  • Genetic associations at 53 loci highlight cell types and biological pathways relevant for kidney function
  • 2016
  • Ingår i: Nature Communications. - : Springer Science and Business Media LLC. - 2041-1723. ; 7
  • Tidskriftsartikel (refereegranskat)abstract
    • Reduced glomerular filtration rate defines chronic kidney disease and is associated with cardiovascular and all-cause mortality. We conducted a meta-analysis of genome-wide association studies for estimated glomerular filtration rate (eGFR), combining data across 133,413 individuals with replication in up to 42,166 individuals. We identify 24 new and confirm 29 previously identified loci. Of these 53 loci, 19 associate with eGFR among individuals with diabetes. Using bioinformatics, we show that identified genes at eGFR loci are enriched for expression in kidney tissues and in pathways relevant for kidney development and transmembrane transporter activity, kidney structure, and regulation of glucose metabolism. Chromatin state mapping and DNase I hypersensitivity analyses across adult tissues demonstrate preferential mapping of associated variants to regulatory regions in kidney but not extra-renal tissues. These findings suggest that genetic determinants of eGFR are mediated largely through direct effects within the kidney and highlight important cell types and biological pathways.

  • Gateway-based call admission in distributed object oriented systems
  • 2000
  • Ingår i: Proceedings fifteenth Nordic Teletraffic Seminar, NTS-15, Lund University, August 22-24, 2000.
  • Konferensbidrag (övrigt vetenskapligt/konstnärligt)abstract
    • Many applications in telecommunications will depend on distributed systems to provide enough capacity. In a distributed system a service is split up into a number of modules (often called objects) that can be placed at different nodes or processors in a network. A service can be seen as a number of invocations of the objects in a certain order. There are a number of performance problems which have to be solved. How shall objects be distributed on the nodes? How shall the load be distributed among the nodes for a given object distribution? How shall the distributed system be protected from temporary overload situations? In our paper we investigate these questions for distributed systems where requests for service arrive to a gateway from which they distributed to the nodes of the system. We assume that the object distribution is given and we concentrate on protecting the system using three different external load control mechanisms: Percent Thinning, Call Gapping and Tokens. Using simulations, we nd that Tokens provide the best system protection.

  • On overload control through queue length for web servers
  • 2002
  • Ingår i: Sixteenth Nordic Teletraffic Seminar NTS 16 : Helsinki University of Technology, Espoo August 21-23, 2002 : proceedings (Report / Helsinki University of Technology, Networking Laboratory). - 1458-0322.
  • Konferensbidrag (övrigt vetenskapligt/konstnärligt)abstract
    • We investigate the performance of overload control through queue length for two different web server architectures. Both of them use finite queue lengths to prevent servers from being overloaded. One architecture prioritizes requests from established sessions while the other treats all requests equally. First, we introduce queueing models for these two systems. Then, we define and explain a new web server performance metric that is a function of session throughput, error rate for connection within sessions and average request response time. Finally, we use simulation to evaluate the performance of these two types of web servers. The result suggests that the benefit of request prioritization is noticeable only when the capacities of the sub-systems match each other.

  • Determinants of morbidity and mortality following emergency abdominal surgery in children in low-income and middle-income countries
  • 2016
  • Ingår i: BMJ Global Health. - : BMJ Publishing Group Ltd. - 2059-7908. ; 1:4
  • Tidskriftsartikel (refereegranskat)abstract
    • Background: Child health is a key priority on the global health agenda, yet the provision of essential and emergency surgery in children is patchy in resource-poor regions. This study was aimed to determine the mortality risk for emergency abdominal paediatric surgery in low-income countries globally. Methods: Multicentre, international, prospective, cohort study. Self-selected surgical units performing emergency abdominal surgery submitted prespecified data for consecutive children aged <16 years during a 2-week period between July and December 2014. The United Nation's Human Development Index (HDI) was used to stratify countries. The main outcome measure was 30-day postoperative mortality, analysed by multilevel logistic regression. Results: This study included 1409 patients from 253 centres in 43 countries; 282 children were under 2 years of age. Among them, 265 (18.8%) were from low-HDI, 450 (31.9%) from middle-HDI and 694 (49.3%) from high-HDI countries. The most common operations performed were appendectomy, small bowel resection, pyloromyotomy and correction of intussusception. After adjustment for patient and hospital risk factors, child mortality at 30 days was significantly higher in low-HDI (adjusted OR 7.14 (95% CI 2.52 to 20.23), p<0.001) and middle-HDI (4.42 (1.44 to 13.56), p=0.009) countries compared with high-HDI countries, translating to 40 excess deaths per 1000 procedures performed. Conclusions: Adjusted mortality in children following emergency abdominal surgery may be as high as 7 times greater in low-HDI and middle-HDI countries compared with high-HDI countries. Effective provision of emergency essential surgery should be a key priority for global child health agendas.

  • Overload control strategies in a multiprocessor system with distributed data
  • 1995
  • Ingår i: Proceedings of the 5th IEE Conference on Telecommunications, Brighton, UK, March 1995. - 0852966342 ; , s. 145-149
  • Konferensbidrag (refereegranskat)abstract
    • This paper addresses overload control issues for distributed control systems, i.e. multiprocessor systems, in telephone exchanges, signalling transfer points, service control points, home location registers, etc. Three different strategies for rejecting calls, due to severe overload conditions, are examined. The strategies differ in that they allow call rejection during different time epochs of the entire life time of the call. The optimal solution, i.e. the largest throughput, can only be obtained by a centrally placed overload control mechanism which collects data from all processors in the system

  • From the First Selective Non-Peptide AT(2) Receptor Agonist to Structurally Related Antagonists
  • 2012
  • Ingår i: Journal of Medicinal Chemistry. - : American Chemical Society (ACS). - 0022-2623 .- 1520-4804. ; 55:5, s. 2265-2278
  • Tidskriftsartikel (refereegranskat)abstract
    • A para substitution pattern of the phenyl ring is a characteristic feature of the first reported selective AT(2) receptor agonist M024/C21 (1) and all the nonpeptidic AT(2) receptor agonists described so far. Two series of compounds structurally related to 1 but with a meta substitution pattern have now been synthesized and biologically evaluated for their affinity to the AT(1) and AT(2) receptors. A high AT(2)/AT(1) receptor selectivity was obtained with all 41 compounds synthesized, and the majority exhibited K-i ranging from 2 to 100 nM. Five compounds were evaluated for their functional activity at the AT(2) receptor, applying a neurite outgrowth assay in NG108-15 cells.. Notably, four of the five compounds, with representatives from both series, acted as potent AT(2) receptor antagonists. These compounds were found to be considerably more effective than PD 123,319, the standard AT(2) receptor antagonist used in most laboratories. No AT(2) receptor antagonists were previously reported among the derivatives with a para substitution pattern. Hence, by a minor modification of the agonist 1 it could be transformed into the antagonist, compound 38. These compounds should serve as valuable tools in the assessment of the role of the AT(2) receptor in more complex physiological models.
