  • THE STRUCTURE AND EMISSION MODEL OF THE RELATIVISTIC JET IN THE QUASAR 3C 279 INFERRED FROM RADIO TO HIGH-ENERGY gamma-RAY OBSERVATIONS IN 2008-2010

    • We present time-resolved broadband observations of the quasar 3C 279 obtained from multi-wavelength campaigns conducted during the first two years of the Fermi Gamma-ray Space Telescope mission. While investigating the previously reported gamma-ray/optical flare accompanied by a change in optical polarization, we found that the optical emission appears to be delayed with respect to the gamma-ray emission by about 10 days. X-ray observations reveal a pair of isolated flares separated by similar to 90 days, with only weak gamma-ray/optical counterparts. The spectral structure measured by Spitzer reveals a synchrotron component peaking in the mid-infrared band with a sharp break at the far-infrared band during the gamma-ray flare, while the peak appears in the millimeter (mm)/submillimeter (sub-mm) band in the low state. Selected spectral energy distributions are fitted with leptonic models including Comptonization of external radiation produced in a dusty torus or the broad-line region. Adopting the interpretation of the polarization swing involving propagation of the emitting region along a curved trajectory, we can explain the evolution of the broadband spectra during the gamma-ray flaring event by a shift of its location from similar to 1 pc to similar to 4 pc from the central black hole. On the other hand, if the gamma-ray flare is generated instead at sub-pc distance from the central black hole, the far-infrared break can be explained by synchrotron self-absorption. We also model the low spectral state, dominated by the mm/sub-mm peaking synchrotron component, and suggest that the corresponding inverse-Compton component explains the steady X-ray emission.

  • Congenital Zika virus infection as a silent pathology with loss of neurogenic output in the fetal brain

    • Zika virus (ZIKV) is a flavivirus with teratogenic effects on fetal brain, but the spectrum of ZIKV-induced brain injury is unknown, particularly when ultrasound imaging is normal. In a pregnant pigtail macaque (Macaca nemestrina) model of ZIKV infection, we demonstrate that ZIKV-induced injury to fetal brain is substantial, even in the absence of microcephaly, and may be challenging to detect in a clinical setting. A common and subtle injury pattern was identified, including (i) periventricular T2-hyperintense foci and loss of fetal noncortical brain volume, (ii) injury to the ependymal epithelium with underlying gliosis and (iii) loss of late fetal neuronal progenitor cells in the subventricular zone (temporal cortex) and subgranular zone (dentate gyrus, hippocampus) with dysmorphic granule neuron patterning. Attenuation of fetal neurogenic output demonstrates potentially considerable teratogenic effects of congenital ZIKV infection even without microcephaly. Our findings suggest that all children exposed to ZIKV in utero should receive long-term monitoring for neurocognitive deficits, regardless of head size at birth.

  • STRONG FAR-INFRARED COOLING LINES, PECULIAR CO KINEMATICS, AND POSSIBLE STAR-FORMATION SUPPRESSION IN HICKSON COMPACT GROUP 57

    • We present [C II] and [O I] observations from Herschel and CO(1-0) maps from the Combined Array for Research in Millimeter Astronomy (CARMA) of the Hickson compact group HCG 57, focusing on the galaxies HCG 57a and HCG 57d. HCG 57a has been previously shown to contain enhanced quantities of warm molecular hydrogen consistent with shock or turbulent heating. Our observations show that HCG 57d has strong [C II] emission compared to L-FIR and weak CO(1-0), while in HCG 57a, both the [C II] and CO(1-0) are strong. HCG 57a lies at the upper end of the normal distribution of the [C II]/CO and [C II]/FIR ratios, and its far-infrared (FIR) cooling supports a low-density, warm, diffuse gas that falls close to the boundary of acceptable models of a photon-dominated region. However, the power radiated in the [C II] and warm H-2 emissions have similar magnitudes, as seen in other shock-dominated systems and predicted by recent models. We suggest that shock heating of the [C II] is a viable alternative to photoelectric heating in violently disturbed, diffuse gas. The existence of shocks is also consistent with the peculiar CO kinematics in the galaxy, indicating that highly noncircular motions are present. These kinematically disturbed CO regions also show evidence of suppressed star formation, falling a factor of 10-30 below normal galaxies on the Kennicutt-Schmidt relation. We suggest that the peculiar properties of both galaxies are consistent with a highly dissipative, off-center collisional encounter between HCG 57d and 57a, creating ring-like morphologies in both systems. Highly dissipative gas-on-gas collisions may be more common in dense groups because of the likelihood of repeated multiple encounters. The possibility of shock-induced star-formation suppression may explain why a subset of these HCG galaxies has been found previously to fall in the mid-infrared green valley.

