  • Genetic variation in GIPR influences the glucose and insulin responses to an oral glucose challenge

    • Glucose levels 2 h after an oral glucose challenge are a clinical measure of glucose tolerance used in the diagnosis of type 2 diabetes. We report a meta-analysis of nine genome-wide association studies (n = 15,234 nondiabetic individuals) and a follow-up of 29 independent loci (n = 6,958–30,620). We identify variants at the GIPR locus associated with 2-h glucose level (rs10423928, β (s.e.m.) = 0.09 (0.01) mmol/l per A allele, P = 2.0 × 10−15). The GIPR A-allele carriers also showed decreased insulin secretion (n = 22,492; insulinogenic index, P = 1.0 × 10−17; ratio of insulin to glucose area under the curve, P = 1.3 × 10−16) and diminished incretin effect (n = 804; P = 4.3 × 10−4). We also identified variants at ADCY5 (rs2877716, P = 4.2 × 10−16), VPS13C (rs17271305, P = 4.1 × 10−8), GCKR (rs1260326, P = 7.1 × 10−11) and TCF7L2 (rs7903146, P = 4.2 × 10−10) associated with 2-h glucose. Of the three newly implicated loci (GIPR, ADCY5 and VPS13C), only ADCY5 was found to be associated with type 2 diabetes in collaborating studies (n = 35,869 cases, 89,798 controls, OR = 1.12, 95% CI 1.09–1.15, P = 4.8 × 10−18).

  • Residence-based priorities and school choice

    • Many countries use centralized school choice procedures to assign pupils to schools. To address excess demand for a particular school, ties are broken according to priority points granted based on various criteria, such as proximity to the school. Using a unique reform undertaken in Madrid (Spain), we estimate the impact of abolishing residence-based priorities on families’ school choices, the stated motivation for choosing a school, and the final school allocation. Utilizing several administrative datasets on school applications, we find that the reform increases families’ out-of-district school assignments and assignments to schools further away from their home address. Parents of immigrant children did not change their application behavior in the first years of the reform but caught up with natives three years after its implementation. Children generally accessed slightly better-performing schools, particularly those from lower-educated backgrounds.

  • School choice priorities and school segregation : Evidence from Madrid

    • Many countries use centralized school choice procedures to assign pupils to schools. To address excess demand for a particular school, ties are broken according to priority points that are granted based on various criteria, such as proximity to the school. We use two unique reforms undertaken in a large-scale educational market, one abolishing resident-based priorities and the other reducing low-income priority points, to estimate whether these changes impact families’ school choices and school segregation. We exploit comprehensive administrative data on parents’ applications to schools that allow us to control for residential segregation confounders. We find that abolishing school district boundaries increases families’ out-of-district school assignment and raises assignment to schools located farther from their home address but has no detrimental effect on segregation. In contrast, we find that sharply reducing low-income priority points substantially increases school segregation. This shows that priority points are an important policy tool in determining both families’ school choices and segregation.

  • Elevation alters ecosystem properties across temperate treelines globally

    • Temperature is a primary driver of the distribution of biodiversity as well as of ecosystem boundaries(1,2). Declining temperature with increasing elevation in montane systems has long been recognized as a major factor shaping plant community biodiversity, metabolic processes, and ecosystem dynamics(3,4). Elevational gradients, as thermoclines, also enable prediction of long-term ecological responses to climate warming(5-7). One of the most striking manifestations of increasing elevation is the abrupt transitions from forest to treeless alpine tundra(8). However, whether there are globally consistent above-and belowground responses to these transitions remains an open question(4). To disentangle the direct and indirect effects of temperature on ecosystem properties, here we evaluate replicate treeline ecotones in seven temperate regions of the world. We find that declining temperatures with increasing elevation did not affect tree leaf nutrient concentrations, but did reduce ground-layer community-weighted plant nitrogen, leading to the strong stoichiometric convergence of ground-layer plant community nitrogen to phosphorus ratios across all regions. Further, elevation-driven changes in plant nutrients were associated with changes in soil organic matter content and quality (carbon to nitrogen ratios) and microbial properties. Combined, our identification of direct and indirect temperature controls over plant communities and soil properties in seven contrasting regions suggests that future warming may disrupt the functional properties of montane ecosystems, particularly where plant community reorganization outpaces treeline advance.

