  • Top 10 priorities for future infertility research: an international consensus development study

    • STUDY QUESTION: Can the priorities for future research in infertility be identified? SUMMARY ANSWER: The top 10 research priorities for the four areas of male infertility, female and unexplained infertility, medically assisted reproduction and ethics, access and organization of care for people with fertility problems were identified. WHAT IS KNOWN ALREADY: Many fundamental questions regarding the prevention, management and consequences of infertility remain unanswered. This is a barrier to improving the care received by those people with fertility problems. STUDY DESIGN, SIZE, DURATION: Potential research questions were collated from an initial international survey, a systematic review of clinical practice guidelines and Cochrane systematic reviews. A rationalized list of confirmed research uncertainties was prioritized in an interim international survey. Prioritized research uncertainties were discussed during a consensus development meeting. Using a formal consensus development method, the modified nominal group technique, diverse stakeholders identified the top 10 research priorities for each of the categories male infertility, female and unexplained infertility, medically assisted reproduction and ethics, access and organization of care. PARTICIPANTS/MATERIALS, SETTING, METHODS: Healthcare professionals, people with fertility problems and others (healthcare funders, healthcare providers, healthcare regulators, research funding bodies and researchers) were brought together in an open and transparent process using formal consensus methods advocated by the James Lind Alliance. MAIN RESULTS AND THE ROLE OF CHANCE: The initial survey was completed by 388 participants from 40 countries, and 423 potential research questions were submitted. Fourteen clinical practice guidelines and 162 Cochrane systematic reviews identified a further 236 potential research questions. A rationalized list of 231 confirmed research uncertainties was entered into an interim prioritization survey completed by 317 respondents from 43 countries. The top 10 research priorities for each of the four categories male infertility, female and unexplained infertility (including age-related infertility, ovarian cysts, uterine cavity abnormalities and tubal factor infertility), medically assisted reproduction (including ovarian stimulation, IUI and IVF) and ethics, access and organization of care were identified during a consensus development meeting involving 41 participants from I I countries. These research priorities were diverse and seek answers to questions regarding prevention, treatment and the longer-term impact of infertility. They highlight the importance of pursuing research which has often been overlooked, including addressing the emotional and psychological impact of infertility, improving access to fertility treatment, particularly in lower resource settings and securing appropriate regulation. Addressing these priorities will require diverse research methodologies, including laboratory-based science, qualitative and quantitative research and population science. LIMITATIONS, REASONS FOR CAUTION: We used consensus development methods, which have inherent limitations, including the representativeness of the participant sample, methodological decisions informed by professional judgment and arbitrary consensus definitions. WIDER IMPLICATIONS OF THE FINDINGS: We anticipate that identified research priorities, developed to specifically highlight the most pressing clinical needs as perceived by healthcare professionals, people with fertility problems and others, will help research funding organizations and researchers to develop their future research agenda.

    • Alpine grassland vegetation supports globally important biodiversity and ecosystems that are increasingly threatened by climate warming and other environmental changes. Trait-based approaches can support understanding of vegetation responses to global change drivers and consequences for ecosystem functioning. In six sites along a 1314 m elevational gradient in Puna grasslands in the Peruvian Andes, we collected datasets on vascular plant composition, plant functional traits, biomass, ecosystem fluxes, and climate data over three years. The data were collected in the wet and dry season and from plots with different fire histories. We selected traits associated with plant resource use, growth, and life history strategies (leaf area, leaf dry/wet mass, leaf thickness, specific leaf area, leaf dry matter content, leaf C, N, P content, C and N isotopes). The trait dataset contains 3,665 plant records from 145 taxa, 54,036 trait measurements (increasing the trait data coverage of the regional flora by 420%) covering 14 traits and 121 plant taxa (ca. 40% of which have no previous publicly available trait data) across 33 families.

  • Global patterns and drivers of ecosystem functioning in rivers and riparian zones

    • River ecosystems receive and process vast quantities of terrestrial organic carbon, the fate of which depends strongly on microbial activity. Variation in and controls of processing rates, however, are poorly characterized at the global scale. In response, we used a peer-sourced research network and a highly standardized carbon processing assay to conduct a global-scale field experiment in greater than 1000 river and riparian sites. We found that Earth's biomes have distinct carbon processing signatures. Slow processing is evident across latitudes, whereas rapid rates are restricted to lower latitudes. Both the mean rate and variability decline with latitude, suggesting temperature constraints toward the poles and greater roles for other environmental drivers (e.g., nutrient loading) toward the equator. These results and data set the stage for unprecedented "next-generation biomonitoring" by establishing baselines to help quantify environmental impacts to the functioning of ecosystems at a global scale.

