  • Association Between Telomere Length and Risk of Cancer and Non-Neoplastic Diseases A Mendelian Randomization Study

    • IMPORTANCE: The causal direction and magnitude of the association between telomere length and incidence of cancer and non-neoplastic diseases is uncertain owing to the susceptibility of observational studies to confounding and reverse causation. OBJECTIVE: To conduct a Mendelian randomization study, using germline genetic variants as instrumental variables, to appraise the causal relevance of telomere length for risk of cancer and non-neoplastic diseases. DATA SOURCES: Genomewide association studies (GWAS) published up to January 15, 2015. STUDY SELECTION: GWAS of noncommunicable diseases that assayed germline genetic variation and did not select cohort or control participants on the basis of preexisting diseases. Of 163 GWAS of noncommunicable diseases identified, summary data from 103 were available. DATA EXTRACTION AND SYNTHESIS: Summary association statistics for single nucleotide polymorphisms (SNPs) that are strongly associated with telomere length in the general population. MAIN OUTCOMES AND MEASURES: Odds ratios (ORs) and 95% confidence intervals (CIs) for disease per standard deviation (SD) higher telomere length due to germline genetic variation. RESULTS: Summary data were available for 35 cancers and 48 non-neoplastic diseases, corresponding to 420 081 cases (median cases, 2526 per disease) and 1 093 105 controls (median, 6789 per disease). Increased telomere length due to germline genetic variation was generally associated with increased risk for site-specific cancers. The strongest associations (ORs [ 95% CIs] per 1-SD change in genetically increased telomere length) were observed for glioma, 5.27 (3.15-8.81); serous low-malignant-potential ovarian cancer, 4.35 (2.39-7.94); lung adenocarcinoma, 3.19 (2.40-4.22); neuroblastoma, 2.98 (1.92-4.62); bladder cancer, 2.19 (1.32-3.66); melanoma, 1.87 (1.55-2.26); testicular cancer, 1.76 (1.02-3.04); kidney cancer, 1.55 (1.08-2.23); and endometrial cancer, 1.31 (1.07-1.61). Associations were stronger for rarer cancers and at tissue sites with lower rates of stem cell division. There was generally little evidence of association between genetically increased telomere length and risk of psychiatric, autoimmune, inflammatory, diabetic, and other non-neoplastic diseases, except for coronary heart disease (OR, 0.78 [ 95% CI, 0.67-0.90]), abdominal aortic aneurysm (OR, 0.63 [ 95% CI, 0.49-0.81]), celiac disease (OR, 0.42 [ 95% CI, 0.28-0.61]) and interstitial lung disease (OR, 0.09 [ 95% CI, 0.05-0.15]). CONCLUSIONS AND RELEVANCE: It is likely that longer telomeres increase risk for several cancers but reduce risk for some non-neoplastic diseases, including cardiovascular diseases.

  • Isotope effect on the formation of CN by radiative association reactions

    • The formation of CN and its isotopologue (CN)-N-15 through radiative association has been investigated. We considered two processes for the collisions of ground state C(P-3) and N(S-4) atoms (A(2)Pi -> X-2 Sigma(+) and b(4)Pi -> a(4)Sigma(+)), as well as, for the first time, we investigated the radiative association of ground state C(P-3) and excited N(D-2) atoms (B-2 Sigma(+) -> X-2 Sigma(+)). The cross-sections for the three processes have been calculated using semi-classical, quantum mechanical, and Breit-Wigner theories. The rate constants, derived from the combination of semi-classical and Breit-Wigner results, have been fitted to the Kooij formula to be used in astrochemical modeling. The rate constant for the B-2 Sigma(+) -> X-2 Sigma(+) process dominates in the investigated temperature range (10-10 000 K), when its own asymptotic energy is used as a reference level. Moreover, the B-2 Sigma(+) -> X-2 Sigma(+ )process presents the most pronounced isotope effect on the rate constant. We suggest considering these newly investigated radiative association processes for the formation of CN and (CN)-N-15 in the interstellar medium.

  • Semi-full-bridge submodule for modular multilevel converters

    • The energy variations in each arm of the modular multilevel converter comprises two components. The first component relates to the difference between the instantaneous input and output power of each phase-leg, and the second component relates to the energy which is moved back and forth between the two arms of the phase-leg. The latter component can be reduced or even eliminated if the peak-to-peak amplitude of the alternating voltage is greater than the pole-to-pole voltage of the dc link. This will, however, require submodules which can insert negative voltages. Therefore, a semi-full-bridge submodule which uses less semiconductors than the conventional full-bridge is proposed. Simulation results shows that by using the negative voltage-levels the capacitor voltage ripple can be reduced by up to 59%. Experimental results also shows that a 7-level voltage waveform can be generated using only one semi-full-bridge submodule with two capacitors per arm.

    • Across multiancestry groups, we analyzed Human Leukocyte Antigen (HLA) associations in over 176,000 individuals with Parkinson's disease (PD) and Alzheimer's disease (AD) versus controls. We demonstrate that the two diseases share the same protective association at the HLA locus. HLA-specific fine-mapping showed that hierarchical protective effects of HLA-DRB1*04 subtypes best accounted for the association, strongest with HLA-DRB1*04:04 and HLA-DRB1*04:07, and intermediary with HLA-DRB1*04:01 and HLA-DRB1*04:03. The same signal was associated with decreased neurofibrillary tangles in postmortem brains and was associated with reduced tau levels in cerebrospinal fluid and to a lower extent with increased Aβ42. Protective HLA-DRB1*04 subtypes strongly bound the aggregation-prone tau PHF6 sequence, however only when acetylated at a lysine (K311), a common posttranslational modification central to tau aggregation. An HLA-DRB1*04-mediated adaptive immune response decreases PD and AD risks, potentially by acting against tau, offering the possibility of therapeutic avenues.

    • This paper reports on fabrication and modeling of 4H- and 6H-SiC metal-semiconductor-metal (MSM) photodetectors (PDs). MSM PDs have been fabricated on 4H-SiC and 6H-SiC epitaxial layers, and their performance analyzed by MEDICI simulation and measurements. The simulations were also used to optimize the sensitivity by varying the width and spacing of the interdigitated electrodes. The fabricated PDs with 2 ÎŒm wide metal electrodes and 3 ÎŒm spacing between the electrodes exhibited, under UV illumination, a peak current to dark current ratio of 10 5 and 10 4 in 4H-SiC and 6H-SiC, respectively. The measured spectral responsivity of 6H-SiC PDs was higher compared to that of 4H-SiC PDs, with a cutoff at 407 nm compared to 384 nm in 4H-SiC PDs. Also the peak responsivity occurred at a shorter wavelength in 6H material. A high rejection ratio between the photocurrent and dark current was found in both cases. These experimental results were in agreement with simulation.

    • The interest in permanent-magnet synchronous motors:(PMSMs) is increasing in a wide area of applications. Since most PMSMs will operate without a shaft sensor in the future, valuable: information for experimental determination of machine parameters will be lost,In this paper, therefore, a method is presented where the induced EMF and the d-axis reactance are determined in a no-load test and the q-axis reactance is determined in a load test. The load angle delta is determined from the load test by means of a new analytical method. In this way, no separate measurement of the load angle is required. The method is especially suitable for line-start PMSMs which normally operate with negative d-axis current and, therefore, are not saturated in the d-axis flux paths. Moreover, the method is very simple to carry out for any laboratory technician, since the only tests that have to be made are standard tests which are made on standard induction motors on a regular basis.
