  • Beyond the global brain differences : intraindividual variability differences in 1q21.1 distal and 15q11.2 bp1-bp2 deletion carriers

    • Background: Carriers of the 1q21.1 distal and 15q11.2 BP1-BP2 copy number variants exhibit regional and global brain differences compared with noncarriers. However, interpreting regional differences is challenging if a global difference drives the regional brain differences. Intraindividual variability measures can be used to test for regional differences beyond global differences in brain structure.Methods: Magnetic resonance imaging data were used to obtain regional brain values for 1q21.1 distal deletion (n = 30) and duplication (n = 27) and 15q11.2 BP1-BP2 deletion (n = 170) and duplication (n = 243) carriers and matched noncarriers (n = 2350). Regional intra-deviation scores, i.e., the standardized difference between an individual's regional difference and global difference, were used to test for regional differences that diverge from the global difference.Results: For the 1q21.1 distal deletion carriers, cortical surface area for regions in the medial visual cortex, posterior cingulate, and temporal pole differed less and regions in the prefrontal and superior temporal cortex differed more than the global difference in cortical surface area. For the 15q11.2 BP1-BP2 deletion carriers, cortical thickness in regions in the medial visual cortex, auditory cortex, and temporal pole differed less and the prefrontal and somatosensory cortex differed more than the global difference in cortical thickness.Conclusions: We find evidence for regional effects beyond differences in global brain measures in 1q21.1 distal and 15q11.2 BP1-BP2 copy number variants. The results provide new insight into brain profiling of the 1q21.1 distal and 15q11.2 BP1-BP2 copy number variants, with the potential to increase understanding of the mechanisms involved in altered neurodevelopment.

    • Adenoid cystic carcinoma (ACC) of the head and neck is a malignant tumor with poor long-term prognosis. Besides the recently identified MYB-NFIB fusion oncogene generated by a t(6;9) translocation, little is known about other genetic alterations in ACC. Using high-resolution, array-based comparative genomic hybridization, and massively paired-end sequencing, we explored genomic alterations in 40 frozen ACCs. Eighty-six percent of the tumors expressed MYB-NFIB fusion transcripts and 97% overexpressed MYB mRNA, indicating that MYB activation is a hallmark of ACC. Thirty-five recurrent copy number alterations (CNAs) were detected, including losses involving 12q, 6q, 9p, 11q, 14q, 1p, and 5q and gains involving 1q, 9p, and 22q. Grade III tumors had on average a significantly higher number of CNAs/tumor compared to Grade I and II tumors (P = 0.007). Losses of 1p, 6q, and 15q were associated with high-grade tumors, whereas losses of 14q were exclusively seen in Grade I tumors. The t(6;9) rearrangements were associated with a complex pattern of breakpoints, deletions, insertions, inversions, and for 9p also gains. Analyses of fusion-negative ACCs using high-resolution arrays and massively paired-end sequencing revealed that MYB may also be deregulated by other mechanisms in addition to gene fusion. Our studies also identified several down-regulated candidate tumor suppressor genes (CTNNBIP1, CASP9, PRDM2, and SFN) in 1p36.33-p35.3 that may be of clinical significance in high-grade tumors. Further, studies of these and other potential target genes may lead to the identification of novel driver genes in ACC.

    • Organic-rich pulp and paper mill sludge (PPMS) has the potential to become a renewable carbon source for producing alternatives to fossil-based product. In this work, PPMS treated by hydrothermal carbonization (HTC) was investigated based on its pyrolysis properties. The pyrolytic mechanism, kinetics data and product of the sample were studied using TG as well as pyrolysis tests in Py-GC/MS and a bench-scale reactor at 450, 550, and 650 degrees C. The results show that the thermal decomposition of feedstock is a two-stage reaction. The mean activation energy of the pyrolysis of HTC treated PPMS was estimated as 233.08 kl/mol, which is higher than that of the pyrolysis of paper sludge reported before. The changes in enthalpies, entropies and Gibbs free energies from the reactants to the activated complex were estimated. The concentration of monocyclic aromatic hydrocarbons in the derived organic liquid fraction shows a positive correlation with the pyrolysis temperature. At 550 degrees C, the organic liquid fraction reached its highest yield at 13.7% with an oxygen level of 10.7 wt% and a higher heating value of 35.9 MJ/ kg. The pyrolytic chars show that a molar ratio of O:C is less than 0.2, which shows potential for use as a carbon sink.

  • Assessment of Self-Heating in Wood Pellets by FE Modelling

    • The self-heating process in a laboratory scale experiment has been modelled using the Comsol Multiphysics software. In the simulations the gas flow and air movement in the volume and heat diffusion in the bulk were taken into account however only one reaction in the pellets bulk is considered. The input data is found from measurements of the reaction chemistry and the heat transfer properties. It is found that all relevant physics is needed in order to obtain reasonable predictions in particular the heat transfer between the bulk and the gas is important but also condensation and evaporation of moisture.