  • Hyaluronidase Impairs Neutrophil Function and Promotes Group B Streptococcus Invasion and Preterm Labor in Nonhuman Primates

    • Invasive bacterial infections during pregnancy are a major risk factor for preterm birth, stillbirth, and fetal injury. Group B streptococci (GBS) are Gram-positive bacteria that asymptomatically colonize the lower genital tract but infect the amniotic fluid and induce preterm birth or stillbirth. Experimental models that closely emulate human pregnancy are pivotal for the development of successful strategies to prevent these adverse pregnancy outcomes. Using a unique nonhuman primate model that mimics human pregnancy and informs temporal events surrounding amniotic cavity invasion and preterm labor, we show that the animals inoculated with hyaluronidase (HylB)-expressing GBS consistently exhibited microbial invasion into the amniotic cavity, fetal bacteremia, and preterm labor. Although delayed cytokine responses were observed at the maternal-fetal interface, increased prostaglandin and matrix metalloproteinase levels in these animals likely mediated pre term labor. HylB-proficient GBS dampened reactive oxygen species production and exhibited increased resistance to neutrophils compared to an isogenic mutant. Together, these findings demonstrate how a bacterial enzyme promotes GBS amniotic cavity invasion and preterm labor in a model that closely resembles human pregnancy. IMPORTANCE Group B streptococci (GBS) are bacteria that commonly reside in the female lower genital tract as asymptomatic members of the microbiota. However, during pregnancy, GBS can infect tissues at the maternal-fetal interface, leading to preterm birth, stillbirth, or fetal injury. Understanding how GBS evade host defenses during pregnancy is key to developing improved preventive therapies for these adverse outcomes. In this study, we used a unique nonhuman primate model to show that an enzyme secreted by GBS, hyaluronidase (HylB) promotes bacterial invasion into the amniotic cavity and fetus. Although delayed immune responses were seen at the maternal-fetal interface, animals infected with hyaluronidase-expressing GBS exhibited premature cervical ripening and preterm labor. These observations reveal that HylB is a crucial GBS virulence factor that promotes bacterial invasion and preterm labor in a pregnancy model that closely emulates human pregnancy. Therefore, hyaluronidase inhibitors may be useful in therapeutic strategies against ascending GBS infection.

    • This study aims to unravel the spatial and temporal distribution of diagenetic alterations of the Mheiherrat Member, the Rudeis Formation (lower Miocene) of the Gulf of Suez rift, Egypt within depositional fades and sequence stratigraphy. The Mheiherrat member is represented by shoreface calcarenites and hybrid arenites (transgressive and highstand systems tracts; TST and HST; respectively) and deltaic rudites and coarse-grained calcarenites and hybrid arenites (lowstand systems tracts; LST). Petrographic, stable O- and C-isotopes, mineral chemical and geochemical analyses have revealed that the arenites are pervasively cemented by eogenetic carbonates and, to small extent, by zeolite and pyrite as well as by telogenetic palygorskite. The shoreface TST and HST calcarenites and hybrid arenites were dominantly cemented by microcrystalline grain-rimming and inter- and intragranular pore-filling calcite (delta O-13(V-PDB) = -3.6 parts per thousand to -0.3 parts per thousand and delta C-13(V-PDB) = -2.3 parts per thousand to -0.7 parts per thousand) and rhombic dolomite (delta C-13(V-PDB) = -3.9 parts per thousand to +0.9 parts per thousand and delta C-13(V-PDB) = -2.5 parts per thousand to -0.7 parts per thousand). These cements are interpreted to be formed by marine to brackish waters with delta O-13(V-SMOW) -1.2 parts per thousand to +3.2 parts per thousand, at temperature of 20-55 degrees C. The deltaic LST coarse-grained calcarenites and hybrid arenites were dominantly cemented by coarse-crystalline, inter- and intragranular pore-filling calcite (delta O-13(V-PDB) -4.4 parts per thousand to -2.3% and delta C-13(V-PDB) = -2.8 parts per thousand. to -1.3 parts per thousand.), which are interpreted to have precipitated from pore waters with delta O-13(V-SWOM) +3.5 parts per thousand. to +5.5 parts per thousand, at temperatures of greater than 55 degrees C. Such pervasive cementation by carbonates is attributed to the occurrence of abundant intrabasinal carbonate grains. The carbonate cement texture is suggested to be controlled by changes in pore-water chemistry owing to transgression and regression events. This case study revealed that better understanding of factors controlling the extent and textural habits of carbonate cements can be achieved when it is linked with depositional facies and sequence stratigraphy.

