    • Surface texture parameters can provide a link between texture, its processing and function. Recent surveys and industrial experience have shown that the ISO 25178 areal surface texture parameters have not received the level of traction in industry that was predicted when introduced despite the fact that the areal parameters were predicted to have more functionally relevant characterisations than the ISO 4287 profile parameters. The objective of the paper is to enable more functionally relevant specifications of surface texture to be taken up by industry and the scientific community by increasing the knowledge of the ISO 25178 texture and novel feature parameters, and their potential use, as well as knowledge about methods for establishing functionally relevant surface texture specifications. In the paper, existing methods for functional tolerancing of surface texture are reviewed and discussed, examples of applications are given and a direction for continued research is presented. © 2020 The Authors

  • Cloning and sequencing of human gp330, a Ca2+ -binding receptor with potential intracellular signaling properties

    • We present here the complete primary structure of human gp330, the human variant of the principal kidney autoantigen causing Heymann membranous glomerulonephritis in rats. The deduced 4655 amino acid residues give a calculated molecular mass of 519636 Da for the mature protein and consists of a probable 25-amino-acid N-terminal signal peptide sequence, an extracellular region of 4398 amino acids, a single transmembrane-spanning domain of 23 amino acids, and an intracellular C-terminal region of 209 amino acid residues. Three types of cysteine-rich repeats characteristic of the low density lipoprotein receptor (LDLR) superfamily are present in human gp330. In the extracellular region, there are a total of 36 LDLR ligand-binding repeats, comprising four distinct domains, 16 growth factor repeats separated by eight YWTD spacer regions, and one epidermal growth factor-like repeat. No consensus cleavage sequence for the processing endoprotease furin is detected in human gp330. The intracellular tail contains not only two copies of the F(X)NPXY coated-pit mediated internalization signal characteristic of LDLR superfamily members, but also intriguing and potentially functional motifs including several Src-homology 3 recognition motifs, one Src-homology 2 recognition motif for the p85 regulatory subunit of phosphatidylinositol 3-kinase, and additional sites for protein kinase C, casein kinase II and cAMP-/cGMP-dependent protein kinase. There is approximately 77% amino acid identity between human and rat gp330 with minor differences between the extracellular and intracellular regions. Recently gp330 has been implicated in Ca2+ regulation in the parathyroid, the placenta, and the renal tubule, but its overall physiological and pathological role still remains uncertain.

    • Variability is unavoidable in the realization of products. While design must specify ideal geometry, it shall also describe limits of variability (tolerances) that must be met in order to maintain proper product function. Although tolerancing is a mature field, new manufacturing processes and design methodologies are creating new avenues of research, and modelling standards must also evolve to support these processes. In addition, the study of uncertainty has produced widely-accepted methods of quantifying variability, and modern tolerancing tools should support these methods. The challenges introduced by new processes and design methodologies continue to make tolerancing research a fertile and productive area.

  • Impact of liver tumour burden, alkaline phosphatase elevation, and target lesion size on treatment outcomes with Lu-177-Dotatate : an analysis of the NETTER-1 study

    • Purpose To assess the impact of baseline liver tumour burden, alkaline phosphatase (ALP) elevation, and target lesion size on treatment outcomes with Lu-177-Dotatate. Methods In the phase 3 NETTER-1 trial, patients with advanced, progressive midgut neuroendocrine tumours (NET) were randomised to 177Lu-Dotatate (every 8 weeks, four cycles) plus octreotide long-acting release (LAR) or to octreotide LAR 60 mg. Primary endpoint was progression-free survival (PFS). Analyses of PFS by baseline factors, including liver tumour burden, ALP elevation, and target lesion size, were performed using Kaplan-Meier estimates; hazard ratios (HRs) with corresponding 95% CIs were estimated using Cox regression. Results Significantly prolonged median PFS occurred with Lu-177-Dotatate versus octreotide LAR 60 mg in patients with low (< 25%), moderate (25-50%), and high (> 50%) liver tumour burden (HR 0.187, 0.216, 0.145), and normal or elevated ALP (HR 0.153, 0.177), and in the presence or absence of a large target lesion (diameter > 30 mm; HR, 0.213, 0.063). Within the Lu-177-Dotatate arm, no significant difference in PFS was observed amongst patients with low/moderate/high liver tumour burden (P = 0.7225) or with normal/elevated baseline ALP (P = 0.3532), but absence of a large target lesion was associated with improved PFS (P = 0.0222). Grade 3 and 4 liver function abnormalities were rare and did not appear to be associated with high baseline liver tumour burden. Conclusions Lu-177-Dotatate demonstrated significant prolongation in PFS versus high-dose octreotide LAR in patients with advanced, progressive midgut NET, regardless of baseline liver tumour burden, elevated ALP, or the presence of a large target lesion. : NCT01578239, EudraCT: 2011-005049-11

