    • Using population pharmacokinetic analysis (PPK), we attempted to identify predictors of S-warfarin clearance (CL(S)) and to clarify population differences in S-warfarin pharmacokinetics among a cohort of 378 African American, Asian and white patients. Significant predictors of CL(S) included clinical (age, body weight and sex) and genotypic (CYP2C9*2,*3 and *8) factors, as well as African American ethnicity, the median CL(S) being 30% lower in the latter than in Asians and whites (170 versus 243 and 250 ml h(-1), P<0.01). The plasma S-warfarin (Cp(S)) time courses following the genotype-based dosing algorithms simulated using the PPK estimates showed African Americans with CYP2C9*1/*1 and any of the VKORC1 genotypes would have an average Cp(S) at steady state 1.5-1.8 times higher than in Asians and whites. These results indicate warfarin dosing algorithms should be evaluated in each respective ethnic population. Further study of a large African American cohort will be necessary to confirm the present findings.

  • Superhard Semiconducting Optically Transparent High Pressure Phase of Boron

    • An orthorhombic (space group Pnnm) boron phase was synthesized at pressures above 9 GPa and high temperature, and it was demonstrated to be stable at least up to 30 GPa. The structure, determined by single-crystal x-ray diffraction, consists of B-12 icosahedra and B-2 dumbbells. The charge density distribution obtained from experimental data and ab initio calculations suggests covalent chemical bonding in this phase. Strong covalent interatomic interactions explain the low compressibility value (bulk modulus is K-300=227 GPa) and high hardness of high-pressure boron (Vickers hardness H-V=58 GPa), after diamond the second hardest elemental material.

    • Several different experimental techniques are normally used to evaluate tribological properties of thin hard coatings. However, most of the tests cannot assess properties of coating, substrate and interface independently. In this study, the potential of a Micro-Slurry-jet Erosion (MSE) test to swiftly evaluate the intrinsic wear properties of thin single layered and multilayered coatings is demonstrated. A slurry containing 1.2 mu m alumina particles was impacted at high velocity perpendicular to CVD TiC/TiN (TiN on top of TiC) and Tic coatings deposited on cemented carbide. In addition, nano-indentation and XRD, GDOES, AFM analyses were done for the original and worn surfaces. For the TiC/TiN coating, the wear depth of the TiN layer increased linearly until the Tic layer was reached. There after the wear occurred at a lower rate but tended to increase towards the TiC/substrate interlayer. The single layered Tic coating showed the same wear behaviour as the TiC layer of the TiC/TiN coating. The wear rate was constant within the individual TiN and Tic layers, but changed near the interfaces. The micro-topographies of the worn surfaces observed by AFM correlated to the difference in the wear rate of the individual layers and the change in wear rate at the interface to the substrate. The Tic layer proved to have about two times higher wear resistance than the TiN layer. Consequently, the MSE test can distinguish between small differences in the properties of the coatings and of the interlayer zones, and can be used as a screening test when evaluating multilayered as well as single layered coatings.

  • Synthesis of Ultra‐Incompressible and Recoverable Carbon Nitrides Featuring CN4 Tetrahedra

    • Carbon nitrides featuring three-dimensional frameworks of CN4 tetrahedra are one of the great aspirations of materials science, expected to have a hardness greater than or comparable to diamond. After more than three decades of efforts to synthesize them, no unambiguous evidence of their existence has been delivered. Here, the high-pressure high-temperature synthesis of three carbon-nitrogen compounds, tI14-C3N4, hP126-C3N4, and tI24-CN2, in laser-heated diamond anvil cells, is reported. Their structures are solved and refined using synchrotron single-crystal X-ray diffraction. Physical properties investigations show that these strongly covalently bonded materials, ultra-incompressible and superhard, also possess high energy density, piezoelectric, and photoluminescence properties. The novel carbon nitrides are unique among high-pressure materials, as being produced above 100 GPa they are recoverable in air at ambient conditions.