  • Diagenetic Alterations and Reservoir Quality Evolution of Lower Cretaceous Fluvial Sandstones : Nubian Formation, Sirt Basin, North-Central Libya

    • Lower Cretaceous meandering and braided fluvial sandstones of the Nubian Formation form some of the most important subsurface reservoir rocks in the Sirt Basin, north-central Libya. Mineralogical, petrographical and geochemical analyses of sandstone samples from well BB6-59, Sarir oilfield, indicate that the meandering fluvial sandstones are fine- to very fine-grained subarkosic arenites (av. Q(91)F(5)L(4)), and that braided fluvial sandstones are medium- to very coarse-grained quartz arenites (av. Q(96)F(3)L(1)). The reservoir qualities of these sandstones were modified during both eodiagenesis (ca. <70 degrees C; <2 km) and mesodiagenesis (ca. >70 degrees C; >2 km). Reservoir quality evolution was controlled primarily by the dissolution and kaolinitization of feldspars, micas and mud intraclasts during eodiagenesis, and by the amount and thickness of grain-coating clays, chemical compaction and quartz overgrowths during mesodiagenesis. However, dissolution and kaolinitization of feldspars, micas and mud intraclasts resulted in the creation of intercrystalline micro- and mouldic macro-porosity and permeability during eodiagenesis, which were more widespread in braided fluvial than in meandering fluvial sandstones. This was because of the greater depositional porosity and permeability in the braided fluvial sandstones which enhanced percolation of meteoric waters. The development of only limited quartz overgrowths in the braided fluvial sandstones, in which quartz grains are coated by thick illite layers, retained high porosity and permeability (12-23% and 30-600 mD). By contrast, meandering fluvial sandstones underwent porosity loss as a result of quartz overgrowth development on quartz grains which lack or have thin and incomplete grain-coating illite (2-15% and 0-0.1 mD). Further loss of porosity in the meandering fluvial sandstones occurred as a result of chemical compaction (pressure dissolution) induced by the occurrence of micas along grains contacts. Other diagenetic alterations, such as the growth of pyrite, siderite, dolomite/ankerite and albitization, had little impact on reservoir quality. The albitization of feldspars may have had minor positive influence on reservoir quality through the creation of intercrystalline micro-porosity between albite crystals. The results of this study show that diagenetic modifications of the braided and meandering fluvial sandstones in the Nubian Formation, and resulting changes in reservoir quality, are closely linked to depositional porosity and permeability. They are also linked to the thickness of grain-coating infiltrated clays, and to variations in detrital composition, particularly the amounts of mud intraclasts, feldspars and mica grains as well as climatic conditions.