  • Physics-based modelling and measurement of advanced manufacturing machinery’s positioning accuracy : Machine tools, industrial manipulators and their positioning accuracy

    • Advanced manufacturing machinery is a corner stone of essential industries of technologicallydeveloped societies. Their accuracy permits the production of complexproducts according to tight geometric dimensions and tolerances for high efficiency,interchangeability and sustainability. The accuracy of advanced manufacturingmachinery can be quantified by the performance measure of positioning accuracy.Positioning accuracy measures the closeness between a commanded and an attainedposition on a machine tool or industrial manipulator, and it is ruled by lawsof physics in classical mechanics and thermodynamics. These laws can be applied tomodel how much the machinery deflects due to gravity, expands due to a change intemperature and how much and how long it vibrates due to process forces; hence,one can quantify how much the accuracy decreases. Thus, to produce machinerywith ever higher accuracy and precision one can design machines which deflect,expand and vibrate less or one can understand and model the actual behaviour ofthe machinery to compensate for it.This licentiate thesis uses physics-based modelling to quantify the positioningaccuracy of machine tools and industrial robots. The work investigates the potentialincrease in positioning accuracy because of the simultaneous modelling of the kinematics,static deflections, vibrations and thermo-elasticity as a lumped-parametermodel of the machinery. Consequently the models can be used to quantify thechange of the accuracy throughout the workspace.The lumped parameter models presented in this work require empirical modelcalibration and validation. The success of both, calibration and validation, dependson the availability of the right measurement instruments, as these need to be ableto capture the actual positioning accuracy of machinery. This thesis focuses on theimportance of measurement instruments in industry and metrology and creates acatalogue of requirements and trends to identify the features of the measurementinstruments required for the factories of the future. These novel measurement instrumentsshall be able to improve model calibration and validation for an improvedoverall equipment effectiveness, improved product quality, reduced costs, improvedsafety and sustainability as a result of physics-based modelling and measurementof advanced manufacturing machinery.

  • Phase III study of pasireotide long-acting release in patients with metastatic neuroendocrine tumors and carcinoid symptoms refractory to available somatostatin analogues

    • In a randomized, double-blind, Phase III study, we compared pasireotide long-acting release (pasireotide LAR) with octreotide long-acting repeatable (octreotide LAR) in managing carcinoid symptoms refractory to first-generation somatostatin analogues. Adults with carcinoid tumors of the digestive tract were randomly assigned (1:1) to receive pasireotide LAR (60 mg) or octreotide LAR (40 mg) every 28 days. Primary outcome was symptom control based on frequency of bowel movements and flushing episodes. Objective tumor response was a secondary outcome. Progression-free survival (PFS) was calculated in a post hoc analysis. Adverse events were recorded. At the time of a planned interim analysis, the data monitoring committee recommended halting the study because of a low predictive probability of showing superiority of pasireotide over octreotide for symptom control (n=43 pasireotide LAR, 20.9%; n=45 octreotide LAR, 26.7%; odds ratio, 0.73; 95% confidence interval [CI], 0.27-1.97; P=0.53). Tumor control rate at month 6 was 62.7% with pasireotide and 46.2% with octreotide (odds ratio, 1.96; 95% CI, 0.89-4.32; P=0.09). Median (95% CI) PFS was 11.8 months (11.0 - not reached) with pasireotide versus 6.8 months (5.6 - not reached) with octreotide (hazard ratio, 0.46; 95% CI, 0.20-0.98; P=0.045). The most frequent drug-related adverse events (pasireotide vs octreotide) included hyperglycemia (28.3% vs 5.3%), fatigue (11.3% vs 3.5%), and nausea (9.4% vs 0%). We conclude that, among patients with carcinoid symptoms refractory to available somatostatin analogues, similar proportions of patients receiving pasireotide LAR or octreotide LAR achieved symptom control at month 6. Pasireotide LAR showed a trend toward higher tumor control rate at month 6, although it was statistically not significant, and was associated with a longer PFS than octreotide LAR.

    • The implementation of precision engineering principles in the design of precision systems is fundamental to achieve high accuracy. Although these principles may be independent of the system working range, their implementation is not. The working range, the accuracy, and the load of the system limit, for instance, the selection of materials, the structural design, or the positioning system. This article analyzes the applicability of precision engineering design principles depending on the working range of the system in order to establish their scalability or lack of it in small, medium, and large range machines and instruments.
