  • Plant functional trait change across a warming tundra biome

    • The tundra is warming more rapidly than any other biome on Earth, and the potential ramifications are far-reaching because of global feedback effects between vegetation and climate. A better understanding of how environmental factors shape plant structure and function is crucial for predicting the consequences of environmental change for ecosystem functioning. Here we explore the biome-wide relationships between temperature, moisture and seven key plant functional traits both across space and over three decades of warming at 117 tundra locations. Spatial temperature–trait relationships were generally strong but soil moisture had a marked influence on the strength and direction of these relationships, highlighting the potentially important influence of changes in water availability on future trait shifts in tundra plant communities. Community height increased with warming across all sites over the past three decades, but other traits lagged far behind predicted rates of change. Our findings highlight the challenge of using space-for-time substitution to predict the functional consequences of future warming and suggest that functions that are tied closely to plant height will experience the most rapid change. They also reveal the strength with which environmental factors shape biotic communities at the coldest extremes of the planet and will help to improve projections of functional changes in tundra ecosystems with climate warming.

    • RHO family proteins are important for the function of inflammatory cells. They are modified with a 20-carbon geranylgeranyl lipid in a process catalyzed by protein geranylgeranyltransferase type I (GGTase-I). Geranylgeranylation is viewed as essential for the membrane targeting and activity of RHO proteins. Consequently, inhibiting GGTase-I to interfere with RHO protein activity has been proposed as a strategy to treat inflammatory disorders. However, here we show that mice lacking GGTase-I in macrophages develop severe joint inflammation resembling erosive rheumatoid arthritis. The disease was initiated by the GGTase-I-deficient macrophages and was transplantable and reversible in bone marrow transplantation experiments. The cells accumulated high levels of active GTP-bound RAC1, CDC42, and RHOA, and RAC1 remained associated with the plasma membrane. Moreover, GGTase-I deficiency activated p38 and NF-κB and increased the production of proinflammatory cytokines. The results challenge the view that geranylgeranylation is essential for the activity and localization of RHO family proteins and suggest that reduced geranylgeranylation in macrophages can initiate erosive arthritis.

    • RAS and RHO proteins, which contribute to tumorigenesis and metastasis, undergo posttranslational modification with an isoprenyl lipid by protein farnesyltransferase (FTase) or protein geranylgeranyltransferase-I (GGTase-I). Inhibitors of FTase and GGTase-I were developed to block RAS-induced malignancies, but their utility has been difficult to evaluate because of off-target effects, drug resistance, and toxicity. Moreover, the impact of FTase deficiency and combined FTase/GGTase-I deficiency has not been evaluated with genetic approaches. We found that inactivation of FTase eliminated farnesylation of HDJ2 and H-RAS, prevented H-RAS targeting to the plasma membrane, and blocked proliferation of primary and K-RAS(G12D)-expressing fibroblasts. FTase inactivation in mice with K-RAS-induced lung cancer reduced tumor growth and improved survival, similar to results obtained previously with inactivation of GGTase-I. Simultaneous inactivation of FTase and GGTase-I markedly reduced lung tumors and improved survival without apparent pulmonary toxicity. These data shed light on the biochemical and therapeutic importance of FTase and suggest that simultaneous inhibition of FTase and GGTase-I could be useful in cancer therapeutics.

  • On-board Impedance Diagnostics Method of Li-ion Traction Batteries Using Pseudo-Random Binary Sequences

    • This work deals with the on-board impedance measurements of Li-ion batteries on hybrid electric vehicles/electric vehicles by using pseudo-random binary sequences (PRBSs). The impedance of the battery can be related to its state of charge (SOC) and state of health (SOH). However the battery impedance is frequency dependent which makes it difficult to perform the measurement on a pack level in the vehicles. By using an excitation signal like PRBS, it is possible to extract the impedance information of the battery packs. Experiments are performed with different set-ups to verify the PRBS method. A non-parametric method is used to process the data and extract the impedance measurement. Experiments in the laboratory at different state of charge levels and temperatures are made to validate the PRBS method. It is shown that the PRBS method can produce a valid electrochemical impedance spectrum in a limited frequency range, similar to the result from a high accuracy laboratory impedance analyzer. The method is stable at different SOC levels and temperatures. A simulation of the excitation signal in the vehicle is performed where the electric motor is used as the load. It shows that it is possible to some extent to use the driveline in a hybrid electric vehicle/electric vehicle to perform an on-board battery impedance measurement.

  • PFAS – A threat for groundwater and drinking water supply in Sweden?