  • Patchy field sampling biases understanding of climate change impacts across the Arctic

    • Effective societal responses to rapid climate change in the Arctic rely on an accurate representation of region-specific ecosystem properties and processes. However, this is limited by the scarcity and patchy distribution of field measurements. Here, we use a comprehensive, geo-referenced database of primary field measurements in 1,840 published studies across the Arctic to identify statistically significant spatial biases in field sampling and study citation across this globally important region. We find that 31% of all study citations are derived from sites located within 50 km of just two research sites: Toolik Lake in the USA and Abisko in Sweden. Furthermore, relatively colder, more rapidly warming and sparsely vegetated sites are under-sampled and under-recognized in terms of citations, particularly among microbiology-related studies. The poorly sampled and cited areas, mainly in the Canadian high-Arctic archipelago and the Arctic coastline of Russia, constitute a large fraction of the Arctic ice-free land area. Our results suggest that the current pattern of sampling and citation may bias the scientific consensuses that underpin attempts to accurately predict and effectively mitigate climate change in the region. Further work is required to increase both the quality and quantity of sampling, and incorporate existing literature from poorly cited areas to generate a more representative picture of Arctic climate change and its environmental impacts.

  • Direction of actin flow dictates integrin LFA-1 orientation during leukocyte migration

    • Integrin αβ heterodimer cell surface receptors mediate adhesive interactions that provide traction for cell migration. Here, we test whether the integrin, when engaged to an extracellular ligand and the cytoskeleton, adopts a specific orientation dictated by the direction of actin flow on the surface of migrating cells. We insert GFP into the rigid, ligand-binding head of the integrin, model with Rosetta the orientation of GFP and its transition dipole relative to the integrin head, and measure orientation with fluorescence polarization microscopy. Cytoskeleton and ligand-bound integrins orient in the same direction as retrograde actin flow with their cytoskeleton-binding β-subunits tilted by applied force. The measurements demonstrate that intracellular forces can orient cell surface integrins and support a molecular model of integrin activation by cytoskeletal force. Our results place atomic, Å-scale structures of cell surface receptors in the context of functional and cellular, μm-scale measurements.

  • Validation of an open source, remote web-based eye-tracking method (WebGazer) for research in early childhood

    • Measuring eye movements remotely via the participant's webcam promises to be an attractive methodological addition to in-person eye-tracking in the lab. However, there is a lack of systematic research comparing remote web-based eye-tracking with in-lab eye-tracking in young children. We report a multi-lab study that compared these two measures in an anticipatory looking task with toddlers using WebGazer.js and jsPsych. Results of our remotely tested sample of 18-27-month-old toddlers (N=125) revealed that web-based eye-tracking successfully captured goal-based action predictions, although the proportion of the goal-directed anticipatory looking was lower compared to the in-lab sample (N=70). As expected, attrition rate was substantially higher in the web-based (42%) than the in-lab sample (10%). Excluding trials based on visual inspection of the match of time-locked gaze coordinates and the participant's webcam video overlayed on the stimuli was an important preprocessing step to reduce noise in the data. We discuss the use of this remote web-based method in comparison with other current methodological innovations. Our study demonstrates that remote web-based eye-tracking can be a useful tool for testing toddlers, facilitating recruitment of larger and more diverse samples; a caveat to consider is the larger drop-out rate.

    • Flow in a stably stratified environment is characterized by anisotropic and intermittent turbulence and wavelike motions of varying amplitudes and periods. Understanding turbulence intermittency and wave-turbulence interactions in a stably stratified flow remains a challenging issue in geosciences including planetary atmospheres and oceans. The stable atmospheric boundary layer (SABL) commonly occurs when the ground surface is cooled by longwave radiation emission such as at night over land surfaces, or even daytime over snow and ice surfaces, and when warm air is advected over cold surfaces. Intermittent turbulence intensification in the SABL impacts human activities and weather variability, yet it cannot be generated in state-of-the-art numerical forecast models. This failure is mainly due to a lack of understanding of the physical mechanisms for seemingly random turbulence generation in a stably stratified flow, in which wave-turbulence interaction is a potential mechanism for turbulence intermittency. A workshop on wave-turbulence interactions in the SABL addressed the current understanding and challenges of wave-turbulence interactions and the role of wavelike motions in contributing to anisotropic and intermittent turbulence from the perspectives of theory, observations, and numerical parameterization. There have been a number of reviews on waves, and a few on turbulence in stably stratified flows, but not much on wave-turbulence interactions. This review focuses on the nocturnal SABL; however, the discussions here on intermittent turbulence and wave-turbulence interactions in stably stratified flows underscore important issues in stably stratified geophysical dynamics in general.
