  • Limits on dark matter annihilation signals from the Fermi LAT 4-year measurement of the isotropic gamma-ray background

    • We search for evidence of dark matter (DM) annihilation in the isotropic gamma-ray background (IGRB) measured with 50 months of Fermi Large Area Telescope (LAT) observations. An improved theoretical description of the cosmological DM annihilation signal, based on two complementary techniques and assuming generic weakly interacting massive particle (WIMP) properties, renders more precise predictions compared to previous work. More specifically, we estimate the cosmologically-induced gamma-ray intensity to have an uncertainty of a factor similar to 20 in canonical setups. We consistently include both the Galactic and extragalactic signals under the same theoretical framework, and study the impact of the former on the IGRB spectrum derivation. We find no evidence for a DM signal and we set limits on the DM-induced isotropic gamma-ray signal. Our limits are competitive for DM particle masses up to tens of TeV and, indeed, are the strongest limits derived from Fermi LAT data at TeV energies. This is possible thanks to the new Fermi LAT IGRB measurement, which now extends up to an energy of 820 GeV. We quantify uncertainties in detail and show the potential this type of search offers for testing the WIMP paradigm with a complementary and truly cosmological probe of DM particle signals.

  • Characterization of the Aquaporin-9 Inhibitor RG100204 In Vitro and in db/db Mice

    • Aquaporin-9 (AQP9) is a facilitator of glycerol and other small neutral solute transmembrane diffusion. Identification of specific inhibitors for aquaporin family proteins has been difficult, due to high sequence similarity between the 13 human isoforms, and due to the limited channel surface areas that permit inhibitor binding. The few AQP9 inhibitor molecules described to date were not suitable for in vivo experiments. We now describe the characterization of a new small molecule AQP9 inhibitor, RG100204 in cell-based calcein-quenching assays, and by stopped-flow light-scattering recordings of AQP9 permeability in proteoliposomes. Moreover, we investigated the effects of RG100204 on glycerol metabolism in mice. In cell-based assays, RG100204 blocked AQP9 water permeability and glycerol permeability with similar, high potency (~5 × 10 -8 M). AQP9 channel blocking by RG100204 was confirmed in proteoliposomes. After oral gavage of db/db mice with RG100204, a dose-dependent elevation of plasma glycerol was observed. A blood glucose-lowering effect was not statistically significant. These experiments establish RG100204 as a direct blocker of the AQP9 channel, and suggest its use as an experimental tool for in vivo experiments on AQP9 function.

  • Characterising the Extended Morphologies of BL Lacertae Objects at 144 MHz with LOFAR

    • We present a morphological and spectral study of a sample of 99 BL Lac objects using the LOFAR Two-Metre Sky Survey Second Data Release (LDR2). Extended emission has been identified at gigahertz frequencies around BL Lac objects, but with LDR2 it is now possible to systematically study their morphologies at 144 MHz, where more diffuse emission is expected. LDR2 reveals the presence of extended radio structures around 66/99 of the BL Lac nuclei, with angular extents ranging up to 115 '', corresponding to spatial extents of 410 kpc. The extended emission is likely to be both unbeamed diffuse emission and beamed emission associated with relativistic bulk motion in jets. The spatial extents and luminosities of the extended emission are consistent with the unification scheme for active galactic nuclei, where BL Lac objects correspond to low-excitation radio galaxies with the jet axis aligned along the line of sight. While extended emission is detected around the majority of BL Lac objects, the median 144-1400 MHz spectral index and core dominance at 144 MHz indicate that the core component contributes similar to 42% on average to the total low-frequency flux density. A stronger correlation was found between the 144 MHz core flux density and the gamma-ray photon flux (r = 0.69) than between the 144 MHz extended flux density and the gamma-ray photon flux (r = 0.42). This suggests that the radio-to-gamma-ray connection weakens at low radio frequencies because the population of particles that give rise to the gamma-ray flux are distinct from the electrons producing the diffuse synchrotron emission associated with spatially extended features.

  • Influence of fissure inclination and confining pressure on the local behaviour of natural clays

    • In this experimental study the influence of fissuring orientation and confinement pressure on the mechanical behaviour of natural clays is investigated. The tested material, the scaly clay from Santa Croce di Magliano (south of Italy), is characterised by an intense network of pre-existing fissures of single orientation. Several plane strain compression tests have been conducted, under different confinement pressures (i.e., from 50 to 600 kPa), on specimens having fissures with vertical, medium and horizontal inclination. Digital Image Correlation has been used to follow the deformation processes of the specimens throughout the tests by measuring incremental shear and volumetric strain maps. The results showed a strong coupling between the total confinement and the fissure inclination, that is controlling both the onset and the development of the patterns of the localisation processes. The new results have been compared with previous ones carried out on the same material without confinement. The comparison shed light on the role of total confinement that becomes particularly relevant from certain levels of pressures and fissuring inclination.