    • Food webs are central entities mediating processes and external pressures in marine ecosystems. They are essential to understand and predict ecosystem dynamics and provision of ecosystem services. Paradoxically, utilization of food web knowledge in marine environmental conservation and resource management is limited. To better understand the use of knowledge and barriers to incorporation in management, we assess its application related to the management of eutrophication, chemical contamination, fish stocks, and non-indigenous species. We focus on the Baltic, a severely impacted, but also intensely studied and actively managed semi-enclosed sea. Our assessment shows food web processes playing a central role in all four areas, but application varies strongly, from formalized integration in management decisions, to support in selecting indicators and setting threshold values, to informal knowledge explaining ecosystem dynamics and management performance. Barriers for integration are complexity of involved ecological processes and that management frameworks are not designed to handle such information. We provide a categorization of the multi-faceted uses of food web knowledge and benefits of future incorporation in management, especially moving towards ecosystem-based approaches as guiding principle in present marine policies and directives. We close with perspectives on research needs to support this move considering global and regional change.

  • Impact of Climate Change on Fish Population Dynamics in the Baltic Sea : A Dynamical Downscaling Investigation

    • Understanding how climate change, exploitation and eutrophication will affect populations and ecosystems of the Baltic Sea can be facilitated with models which realistically combine these forcings into common frameworks. Here, we evaluate sensitivity of fish recruitment and population dynamics to past and future environmental forcings provided by three ocean-biogeochemical models of the Baltic Sea. Modeled temperature explained nearly as much variability in reproductive success of sprat (Sprattus sprattus; Clupeidae) as measured temperatures during 1973-2005, and both the spawner biomass and the temperature have influenced recruitment for at least 50 years. The three Baltic Sea models estimate relatively similar developments (increases) in biomass and fishery yield during twenty-first century climate change (ca. 28 % range among models). However, this uncertainty is exceeded by the one associated with the fish population model, and by the source of global climate data used by regional models. Knowledge of processes and biases could reduce these uncertainties.

  • Multi-decadal scale variability in the eastern Baltic cod fishery 1550-1860 - Evidence and causes

    • Identification of periods of high and low cod production, and the reasons for these periods, can increase understanding of variability in populations and ecosystems. In this study we investigate the multi-decadal and multi-century scale variations in the cod population in the eastern Baltic Sea (ICES Subdivisions 25-32). Analytically derived estimates of biomass are available since 1966. These estimates show that biomass increased in the late 1970s-early 1980s, but decreased nearly 10-fold until the early 1990s and is still well below the long-term average. Prior to 1966 the biomass of cod is unknown, as is the relative role of fishing, climate variability/regimes, eutrophication and reduction of marine mammal predator populations. We have begun to investigate whether historical fisheries information (landings, effort, distribution) from before the 1880s is available in Baltic archives and museums, and to what extent this information can be used to interpret variations in this population. We have located fisheries data for different parts of the Baltic for different time periods since the 1550s and have interpreted the findings using current process knowledge of oceanographic mechanisms affecting cod reproduction and ecology in the Baltic Sea. The recovered data show that the Baltic ecosystem was able to support modest-large cod populations even though it was oligotrophic and contained large populations of cod predators (e.g., marine mammals). Current ecosystem management policy in the Baltic as developed and implemented by organisations such as the International Council for the Exploration of the Sea (ICES), the Baltic Marine Environment Protection Commission (HELCOM), the nine coastal countries and the European Union includes recovery of the cod population, a reduction in nutrient loading and measures to promote recovery of seal and harbour porpoise populations. If these policies are successful, the role of predatory fish in the future Baltic could again be substantial and comparable to that which we show existed 450 years ago. However, such a scenario will also require a major reduction in cod fishing mortality and suitable hydrographic conditions which promote successful cod reproduction. Historical ecology investigations in the Baltic can contribute to scientifically based fishery and ecosystem management and recovery plans.

  • Comparing reconstructed past variations and future projections of the Baltic sea ecosystem first results from multi model ensemble simulations

    • Multi-model ensemble simulations for the marine biogeochemistry and food web of the Baltic Sea were performed for the period 1850-2098, and projected changes in the future climate were compared with the past climate environment. For the past period 1850-2006, atmospheric, hydrological and nutrient forcings were reconstructed, based on historical measurements. For the future period 1961-2098, scenario simulations were driven by regionalized global general circulation model (GCM) data and forced by various future greenhouse gas emission and air-and riverborne nutrient load scenarios (ranging from a pessimistic 'business-as-usual' to the most optimistic case). To estimate uncertainties, different models for the various parts of the Earth system were applied. Assuming the IPCC greenhouse gas emission scenarios A1B or A2, we found that water temperatures at the end of this century may be higher and salinities and oxygen concentrations may be lower than ever measured since 1850. There is also a tendency of increased eutrophication in the future, depending on the nutrient load scenario. Although cod biomass is mainly controlled by fishing mortality, climate change together with eutrophication may result in a biomass decline during the latter part of this century, even when combined with lower fishing pressure. Despite considerable shortcomings of state-of-the-art models, this study suggests that the future Baltic Sea ecosystem may unprecedentedly change compared to the past 150 yr. As stakeholders today pay only little attention to adaptation and mitigation strategies, more information is needed to raise public awareness of the possible impacts of climate change on marine ecosystems.