    • The reactivity of the α-phase of Al-Zn (Zn-68 wt% Al, Al 5.2 Zn) in deaerated 0.1 M NaOH solution (simulating industrial pretreatments) was investigated and compared with that of pure Al and Zn. The elementary phenomena of metal oxidation, dissolution, oxide formation, and hydrogen evolution were decoupled using atomic emission spectroelectrochemistry. At the open-circuit potential, the Al 5.2 Zn phase reacted similarly as pure Al, undergoing selective Al dissolution to form a Zn(0) enriched layer. The Zn in the alloy shifted the potential to just below the onset of Zn dissolution. Elementary polarization curves showed that Zn dissolution was similar for the Al 5.2 Zn phase as for pure Zn. Near the open-circuit potential, Zn dissolution was faradaic limited by the formation of surface Zn(OH) 2 . At higher temperature, significant amounts of ZnO formed resulting in passivation. For the Al 5.2 Zn phase, the rates of Al and Zn dissolution were determined by a charge transfer mechanism across the ZnO film. Kinetic parameters (activation energies and Tafel slopes) were measured for some of the elementary processes.

    • The surface reactivity and the elemental dissolution of a high-strength aerospace alloy (AA7449-T651) during acid pickling was investigated with element-resolved electrochemistry (AESEC). The results demonstrated a sequential dissolution of Mg, Al, Zn and Cu, following the order of their respective galvanic reactivity in common acid pickling solutions. The enrichment of Cu during pickling with the H2SO4 solution was observed. The effect of oxidizing solution components HNO3 and Fe(III) on Cu dissolution were investigated using an experimental Evans diagram approach.

    • Acid pickling is a one-step process prior to the conversion coating of aluminum alloys, an alternative to the two-step degreasing-deoxidizing. Selective dissolution may lead to enrichment or depletion of alloying elements with important consequences for corrosion resistance. In this work, the mechanisms of selective dissolution were explored using element-resolved electrochemistry for Al-3at.%Cu and Al-3at.%Mg, representing two extremes of the galvanic series. Pickling reactions were investigated in sulfuric and nitric acids with and without Fe(III) additives. The microstructure of the Al-3at.%Cu alloy was varied by heat treatment. Al-3at.%Mg showed congruent dissolution throughout the steady state in all electrolytes while Al-3at.%Cu underwent selective dissolution with surface enrichment of Cu and a significant release of detached Cu-rich particles in sulfuric acid. With the presence of Fe(III) in the electrolyte, Cu dissolved congruently, and the dissolution kinetics of these two alloys were also markedly enhanced. Experimental Evans diagrams were determined to clarify the electrochemical nature of the reactions. © 2023 The Author(s)

  • Neurosteroid activation of gaba-a receptors : a potential treatment target for symptoms in primary biliary cholangitis?

    • Background and Aims: A third of patients with primary biliary cholangitis (PBC) experience poorly understood cognitive symptoms, with a significant impact on quality of life (QOL), and no effective medical treatment. Allopregnanolone, a neurosteroid, is a positive allosteric modulator of gamma-aminobutyricacid-A (GABA-A) receptors, associated with disordered mood, cognition, and memory. This study explored associations between allopregnanolone and a disease-specific QOL scoring system (PBC-40) in PBC patients.Method: Serum allopregnanolone levels were measured in 120 phenotyped PBC patients and 40 age and gender-matched healthy controls. PBC subjects completed the PBC-40 at recruitment. Serum allopregnanolone levels were compared across PBC-40 domains for those with none/mild symptoms versus severe symptoms.Results: There were no overall differences in allopregnanolone levels between healthy controls (median = 0.03 ng/ml (IQR = 0.025)) and PBC patients (0.031 (0.42), p = 0.42). Within the PBC cohort, higher allopregnanolone levels were observed in younger patients (r (120) = -0.53, p < 0.001) but not healthy controls (r (39) = -0.21, p = 0.21). Allopregnanolone levels were elevated in the PBC-40 domains, cognition (u = 1034, p = 0.02), emotional (u = 1374, p = 0.004), and itch (u = 795, p = 0.03). Severe cognitive symptoms associated with a younger age: severe (50 (12)) vs. none (60 (13); u = 423 p = 0.001).Conclusion: Elevated serum allopregnanolone is associated with severe cognitive, emotional, and itch symptoms in PBC, in keeping with its known action on GABA-A receptors. Existing novel compounds targeting allopregnanolone could offer new therapies in severely symptomatic PBC, satisfying a significant unmet need.
