  • An MPER antibody neutralizes HIV-1 using germline features shared among donors

    • The membrane-proximal external region (MPER) of HIV-1 envelope glycoprotein (Env) can be targeted by neutralizing antibodies of exceptional breadth. MPER antibodies usually have long, hydrophobic CDRH3s, lack activity as inferred germline precursors, are often from the minor IgG3 subclass, and some are polyreactive, such as 4E10. Here we describe an MPER broadly neutralizing antibody from the major IgG1 subclass, PGZL1, which shares germline V/D-region genes with 4E10, has a shorter CDRH3, and is less polyreactive. A recombinant sublineage variant pan-neutralizes a 130-isolate panel at 1.4 μg/ml (IC50). Notably, a germline revertant with mature CDR3s neutralizes 12% of viruses and still binds MPER after DJ reversion. Crystal structures of lipid-bound PGZL1 variants and cryo-EM reconstruction of an Env-PGZL1 complex reveal how these antibodies recognize MPER and viral membrane. Discovery of common genetic and structural elements among MPER antibodies from different patients suggests that such antibodies could be elicited using carefully designed immunogens.

    • The effects of estrogens are pleiotropic, affecting multiple bodily systems. Changes from the body’s natural fluctuating levels of estrogens, through surgical removal of the ovaries, natural menopause, or the administration of exogenous estrogens to menopausal women have been independently linked to an altered immune profile, and changes to cognitive processes. Here, we propose that inflammation may mediate the relationship between low levels of estrogens and cognitive decline. In order to determine what is known about this connection, we review the literature on the cognitive effects of decreased estrogens due to oophorectomy or natural menopause, decreased estrogens’ role on inflammation – both peripherally and in the brain – and the relationship between inflammation and cognition. While this review demonstrates that much is unknown about the intersection between estrogens, cognition, inflammation, we propose that there is an important interaction between these literatures.

    • The bright fluorescent cytosine analogue tCO stands out among fluorescent bases due to its virtually unquenched fluorescence emission in duplex DNA. However, like most reported base analogues, it has not been thoroughly characterized in RNA. We here report on the first synthesis and RNA-incorporation of tCO, and characterize its base-mimicking and fluorescence properties in RNA. As in DNA, we find a high quantum yield inside RNA duplexes (< Phi(F)> = 0.22) that is virtually unaffected by the neighbouring bases (Phi(F) = 0.20-=0.25), resulting in an average brightness of 1900 M-1 cm(-1). The average fluorescence lifetime in RNA duplexes is 4.3 ns and generally two lifetimes are required to fit the exponential decays. Fluorescence properties in ssRNA are defined by a small increase in average quantum yield (< Phi(F) > = 0.24) compared to dsRNA, with a broader distribution (Phi(F) = 0.17-=0.34) and slightly shorter average lifetimes. Using circular dichroism, we find that the tCO-modified RNA duplexes form regular A-form helices and in UV-melting experiments the stability of the duplexes is only slightly higher than that of the corresponding natural RNA ( = + 2.3 degrees C). These properties make tC(0) a highly interesting fluorescent RNA base analogue for detailed FRET-based structural measurements, as a bright internal label in microscopy, and for fluorescence anisotropy measurements of RNA dynamics.

  • Mixing, heat fluxes and heat content evolution of the Arctic Ocean mixed layer

    • A comprehensive measurement program was conducted during 16 days of a 3 week long ice pack drift, from 15 August to 1 September 2008 in the central Amundsen Basin, Arctic Ocean. The data, sampled as part of the Arctic Summer Cloud Ocean Study (ASCOS), included upper ocean stratification, mixing and heat transfer as well as transmittance solar radiation through the ice. The observations give insight into the evolution of the upper layers of the Arctic Ocean in the transition period from melting to freezing. The ocean mixed layer was found to be heated from above and, for summer conditions, the net heat flux through the ice accounted for 22% of the observed change in mixed layer heat content. Heat was mixed downward within the mixed layer and a small, downward heat flux across the base of the mixed layer accounted for the accumulated heat in the upper cold halocline during the melting season. On average, the ocean mixed layer was cooled by an ocean heat flux at the ice/ocean interface (1.2 W m(-2)) and heated by solar radiation through the ice (-2.6 W m(-2)). An abrupt change in surface conditions halfway into the drift due to freezing and snowfall showed distinct signatures in the data set and allowed for inferences and comparisons to be made for cases of contrasting forcing conditions. Transmittance of solar radiation was reduced by 59% in the latter period. From hydro-graphic observations obtained earlier in the melting season, in the same region, we infer a total fresh water equivalent of 3.3 m accumulated in the upper ocean, which together with the observed saltier winter mixed layer indicates a transition towards a more seasonal ice cover in the Arctic.
