  • The Beaker phenomenon and the genomic transformation of northwest Europe

    • From around 2750 to 2500 bc, Bell Beaker pottery became widespread across western and central Europe, before it disappeared between 2200 and 1800 bc. The forces that propelled its expansion are a matter of long-standing debate, and there is support for both cultural diffusion and migration having a role in this process. Here we present genome-wide data from 400 Neolithic, Copper Age and Bronze Age Europeans, including 226 individuals associated with Beaker-complex artefacts. We detected limited genetic affinity between Beaker-complex-associated individuals from Iberia and central Europe, and thus exclude migration as an important mechanism of spread between these two regions. However, migration had a key role in the further dissemination of the Beaker complex. We document this phenomenon most clearly in Britain, where the spread of the Beaker complex introduced high levels of steppe-related ancestry and was associated with the replacement of approximately 90% of Britain's gene pool within a few hundred years, continuing the east-to-west expansion that had brought steppe-related ancestry into central and northern Europe over the previous centuries.

  • Observations of Low-frequency Radio Emission from Millisecond Pulsars and Multipath Propagation in the Interstellar Medium

    • Studying the gravitational-wave sky with pulsar timing arrays (PTAs) is a key science goal for the Square Kilometre Array (SKA) and its pathfinder telescopes. With current PTAs reaching sub-microsecond timing precision, making accurate measurements of interstellar propagation effects and mitigating them effectively has become increasingly important to realize PTA goals. As these effects are much stronger at longer wavelengths, low-frequency observations are most appealing for characterizing the interstellar medium (ISM) along the sight lines toward PTA pulsars. The Murchison Widefield Array (MWA) and the Engineering Development Array (EDA), which utilizes MWA technologies, present promising opportunities for undertaking such studies, particularly for PTA pulsars located in the southern sky. Such pulsars are also the prime targets for PTA efforts planned with the South African MeerKAT, and eventually with the SKA. In this paper we report on observations of two bright southern millisecond pulsars, PSR J0437-4715 and PSR J2145-0750, made with these facilities; MWA observations sampled multiple frequencies across the 80-250 MHz frequency range, while the EDA provided direct-sampled baseband data to yield a large instantaneous usable bandwidth of similar to 200 MHz. Using these exploratory observations, we investigate various aspects relating to pulsar emission and ISM properties, such as spectral evolution of the mean pulse shape, scintillation as a function of frequency, chromaticity in interstellar dispersion, and flux density spectra at low frequencies. Systematic and regular monitoring observations will help ascertain the role of low-frequency measurements in PTA experiments, while simultaneously providing a detailed characterization of the ISM toward the pulsars, which will be useful in devising optimal observing strategies for future PTA experiments.

  • Blood-brain barrier opening of the default mode network in Alzheimer's disease with magnetic resonance-guided focused ultrasound

    • In an open-label single-arm trial, Meng et al. investigate the feasibility and tolerability of opening the blood-brain barrier in the default mode network with MR-guided focused ultrasound in patients with Alzheimer's disease. The study intervention led to local amyloid reduction on PET imaging, but no change in disease-specific markers in plasma or CSF. The blood-brain barrier (BBB) protects the brain but is also an important obstacle for the effective delivery of therapeutics in Alzheimer's disease and other neurodegenerative disorders. Transcranial magnetic resonance-guided focused ultrasound (MRgFUS) has been shown to reversibly disrupt the BBB. However, treatment of diffuse regions across the brain along with the effect on Alzheimer's disease relevant pathology need to be better characterized. This study is an open-labelled single-arm trial (NCT04118764) to investigate the feasibility of modulating BBB permeability in the default mode network and the impact on cognition, amyloid and tau pathology as well as BBB integrity. Nine participants [mean age 70.2 +/- 7.2 years, mean Mini-Mental State Examination (MMSE) 21.9] underwent three biweekly procedures with follow-up visits up to 6 months. The BBB permeability of the bilateral hippocampi, anterior cingulate cortex and precuneus was transiently increased without grade 3 or higher adverse events. Participants did not experience worsening trajectory of cognitive decline (ADAS-cog11, MMSE). Whole brain vertex-based analysis of the F-18-florbetaben PET imaging demonstrated clusters of modest SUVR reduction in the right parahippocampal and inferior temporal lobe. However, CSF and blood biomarkers did not demonstrate any amelioration of Alzheimer's disease pathology (P-tau181, amyloid-beta 42/40 ratio), nor did it show persistent BBB dysfunction (plasma PDGFRbeta and CSF-to-plasma albumin ratio). This study provides neuroimaging and fluid biomarker data to characterize the safety profile of MRgFUS BBB modulation in neurodegeneration as a potential strategy for enhanced therapeutic delivery.

  • MWA tied-array processing I: Calibration and beamformation

    • The Murchison Widefield Array is a low-frequency Square Kilometre Array precursor located at the Murchison Radio-astronomy Observatory in Western Australia. Primarily designed as an imaging telescope, but with a flexible signal path, the capabilities of this telescope have recently been extended to include off-line incoherent and tied-array beam formation using recorded antenna voltages. This has provided the capability for high-time and frequency resolution observations, including a pulsar science program. This paper describes the algorithms and pipeline that we have developed to form the tied-array beam products from the summation of calibrated signals of the antenna elements, and presents example polarimetric profiles for PSRs J0437-4715 and J1900-2600 at 185 MHz.

    • Locomotive design is a highly complex task that requires the use of systems engineering that depends upon knowledge from a range of disciplines and is strongly oriented on how to design and manage complex systems that operate under a wide range of different train operational conditions on various types of tracks. Considering that field investigation programs for locomotive operational scenarios involve high costs and cause disruption of train operations on real railway networks and given recent developments in the rollingstock compliance standards in Australia and overseas that allow the assessment of some aspects of rail vehicle behaviour through computer simulations, a great number of multidisciplinary research studies have been performed and these can contribute to further improvement of a locomotive design technique by increasing the amount of computer-based studies. This paper was focused on the presentation of the all-important key components required for locomotive studies, starting from developing a realistic locomotive design model, its validation and further applications for train studies. The integration of all engineering disciplines is achieved by means of advanced simulation approaches that can incorporate existing AC and DC locomotive designs, hybrid locomotive designs, full locomotive traction system models, rail friction processes, the application of simplified and exact wheel-rail contact theories, wheel-rail wear and rolling contact fatigue, train dynamic behaviour and in-train forces, comprehensive track infrastructure details, and the use of co-simulation and parallel computing. The co-simulation and parallel computing approaches that have been implemented on Central Queensland University’s High-Performance Computing cluster for locomotive studies will be presented. The confidence in these approaches is based on specific validation procedures that include a locomotive model acceptance procedure and field test data. The problems and limitations presented in locomotive traction studies in the way they are conducted at the present time are summarised and discussed.
