  • Conceptual uncertainties in modelling the interaction between engineered and natural barriers of nuclear waste repositories in crystalline rocks

    • Nuclear waste disposal in geological formations relies on a multi-barrier concept that includes engineered components – which, in many cases, include a bentonite buffer surrounding waste packages – and the host rock. Contrasts in materials, together with gradients across the interface between the engineered and natural barriers, lead to complex interactions between these two subsystems. Numerical modelling, combined with monitoring and testing data, can be used to improve our overall understanding of rock–bentonite interactions and to predict the performance of this coupled system. Although established methods exist to examine the prediction uncertainties due to uncertainties in the input parameters, the impact of conceptual model decisions on the quantitative and qualitative modelling results is more difficult to assess. A Swedish Nuclear Fuel and Waste Management Company Task Force project facilitated such an assessment. In this project, 11 teams used different conceptualizations and modelling tools to analyse the Bentonite Rock Interaction Experiment (BRIE) conducted at the Äspö Hard Rock Laboratory in Sweden. The exercise showed that prior system understanding along with the features implemented in the available simulators affect the processes included in the conceptual model. For some of these features, sufficient characterization data are available to obtain defensible results and interpretations, whereas others are less supported. The exercise also helped to identify the conceptual uncertainties that led to different assessments of the relative importance of the engineered and natural barrier subsystems. The range of predicted bentonite wetting times encompassed by the ensemble results were considerably larger than the ranges derived from individual models. This is a consequence of conceptual uncertainties, demonstrating the relevance of using a multi-model approach involving alternative conceptualizations.

  • The C-terminus of the transmembrane mucin MUC17 binds to the scaffold protein PDZK1 that stably localizes it to the enterocyte apical membrane in the small intestine

    • The membrane bound mucins have a heavily O-glycosylated extracellular domain, a single pass membrane domain and a short cytoplasmic tail. Three of the membrane bound mucins, MUC3, MUC12 and MUC17, are clustered on chromosome 7 and found in the gastrointestinal tract. These mucins have C-terminal sequences typical for PDZ domain binding proteins. To identify PDZ proteins able to interact with the mucins, we screened PDZ domain arrays using YFP-tagged proteins. MUC17 exhibited a strong binding to PDZK1 whereas the binding to NHERF1 was weak. Furthermore, we showed weak binding of MUC12 to PDZK1, NHERF1 and NHERF2. GST pull-down experiments confirmed that the C-terminal tail of MUC17 co-precipitates with the scaffold protein PDZK1 as identified by mass spectrometry. This was mediated through the C-terminal PDZ-interaction site in MUC17 which was capable of binding to three of the four PDZ domains in PDZK1. Immunostaining of wild-type or Pdzk1-/- mouse jejunum with an antiserum against Muc3(17), the mouse orthologue of human MUC17, revealed strong brush border membrane staining in the wild-type mice compared to an intracellular Muc3(17) staining in the Pdzk1-/- mice. This suggests that Pdzk1 plays a specific role in stabilizing Muc3(17) in the apical membrane of small intestinal enterocytes.

  • Quality and Renewal 2017 (Kvalitet och förnyelse 2017) : Research Environment Evaluation at Uppsala University

    • This is the final report of the enhancement-led research evaluation Quality and Renewal 2017, Q&R17 (in Swedish, Kvalitet och förnyelse, KoF17), carried out at Uppsala University between February 2016 and October 2017. The project has been a major undertaking, aiming to strengthen research at Uppsala University through a broad analysis of the functioning of its various research environments, with particular focus on the preconditions and processes that underpin research quality and renewal. To this end, an internet-based survey was carried out, in which around 3,700 active researchers at Uppsala University shared their perceptions of and opinions on their local research environments at the University. Together with some bibliometric analyses, the survey results served as background material for departmental self-evaluations, which in turn were subjected to external peer review. In this process, almost 130 ‘critical friends’, most of them from outside Sweden, evaluated 54 evaluation units to assess strengths and weaknesses and make recommendations.Q&R17 is the third major research evaluation at Uppsala University. The two previous evaluations, Q&R07 and Q&R11, primarily aimed to identify strong research activities and research initiatives with potential to develop into strong future areas of research, thereby aiding the university management in its continuous strategic decision-making process. In contrast to those two evaluations, Q&R17 has not resulted in any sort of grading of the research carried out at Uppsala University, either in its totality or in its parts. Nevertheless, the panel reports include numerous testimonies of the perceived strength and excellence of research at Uppsala University. More importantly, given the purpose of Q&R17, a number of areas have been identified where action is needed if Uppsala University is to take steps towards reaching its full potential. These relate to: leadership and strategic renewal; talent attraction and retention; quality culture and control; inter-national milieu; external collaboration and outreach; and research-teaching linkages. The conclusions and recommendations coming out of Q&R17 will form the basis for a number of actions throughout the University aiming to further strengthen the international standing of Uppsala University.

  • Intensive care nurses' lived experience of altruism and sacrifices during the Covid-19 pandemic : A phenomenological study

    • AIM: The aim of this study was to understand the lived experience of altruism and sacrifices among Swedish nurses working in intensive care units (ICU) during the COVID-19 pandemic.DESIGN: This was a descriptive phenomenological study.METHODS: The study was conducted between June 2020 and March 2021 and included 20 nurses who were directly involved in the ICU care of COVID-19 patients in Sweden during the pandemic. The text transcripts were analysed using Malterud's Systematic Text Condensation.FINDINGS: The analysis revealed four themes. The work situation changed from 1 day to another-the nurses were brutally confronted with a new and highly demanding situation. Adapting to the chaotic situation-despite fear, anguish and exhaustion, the nurses adapted to the new premises. They shouldered the moral responsibility and responded to the needs of the patients and the health care system since they had the competence. Being confronted with ethical and moral challenges-the nurses were overwhelmed by feelings of helplessness and inadequacy because despite how hard they worked, they were still unable to provide care with dignity and of acceptable quality. The importance of supporting each other-collegiality was fundamental to the nurses' ability to cope with the situation.CONCLUSIONS: Taken together, being exposed to a constantly changing situation, facing the anguish and misery of patients, families, and colleagues, and being confronted with a conflict between the moral obligation to provide care of high quality and the possibility to fulfil this commitment resulted in suffering among the nurses. Collegial back-up and a supportive culture within the caring team were important for the nurses' endurance.IMPACT: The study contributes an understanding of nurses' lived experience of working during the COVID-19 pandemic and highlights the importance of protecting and preparing nurses and nursing organisation for potential future crises.

    • This is the final report of the research project Environmentally differentiated port dues. The purposes of the research are to examine how environmentally differentiated dues and incentives in ports can reduce the environmental impact caused by transport modes that call at the port, and their consequences from legal, political and goods flow perspectives. Both land and sea transport are addressed. The project examines how ports, as important parts of international transport chains, can contribute to the environmental and climate objectives, by introducing environmentally differentiated port dues to promote a shift to more environmentally efficient transport, vehicles, ships, technologies and alternative fuels.
