink (Laplace pressure) on the mechanism of liquid ink writing. In this study it is shown that not only the amount of ink, but also physisorption and surface diffusion are relevant. After a few writing steps, physisorption and surface diffusion outweigh the influence of the amount of ink, allowing consistent patterning governed by dwell times and writing speeds. Polymer matrices can be utilized as a delivery medium to deposit functional particles. DPN patterning of polymer nanocomposites allows for local tuning of the functionality and mechanical strength of the written patterns in high resolution, with the benefit of pattern flexibility. Typically polymer matrices with volatile components are used as a delivery medium for nanoparticle deposition, with subsequent removal of loosely bound matrix material by heating or oxygen plasma. In our study, nanocomposite patterns were constructed, and the differences between polymer and nanocomposite patterning were investigated. Cross-sectional SEM and TEM analysis confirmed that nanoparticles can be deposited with the liquid-polymer ink and are evenly distributed in the polymer matrix.