    • Perfluoroalkyl substances (PFAS) are a group of anthropogenic environmental pollutants that are widely distributed in the global environment. They have multiple industrial uses, including water repellents in clothing, paper coatings and firefighting foam. According to a study released by the Environmental Directorate of the OECD, they are persistent, bioaccumulative and toxic to mammalian species (OECD, 2002). In some municipal drinking water wells in Sweden, measured concentrations of PFAS found to be several hundred times higher than the allowed threshold values. This has created a huge public concern and has recently attracted much media attention in Sweden (e.g. Afzelius et al., 2014; Bergman et al., 2014; Lewis et al., 2014). PFAS findings raised questions such as “What can we do to solve the problem?” When it comes to drinking water, there are a number of techniques that can ensure that PFAS levels are reduced to acceptable levels. This may be a costly challenge, but from a technical point of view it is possible. To ensure the safety of drinking water from a public health perspective is obviously a top priority. However, international experience shows that the cost of cleaning up PFAS in groundwater may be significantly higher than continuously treat drinking water in water works. Approximately fifty percent of Sweden’s drinking water comes from groundwater. As a result, there are several ongoing and planned PFAS-related environmental and drinking-water investigations in Sweden. Many aquifers that supply municipal water plants are located in areas of sand and gravel deposits. Such soils have relatively high permeabilities, which permits extraction of large volumes of water. However, the downside to high permeabilities is that they also allow dissolved contaminants as PFAS to spread over large areas. If one disregards the health risks linked to its presence in drinking water, PFAS have an impact on three of Sweden’s national environmental quality objectives, namely, A Non-Toxic Environment, Flourishing Lakes and Streams and Good-Quality Groundwater. Although the survey of PFAS in our groundwater supplies will take time, it is feasible. Much research in the field of hydrogeology and geochemistry remains before a viable and cost-effective groundwater remediation method can be operational. Until then, it is essential that measures are taken to identify the present distribution and magnitude of PFAS in groundwater and prevents its further spread in our most important aquifers.   • Antipredator defence in butterflies

    • Predation exerts a potent selection mechanism and has resulted in a suite of antipredation adaptations encompassing morphological and behavioral traits in prey. In butterflies, several such traits appear to be directed towards birds which are considered as one of their major predators. In this thesis I have investigated tactics by which adult butterflies may survive close encounters with birds. In paper I, I provide supporting evidence that the small white “comma” on females of the comma butterfly, Polygonia c-album, has a distractive function on blue tits and so reduces the attack risk. In paper II & III, I investigate the antipredator efficiency of sudden wing-flicking in two species, the peacock butterfly, Inachis io, and the European swallowtail, Papilio machaon, when confronted with domestic fowl and great tits, respectively. Peacock butterflies were manipulated to either display visible or painted over eyespots. Interestingly, the birds that confronted peacocks with visible eyespots were more likely to utter antipredator alarm calls, which imply that the eyespots may be perceived as real eyes of a potential predator. On the whole, wing-flicking in both species typically induced evasion in the birds which suggests that the birds became frightened rather than perceiving the butterflies as not profitable to attack for some other reason. Moreover, I use blue tits as predators to investigate the possible function of smaller eyespots of satyrine butterflies in that they serve to divert predator attacks. Evidence suggests that low light conditions accentuated in the UV may enhance the deflective function of marginal eyespots in the woodland brown butterfly, Lopinga achine (paper IV). In paper V, I show that the presence of one marginal eyespot on the speckled wood butterfly, Pararge aegeria, can deflect bird attacks; moreover, when the butterfly is concealed against the background, eyespots can also increase the latency time until bird attack. In conclusion, my thesis underscores that behavioral studies of predators are instrumental to aid our understanding of the adaptive significance of certain behavioral and morphological traits in prey.

  • Bird attacks on a butterfly with marginal eyespots and the role of prey concealment against the background : Marginal eyespots can deflect bird attacks

    • Small eyespots on butterflies have long been thought to deflect attacks and birds are presumptive drivers selecting for these patterns. However, evidence of this function is still ambiguous. Marginal eyespots typically consist of a UV-reflective white pupil, surrounded by one black and one yellowish ring. We have recently shown that blue tits attack such eyespots, but only under low light intensities with accentuated UV-levels. An increased salience of the eyespots relative to the rest of the butterfly probably explains this result. Possibly, a background against which the butterfly is concealed may deceive birds to making similar errors. We therefore presented speckled wood butterflies provided with eyespots (or controls without eyespots) to blue tits against two backgrounds, oak- and birch bark. Results show that (i) eyespots, independent of background, were effective in deflecting attacks, (ii) the time elapsed between a bird’s landing and attack was interactively dependent on background and whether the butterfly bore an eyespot and (iii) the speed at which a butterfly was attacked predicted the outcome, with faster birds being more prone to errors than slower birds. This underscores a speed-accuracy tradeoff in the predators and that background plays a role in the defensive qualities of marginal eyespots.
