- We have identified a novel protein, Leprecan-like 1 (LEPREL1), with profound similarity to the Leprecan family of proteoglycans. The genomic organization of the Leprecan gene family was found to be highly conserved. Expression analysis shows that LEPREL1 is expressed in most tissues as a 3.4 kb transcript encoding an 80 kDa protein. A LEPREL1 specific antibody stains many cell types including adipocytes and neuroendocrine cells of the gastrointestinal epithelium. Muscle tissue contains a specific 6.5 kb transcript and a 200 kDa protein. The 3.4 kb LEPREL1 transcript encodes a 708 amino acid protein containing a signal sequence, four tetratricopeptide repeats (TPRs), a leucine zipper, a P-loop, a prolyl 4-hydroxylase alpha domain (P4Halpha), and a C-terminal KDEL ER-retention motif. LEPREL1 is localized to the ER and Golgi network and over-expressing it affects normal protein disulfide isomerase staining patterns in the ER.

Expression of TGF-beta isoforms, TGF-beta receptors, and SMAD molecules at different stages of human glioma

- Human gliomas express TGF-beta but, so far the expression of downstream mediators has been investigated in only a few cell lines. We have examined tissue specimens of 23 gliomas: 3 astrocytomas grade II (AST), 8 anaplastic astrocytomas grade III (AAST), and 12 glioblastoma multiforme grade IV (GBM). We analyzed the mRNA expression of TGF-beta1, TGF-beta2, TGF-beta3, the TGF-beta receptors type I (TbetaR-I) and type II (TbetaR-II), Smad2, Smad3, and Smad4. mRNA expression of IL-10 and CD95 (FAS/APO-1) were also studied. We detected increased mRNA levels of the 3 TGF-beta isoforms, correlating with the degree of malignancy. TGF-beta3 mRNA was increased, particularly in AST and AAST, while TGF-beta1 and TGF-beta2 mRNAs were strongly expressed in GBM. TGF-beta normally up-regulates the TGF-beta receptors, and TbetaR-I and TbetaR-II showed stronger expression in all gliomas when compared to normal tissues. However, the mRNA expression of Smad2, Smad3, and Smad4 was decreased in GBM. IL-10 mRNA expression was detected in glioma tissues but not in glioma cell lines. No marked increase in the expression of soluble CD95 splicing variants was found in the gliomas compared with normal tissue. However, total CD95 mRNA was elevated among GBM tissues.

- Transport by road is the most dominant mode of transportation in Europe today. An increased use of short sea shipping can balance the European transport system and contribute to a better environmental situation. The aim of this study is to identify and explore some characteristics of short sea shipping and to find general logistic weaknesses in the concept. By using a total logistic management approach, this study shows how short sea shipping is a competitive option with high environmental performance.We present a comparative study between different modes of transportation. A fictitious case of transportation of cargo between the Lake Vänern area in Sweden and Duisburg in Germany is set up and evaluated. The intermodal network between these two regions includes road, rail and shipping. The logistic quality of the total transport chain is measured as a weighted sum of performance parameters such as transport time, transport cost, frequency and flexibility, reliability, logistic management and service, environmental impact and political acceptability.The scope of the study is narrow, but by employing systems engineering techniques, interesting conclusions regarding a strengthened role of short sea shipping in a future European transport system has been made possible.
