  • Comprehensive screening of genomic and metagenomic data reveals a large diversity of tetracycline resistance genes

    • Tetracyclines are broad-spectrum antibiotics used to prevent or treat a variety of bacterial infections. Resistance is often mediated through mobile resistance genes, which encode one of the three main mechanisms: active efflux, ribosomal target protection or enzymatic degradation. In the last few decades, a large number of new tetracycline-resistance genes have been discovered in clinical settings. These genes are hypothesized to originate from environmental and commensal bacteria, but the diversity of tetracycline-resistance determinants that have not yet been mobilized into pathogens is unknown. In this study, we aimed to characterize the potential tetracycline resistome by screening genomic and metagenomic data for novel resistance genes. By using probabilistic models, we predicted 1254 unique putative tetracycline resistance genes, representing 195 gene families (<70% amino acid sequence identity), whereof 164 families had not been described previously. Out of 17 predicted genes selected for experimental verification, 7 induced a resistance phenotype in an Escherichia coli host. Several of the predicted genes were located on mobile genetic elements or in regions that indicated mobility, suggesting that they easily can be shared between bacteria. Furthermore, phylogenetic analysis indicated several events of horizontal gene transfer between bacterial phyla. Our results also suggested that acquired efflux pumps originate from proteobacterial species, while ribosomal protection genes have been mobilized from Firmicutes and Actinobacteria. This study significantly expands the knowledge of known and putatively novel tetracycline resistance genes, their mobility and evolutionary history. The study also provides insights into the unknown resistome and genes that may be encountered in clinical settings in the future.

    • When liquid paperboard based package material ismade aseptic, unsealed edges of the board are exposedto a liquid front which in some circumstances may soakthe material to some extent. This is not desired since itmay lead to aesthetic as well as functional defects. Inorder to make a priori predictions of the edge wickingproperties of a given paper material, something whichis of great interest to paperboard industry as well aspackaging manufacturers, micro simulations are required.To calculate the penetration of fluid in the open edgeof a paper sheet a multi-scale framework is developed.On the fiber micro-scale, virtual paper models are generatedin PaperGeo [6]. In IBOFlow [7] a pore morphologymethod is used to calculate capillary pressurecurves, and the active pores one-phase flow simulationsare performed for relative permeabilities. Theresult is a database of capillary pressure curves andrelative permeabilities as functions of saturation andporosity. The database is used as an input for a twophaseflow simulation on a 2D virtual macro sheet tocalculate the penetration of fluid in the paper. Themulti-scale framework is validated against pressurizededge wick measurements with good agreement.

  • Pregnant under the pressure of a pandemic: a large-scale longitudinal survey before and during the COVID-19 outbreak

    • Background One of the groups that is most vulnerable to the COVID-19 pandemic is pregnant women. They cannot choose to refrain from care; they and their children are at risk of severe complications related to the virus; and they lose comfort and support as clinics prohibit their partners and as societal restrictions demand isolation from friends and relatives. It is urgent to study how this group is faring during the pandemic and we focus here on their health-related worries. Methods A longitudinal survey at a Swedish hospital starting six months before (16 September 2019) and continuing during the COVID-19 outbreak (until 25 August 2020). 6,941 pregnant women and partners of diverse social backgrounds were recruited. 96 percent of birth-giving women in the city take early ultrasounds where recruitment took place. 62 percent of the women with an appointment and 51 percent of their partners gave consent to participate. Results Pregnant women experienced dramatically increased worries for their own health, as well as for their partner’s and their child’s health in the beginning of the pandemic. The worries remained at higher than usual levels throughout the pandemic. Similar, but less dramatic changes, were seen among partners. Conclusions There is a need for heightened awareness of pregnant women’s and partners’ health-related worries as a consequence of the COVID-19 pandemic. Related feelings such as anxiety have been linked to adverse pregnancy outcome and might have long-term effects. The health care system needs to prepare for follow-up visits with these families.

  • Irreversible inhibition of cytosolic thioredoxin reductase 1 as a mechanistic basis for anticancer therapy

    • Cancer cells adapt to their inherently increased oxidative stress through activation of the glutathione (GSH) and thioredoxin (TXN) systems. Inhibition of both of these systems effectively kills cancer cells, but such broad inhibition of antioxidant activity also kills normal cells, which is highly unwanted in a clinical setting. We therefore evaluated targeting of the TXN pathway alone and, more specifically, selective inhibition of the cytosolic selenocysteine-containing enzyme TXN reductase 1 (TXNRD1). TXNRD1 inhibitors were discovered in a large screening effort and displayed increased specificity compared to pan-TXNRD inhibitors, such as auranofin, that also inhibit the mitochondrial enzyme TXNRD2 and additional targets. For our lead compounds, TXNRD1 inhibition correlated with cancer cell cytotoxicity, and inhibitor-triggered conversion of TXNRD1 from an antioxidant to a pro-oxidant enzyme correlated with corresponding increases in cellular production of H2O2. In mice, the most specific TXNRD1 inhibitor, here described as TXNRD1 inhibitor 1 (TRi-1), impaired growth and viability of human tumor xenografts and syngeneic mouse tumors while having little mitochondrial toxicity and being better tolerated than auranofin. These results display the therapeutic anticancer potential of irreversibly targeting cytosolic TXNRD1 using small molecules and present potent and selective TXNRD1 inhibitors. Given the pronounced up-regulation of TXNRD1 in several metastatic malignancies, it seems worthwhile to further explore the potential benefit of specific irreversible TXNRD1 inhibitors for anticancer therapy.

    • Syftet med seminariegrävningen av fornborgen i Botvalde var att kartera och datera fornborgen genom att undersöka stenkonstruktionerna och området inne i borgen som visade höga fosfatvärden. För att kunna utföra dateringar var målet att hitta daterbara artefakter och/eller ekofakter av kol, nötskal och ben i icke omrörda, stratigrafiskt avgränsade kontexter. Dateringen och karteringen av fornborgen går i linje med syftet för forskningsprojektet “I Tjelvars fotspår”, vars mål är att digitalisera och rekonstruera det forntida landskapet kring Lina myr med hjälp av miljöanalyser, genom pollen- och diatomeanalyser, tidigare arkeologiska undersökningar, nya arkeologiska undersökningar samt ett samarbete med lokalbefolkningen och hembygdsföreningar. Målsättningen med projektet är att utarbeta upplevelsebaserade besöksmål i området för att öka kulturturismen att nå ut till den gotländska landsbygden.