    • GDF15 is a stress-induced hormone acting in the hindbrain that activates neural circuitry involved in establishing aversive responses and reducing food intake and body weight in animal models. Anorexia, weight loss, nausea and vomiting are common manifestations of glucocorticoid deficiency, and we hypothesized that glucocorticoid deficiency may be associated with elevated levels of GDF15.To determine the impact of primary adrenal insufficiency (PAI) and glucocorticoid replacement on circulating GDF15 levels.We measured circulating concentrations of GDF15 in a cohort of healthy volunteers and Addison's disease patients following steroid withdrawal. Significantly higher GDF15 (mean ± standard deviation [SD]) was observed in the Addison's cohort, 739.1 ± 225.8 pg/mL compared to healthy controls, 497.9 ± 167.7 pg/mL (P = 0.01). The effect of hydrocortisone replacement on GDF15 was assessed in 3 independent PAI cohorts with classical congenital adrenal hyperplasia or Addison's disease; intravenous hydrocortisone replacement reduced GDF15 in all groups. We examined the response of GDF15 to increasing doses of glucocorticoid replacement in healthy volunteers with pharmacologically mediated cortisol deficiency. A dose-dependent difference in GDF15 (mean ± SD) was observed between the groups with values of 491.0 ± 157.7 pg/mL, 427.0 ± 152.1 pg/mL and 360 ± 143.1 pg/mL, in the low, medium and high glucocorticoid replacement groups, respectively, P < .0001.GDF15 is increased in states of glucocorticoid deficiency and restored by glucocorticoid replacement. Given the site of action of GDF15 in the hindbrain and its effects on appetite, further study is required to determine the effect of GDF15 in mediating the anorexia and nausea that is a common feature of glucocorticoid deficiency.

    • Changes in climate, in combination with intensive exploitation of marine resources, have caused large-scale reorganizations in many of the world's marine ecosystems during the past decades. The Baltic Sea in Northern Europe is one of the systems most affected. In addition to being exposed to persistent eutrophication, intensive fishing, and one of the world's fastest rates of warming in the last two decades of the 20th century, accelerated climate change including atmospheric warming and changes in precipitation is projected for this region during the 21st century. Here, we used a new multi-model approach to project how the interaction of climate, nutrient loads and cod fishing may affect the future of the open Central Baltic Sea food web. Regionally downscaled global climate scenarios were, in combination with three nutrient load scenarios, used to drive an ensemble of three regional biogeochemical models (BGMs). An Ecopath with Ecosim food web model was then forced with the BGM results from different nutrient-climate scenarios in combination with two different cod fishing scenarios. The results showed that regional management is likely to play a major role in determining the future of the Baltic Sea ecosystem. By the end of the 21st century, for example, the combination of intensive cod fishing and high nutrient loads projected a strongly eutrophicated and sprat-dominated ecosystem, while low cod fishing in combination with low nutrient loads resulted in a cod-dominated ecosystem with eutrophication levels close to present. Also, non-linearities were observed in the sensitivity of different trophic groups to nutrient loads or fishing depending on the combination of the two. Finally, many climate variables and species biomasses were projected to levels unseen in the past. Hence, the risk for ecological surprises needs to be addressed, particularly when the results are discussed in the ecosystem-based management context.

    • The current paper summarizes catch and effort data for Swedish fisheries in the Baltic Sea prior to the standardized recording of fish catches, evaluates spatial and temporal variations and gives interpretations of selected findings. Long-term datasets (1868-1913) of species having different environmental preferences (and of different origin) - marine, migratory and freshwater - are presented for several Swedish counties extending from the Bothnian Bay to the Bornholm Basin. Herring (Clupea harengus membras) was the most important commercial fish-based on landings and also on the amount of historical records available. In addition, herring landings have undoubtedly fluctuated the most. Other commercially important fish were cod (Gadus morhua), salmon and trout (Salmo spp.), flounder (Platichthys flesus), eel (Anguilla anguilla) and whitefish (Coregonus lavaretus). Different species exhibited different patterns of catch dynamics. In general, the 1880s and the 1890s can be characterized as 'good fishing years' for the Swedish Baltic fisheries: catches of herring and cod were high while fishing effort indices were relatively stable. Sprat was not represented in the catch data and may have been recorded as herring. For several species, regions and time periods, catch-effort relationship exhibited clear coupling. However, in several cases, insufficient or lack of effort-related information hampered data interpretations. In summary, the Swedish fisheries during the late 19th century generally exploited the same species as presently (including a dominance by clupeids), but the relative contributions of individual species have changed.
