    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

  • COVID-19 in Patients with Glomerular Disease: Follow-Up Results from the IRoc-GN International Registry

    • Background The acute and long-term effects of severe acute respiratory syndrome coronavirus 2 infection in individuals with GN are still unclear. To address this relevant issue, we created the International Registry of COVID-19 infection in GN.Methods We collected serial information on kidney-related and-unrelated outcomes from 125 GN patients (63 hospitalized and 62 outpatients) and 83 non-GN hospitalized patients with coronavirus disease 2019 (COVID-19) and a median follow-up period of 6.4 (interquartile range 2.3-9.6) months after diagnosis. We used logistic regression for the analyses of clinical outcomes and linear mixed models for the longitudinal analyses of eGFR. All multiple regression models were adjusted for age, sex, ethnicity, and renin-angiotensin-aldosterone system inhibitor use.Results After adjustment for pre-COVID-19 eGFR and other confounders, mortality and AKI did not differ between GN patients and controls (adjusted odds ratio for AKI=1.28; 95% confidence interval [CI], 0.46 to 3.60; P=0.64). The main predictor of AKI was pre-COVID-19 eGFR (adjusted odds ratio per 1 SD unit decrease in eGFR=3.04; 95% CI, 1.76 to 5.28; P,0.001). GN patients developing AKI were less likely to recover pre-COVID19 eGFR compared with controls (adjusted 6-month post-COVID-19 eGFR=0.41; 95% CI, 0.25 to 0.56; times preCOVID-19 eGFR). Shorter duration of GN diagnosis, higher pre-COVID-19 proteinuria, and diagnosis of focal segmental glomerulosclerosis or minimal change disease were associated with a lower post-COVID-19 eGFR.Conclusions Pre-COVID-19 eGFR is the main risk factor for AKI regardless of GN diagnosis. However, GN patients are at higher risk of impaired eGFR recovery after COVID-19-associated AKI. These patients (especially those with high baseline proteinuria or a diagnosis of focal segmental glomerulosclerosis or minimal change disease) should be closely monitored not only during the acute phases of COVID-19 but also after its resolution.

    • Atrial fibrillation (AF) is the most common arrhythmia in clinical practice. At an early stage of the disease, AF may terminate spontaneously and is then referred to as paroxysmal AF. On the other hand, when external intervention is required for the arrhythmia to terminate, it is referred to as persistent AF. In this work, a method to discriminate between paroxysmal and persistent AF in the long-term ECGs is presented. The dominant frequency as well as the organization of the atrial activity are employed to characterize AF. The dominant atrial frequency (DAF) is estimated using hidden Markov model based frequency tracking, and organization is estimated by the sample entropy of the main atrial wave (MAW) and the first two harmonics, respectively. Long-term variations in DAF and organization from 50 ECG recordings were evaluated, showing that episodes of paroxysmal AF were consistently associated with lower DAF and organization of the MAW and the harmonics, than was persistent AF. Discrimination of paroxysmal and persistent AF resulted in classification rates of 84.1±26.1%, thus suggesting that it possible to discriminate between paroxysmal and persistent AF in patients without previously known AF history.

    • The problem of classifying short atrial fibrillatory segments in ambulatory ECG recordings as being either paroxysmal or persistent is addressed by investigating a robust approach to signal characterization. The method comprises preprocessing, estimation of the dominant atrial frequency for the purpose of controlling the subbands of a filter bank, and computation of the relative subband (harmonics) energy and the subband sample entropy. Using minimum-error-rate classification of different feature vectors, a dataset consisting of 24-h ambulatory recordings from 50 subjects with either paroxysmal (26) or persistent (24) atrial fibrillation (AF) was analyzed on a 10-s segment basis; a total of 212196 segments were classified. The best performance in terms of area under the receiver operating characteristic curve was obtained for a feature vector defined by the subband sample entropy of the dominant atrial frequency and the relative harmonics energy, resulting in a value of 0.923, whereas that of the dominant atrial frequency was equal to 0.826. It is concluded that paroxysmal and persistent AF can be discriminated from short segments with good accuracy at any time of an ambulatory recording.

  • Separation of Rare-Earth Elements Using Supported Liquid Membrane Extraction in Pilot Scale

    • The use of supported liquid membrane extraction for recovery and separation of rare-earth elements (REEs) has been investigated. Experiments have been carried out using the different configurations: (1) standard hollow fiber supported liquid membrane operation (HFSLM), (2) renewal liquid membrane operation (HFRLM), and (3) emulsion pertraction technology (EPT). The experiments were performed in pilot scale using a hollow fiber module with a mass transfer surface area of 8 m2. Synthetic feed solution was used with compositions based on a process for recovery of REE from an apatite concentrate. The total concentration of REE in the feed was varied from 1 to 22 mM REE and the pH was varied in the range 1.5–3.2. Di(2-ethylhexyl) phosphoric acid (D2HEPA) diluted in kerosene, 10% (v/v), was used as the organic membrane solution, and 3 M HCl was used as stripping solution. In supported liquid membrane extraction, the extraction performance is governed by both the kinetics of REE transport through the membrane and by thermodynamics. The effect of feed composition on the selectivity and transport of REE through the liquid membrane have been investigated. The results show that the liquid membrane is more selective toward the heavy REE at lower pH values and higher REE concentration. HFRLM shows a higher transport rate than HFSLM, while the HFSLM configuration gives a higher selectivity toward individual REE. The membrane performance in HFSLM configuration rapidly decays with time, while in the HFRLM and EPT configurations, the performance is much more stable. Possible mechanisms for decaying membrane performance are discussed, and gel formation is identified as being of significant importance. Gel formation is observed at an organic loading above ∼46% for Nd, 38% for Y, 46% for Dy, and 65% for Er. The work performed in this study serves as an initial step to demonstrate that HFRLM and EPT can provide stable operation and be feasible options for processing of REE liquors. A process flow diagram for the recovery of the REE, present in the apatite concentrate, in three fractions is proposed based on the results from this study.

    • Colorectal cancer (CRC) is a highly prevalent disease worldwide. Patient survival is hampered by tumor relapse and the appearance of drug-resistant metastases, which are sustained by the presence of cancer stem cells (CSC). Specific delivery of anti-CSC chemotherapeutic drugs to tumors by using targeted drug delivery systems that can also target CSC sub-population might substantially improve current clinical outcomes. CD44v6 is a robust biomarker for advanced CRC and CSC, due to its functional role in tumorigenesis and cancer initiation process. Here, we show that CD44v6-targeted polymeric micelles (PM) loaded with niclosamide (NC S), a drug against CSC, is a good therapeutic strategy against colorectal CSC and circulating tumor cells (CTC) in vivo. HCT116 cells were sorted according to their CD44v6 receptor expression into CD44v6+ (high) and CDv44v6- (low) subpopulations. Accordingly, CD44v6+ cells presented stemness properties, such as overexpression of defined stemness markers (ALDH1A1, CD44v3 and CXCR4) and high capacity to form colonspheres in low attachment conditions. NC S-loaded PM functionalized with an antibody fragment against CD44v6 (Fab-CD44v6) presented adequate size, charge, and encapsulation efficiency. In addition, Fab-CD44v6 significantly increased PM internalization in CD44v6+ cells. Further, encapsulation of NCS improved its effectiveness in vitro, particularly against colonspheres, and allowed to increase its intravenous dosage in vivo by increasing the amount of NCS able to be administered without causing toxicity. Remarkably, functionalized PM accumulate in tumors and significantly reduce CTC in vivo. In conclusion, CD44v6 targeted PM meet the essential conditions to become an efficient anti-CSC therapy.
