  • Practical Guide to Measuring Wetland Carbon Pools and Fluxes

    • Wetlands cover a small portion of the world, but have disproportionate influence on global carbon (C) sequestration, carbon dioxide and methane emissions, and aquatic C fluxes. However, the underlying biogeochemical processes that affect wetland C pools and fluxes are complex and dynamic, making measurements of wetland C challenging. Over decades of research, many observational, experimental, and analytical approaches have been developed to understand and quantify pools and fluxes of wetland C. Sampling approaches range in their representation of wetland C from short to long timeframes and local to landscape spatial scales. This review summarizes common and cutting-edge methodological approaches for quantifying wetland C pools and fluxes. We first define each of the major C pools and fluxes and provide rationale for their importance to wetland C dynamics. For each approach, we clarify what component of wetland C is measured and its spatial and temporal representativeness and constraints. We describe practical considerations for each approach, such as where and when an approach is typically used, who can conduct the measurements (expertise, training requirements), and how approaches are conducted, including considerations on equipment complexity and costs. Finally, we review key covariates and ancillary measurements that enhance the interpretation of findings and facilitate model development. The protocols that we describe to measure soil, water, vegetation, and gases are also relevant for related disciplines such as ecology. Improved quality and consistency of data collection and reporting across studies will help reduce global uncertainties and develop management strategies to use wetlands as nature-based climate solutions.

    • Genetic editing technologies have emerged as a potential therapeutic tool in various biomedical fields owing to their applications against cancer, neurological diseases, diabetes, autoimmune disorder, muscu-lar dystrophy, bacterial infections (AMR), and cardiovascular diseases. CRISPR is one such valuable genetic editing tool with extensive therapeutic appliances but with a major challenge in terms of deliv-ery. Herein, we have strived to exploit a synergy of nanocarriers and CRISPR against the aforementioned diseases for their medical applications and explicated their clinical significance including the enhanced delivery via endosomal escape and environmental factors such as light, pH, and stimuli. In addition to highlighting the delivery strategies of nano-carriers for CRISPR and their characterization, we have expounded on the reliant factor of the CRISPR-Cas Complex.

  • Recent Advancements, Limitations, and Future Perspectives of the use of Personalized Medicine in Treatment of Colon Cancer

    • Due to the heterogeneity of colon cancer, surgery, chemotherapy, and radiation are ineffective in all cases. The genomic profile and biomarkers associated with the process are considered in personalized medicine, along with the patient's personal history. It is based on the response of the targeted therapies to specific genetic variations. The patient's genetic transcriptomic and epigenetic features are evaluated, and the best therapeutic approach and diagnostic testing are identified through personalized medicine. This review aims to summarize all the necessary, updated information on colon cancer related to personalized medicine. Personalized medicine is gaining prominence as generalized treatments are finding it challenging to contain colon cancer cases which currently rank fourth among global cancer incidence while being the fifth largest in total death cases worldwide. In personalized therapy, patients are grouped into specific categories, and the best therapeutic approach is chosen based on evaluating their molecular features. Various personalized strategies are currently being explored in the treatment of colon cancer involving immunotherapy, phytochemicals, and other biomarker-specific targeted therapies. However, significant challenges must be overcome to integrate personalized medicine into healthcare systems completely. We look at the various signaling pathways and genetic and epigenetic alterations associated with colon cancer to understand and identify biomarkers useful in targeted therapy. The current personalized therapies available in colon cancer treatment and the strategies being explored to improve the existing methods are discussed. This review highlights the advantages and limitations of personalized medicine in colon cancer therapy. The current scenario of personalized medicine in developed countries and the challenges faced in middle- and low-income countries are also summarized. Finally, we discuss the future perspectives of personalized medicine in colon cancer and how it could be integrated into the healthcare systems.

  • Occurrence, predictors and hazards of elevated groundwater arsenic across India through field observations and regional-scale AI-based modeling

    • Existence of wide spread elevated concentrations of groundwater arsenic (As) across South Asia, including India, has endangered a huge groundwater-based drinking water dependent population. Here, using high-spatial resolution As field-observations (similar to 3 million groundwater sources) across India, we have delineated the regional-scale occurrence of elevated groundwater As (>= 10 mu g/L), along with the possible geologic-geomorphologic-hydrologic and human-sourced predictors that influence the spatial distribution of the contaminant. Using statistical and machine learning method, we also modeled the groundwater As concentrations probability at 1 Km resolution, along with probabilistic delineation of high As-hazard zones across India. The observed occurrence of groundwater As was found to be most strongly influenced by geology-tectonics, groundwater-fed irrigated area (%) and elevation. Pervasive As contamination is observed in major parts of the Himalayan mega-river Indus-Ganges-Brahmaputra basins, however it also occurs in several more-localized pockets, mostly related to ancient tectonic zones, igneous provinces, aquifers in modern delta and chalcophile mineralized regions. The model results suggest As-hazard potential in yet-undetected areas. Our model performed well in predicting groundwater arsenic, with accuracy: 82% and 84%; area under the curve (AUC): 0.89 and 0.88 for test data and validation datasets. An estimated similar to 90 million people across India are found to be exposed to high groundwater As from field-observed data, with the five states with highest hazard are West Bengal (28 million), Bihar (21 million), Uttar Pradesh (15 million), Assam(8.6 million) and Punjab (6 million). However it can be much more if the modeled hazard is considered (>250 million). Thus, our study provides a detailed, quantitative assessment of high groundwater As across India, with delineation of possible intrinsic influences and exogenous forcings. The predictive model is helpful in predicting As-hazard zones in the areas with limited measurements.

    • We have carried out an experimental study of Ni-rich NiMn alloys in a series of compositions across the multicritical point and determined the phase diagram within that range. We have observed ferromagnetic long-range order with reentrant spin-glass/ferro-spin-glass phase for x <= 25, an antiferromagnetic long-range order around x similar to 37, and a gradual change from a canonical spin-glass state to a long-range antiferromagnetic phase in the intermediate composition region. In order to explain the experimental observations, we have examined the physical properties from a density functional based first-principals theoretical analysis and used it to understand the experimental results. Using atomic spin dynamics simulations based on the Landau-Lifshitz-Gilbert equation, we have found the aging behavior and anomalously slow relaxation of magnetization in the composition range where experiment observes spin-glassy behavior.
