  • Deep eutectic solvent delignification to low-energy mechanical pulp to produce papermaking fibers

    • A novel process based on low-energy mechanical pulp and deep eutectic solvents (DESs) was evaluated with the goal of producing fibers suitable for papermaking. Ideally, these fibers could be produced at much lower costs, especially when applied to an existing paper mill equipped with a thermomechanical pulp (TMP) production line that was threatened with shutdown due to the decreasing demand for wood-containing paper grades. The efficiency of DES delignification in Teflon-coated autoclaves and in a specially designed non-standard flow extractor was evaluated. All tested DESs had choline chloride ([Ch]Cl) as the hydrogen bond acceptor. Lactic acid, oxalic acid, malic acid, or urea acted as hydrogen bond donors. The temperatures and times of the delignification tests were varied. Chemical analysis of the pulp samples revealed that DESs containing lactic acid, oxalic acid, or urea decreased the lignin content by approximately 50%. The DES delignification based on [Ch]Cl and urea exhibited good hemicellulose retention while DES systems based on organic acids resulted in varying hemicellulose losses. The [Ch]Cl / urea mixture did not appear to be corrosive to stainless steel, which was another advantage of this DES system.

  • Experiences in implementing immunopsychiatry in real life

    • Immunological mechanisms, both alone and in combination, are associated with a broad range of psychiatric disorders encompassing autoimmune, autoinflammatory disorders but also genetic, metabolic, or other immunological disorders. Early treatment improves the outcome for autoimmune disorders, but early diagnosis is more difficult when isolated psychiatric symptoms are manifestations of autoimmunity. Treatment of these cases must encompass integrated models of disease, as both systemic autoimmunity and psychological processes influence mental health. Several challenges need to be overcome to efficiently merge psychiatric and somatic disease paradigms and medical care ranging from language and conceptual barriers to organizational barriers. Since 2015, the Immunopsychiatry team at Uppsala University has developed a collaborative multidisciplinary approach to improve and integrate care for patients with moderate to severe psychiatric disorders. Based on this experience, we have outlined the obstacles to be overcome in taking steps forward to achieve the long-term goal of understanding and early detection and identification of treatable immunological conditions within the psychiatric patient population; the described framework of evaluations and work-flow may serve as a model for other centers.

  • Cancer Patients’ Satisfaction with Doctors’ Care : Consequences and Contributing Conditions

    • The main aims were to: explore whether there is a relation between doctors’ ability to identify patients’ worry and wish for information and self-efficacy with regard to communicating with patients about difficult matters; describe which cues doctors consider when estimating patients’ worry and wish for information, and investigate whether there is a relation between patients’ satisfaction with doctors’ care and patients’ psychosocial function. Eleven doctors and 69 patients (of which 36 patients participated in the longitudinal study) with carcinoid tumours participated. Doctors’ self-efficacy, and ability to identify patients’ worry/wish for information were investigated at patients’ first admission. Doctors were interviewed about which cues they considered when estimating patients worry/wish for information. Patients’ satisfaction with care (CASC SF 4.0) and psychosocial function (EORTC QLQ-C30, HADS) were measured longitudinally, during the first year after diagnosis. Doctors reported higher self-efficacy when showing good ability to identify patients’ wish for information, than when showing less good ability, overestimated patients’ worry and underestimated patients’ wish for information. Doctors considered patients’ verbal behaviour and body language together with knowledge and experience when estimating patients worry and wish for information. Patients who met doctors showing good ability to identify their wish for information, reported a higher cognitive function than patients who met doctors showing less good ability. At all assessments patients expressed high satisfaction with doctor’ care and patients’ satisfaction did not change over time. Patients’ satisfaction with doctors’ care were related to their psychosocial function shortly after the first three admissions to specialist care. Patients with carcinoid tumours in some respects reported a worse HRQoL than the general Swedish population. Fatigue, diarrhoea, limited possibilities to work/pursue daily activities, and worry that the illness will get worse were among the most prevalent, and worst, aspects of disease- and treatment related distress.

  • A generic emission model to predict release of organic substances from materials in consumer goods

    • Organic chemicals may be released when consumer goods are used, contributing to environmental and human levels of potentially hazardous chemicals. A generic model was developed to predict emissions of organic chemicals from various materials in consumer products. The model involved three modules, which each predict a key parameter needed to calculate the mass of individual chemicals emitted. Partition coefficients between a material and the surrounding air were predicted using Abraham solvation parameters, diffusion coefficients in materials were calculated using the Piringer equation, and convective mass transfer coefficients were evaluated by applying the Chilton–Colburn analogy. The calculated emission rates from predicted parameters were evaluated and agreed well with literature data. The release of plasticizers from vinyl flooring used in Sweden was calculated to demonstrate the utility of the generic model. The estimated emitted masses of di(2-ethylhexyl)phthalate (DEHP), di-iso-nonylphthalate (DINP), and 1,2-cyclohexanedicarboxylic acid di-iso-nonyl ester (DINCH) in 2012 were 210 kg, 40 kg, and 3.6 kg respectively. Emissions from vinyl flooring were estimated for the period 1990 to 2035 and it was shown that the recent substitution of DEHP with DINP will help to reduce plasticizer emissions. Model calculations for alternative plasticizers revealed that DINCH would yield similar emissions to DINP, whereas use of diethyl hexyl-iso-sorbide or diethyl hexyl adipate would result in higher emissions.

  • Norrlands vattenanknutna kulturmiljöer – en förstudie : rapport från ett FoU-projekt

    • Syftet med föreliggande förstudie har varit att ta fram ett underlag inför ett kommande tvärvetenskapligt forsknings- och digitaliseringsprojekt. Bakgrunden är att Riksantikvarieämbetet under 1940-1980-talen genomförde omfattande arkeologiska, men även naturvetenskapliga och etnologiska, undersökningar vid Norrlands älvar och sjöar som ett led i vattenkraftsutbyggnaden. Materialet kom under 1960-1980-talen att bearbetas tillsammans med Stockholms universitet inom ramen för det tvärvetenskapliga projektet Norrlands Tidiga Bebyggelse. Insatserna resulterade i ett rikt material som idag förvaras i Riksantikvarieämbetets arkiv. Då stora delar av Norrland alltjämt är antikvariskt eftersatta utgör det ett mycket viktigt källmaterial för både forskning och förvaltning av dess kulturmiljöer.Det långsiktiga målet med förstudien och det kommande projektet är att skapa ett kunskapsunderlag för forskning och redskap för landskapsanalyser som leder till ett hållbart bevarande och förvaltning av Norrlands vattenanknutna kulturmiljöer. Inom förstudien har en rad viktiga forsknings- och förvaltningsområden identifierats. För att ta fram ett adekvat underlag inför ansökan har den bland annat mynnat ut i olika förslag på strategier för det kommande projektets genomförande och finansiering, en fördjupning av frågor som rör förvaltning av vattenanknutna kulturmiljöer, utvecklat tvärvetenskapliga forskningsfrågor, tagit fram en digitaliseringsstrategi, formaliserat ett nätverk och en referensgrupp, genomfört en behovsanalys samt en inventering för att identifiera relevant arkivmaterial inom projektdeltagarnas respektive institutioner. Med förstudien har vi således lagt grunden till ett tvärvetenskapligt forsknings- och digitaliseringsprojekt som inkluderar kulturmiljöförvaltning, arkeologi, etnologi, sociologi och naturveten-skapliga ämnen, däribland miljö- och klimatforskning.

  • A transplantable human medullary thyroid carcinoma as a model for RET tyrosine kinase-driven tumorigenesis

    • Hereditary medullary thyroid carcinoma (MTC) is caused by germline mutations in the RET proto-oncogene, resulting in constitutive activation of the RET tyrosine kinase. A substantial proportion of sporadic MTCs also have RET mutations, making the RET tyrosine kinase a potential therapeutic target in MTC. We have established a transplantable MTC in nude mice from a sporadic human MTC carrying a RET C634R mutation. Transplanted tumors had an exponential growth rate with an approximate doubling time of about 3 weeks, and expressed a neuroendocrine phenotype characteristic of MTC, e.g., expression of calcitonin, chromogranin A (CgA), synaptophysin, synaptic vesicle protein 2 (SV2), vesicular monoamine transporter-1 and -2, carcinoembryonic antigen, cytokeratin 8/18, epithelial cadherin, and neural cell adhesion molecule. Plasma calcitonin and CgA levels were elevated in tumor-bearing mice and correlated with tumor size. Cytogenetic analysis, including spectral karyotyping, confirmed the human origin of the xenografted tumors and demonstrated an abnormal, near triploid karyotype. Treatment of tumor-bearing nude mice with the tyrosine kinase inhibitor ZD6474, which specifically inhibits RET, epidermal growth factor receptor (EGFR), and vascular endothelium growth factor receptor (VEGFR) tyrosine kinases, resulted in a dose-dependent inhibition of tumor growth. Oral ZD6474 given once daily (250 mg/kg, 5 days/week) reduced tumor volume to 11% when compared with controls after 4 weeks. Our results show that this transplantable MTC, designated GOT2, represents a novel and useful model for studies of MTC and RET tyrosine kinase-dependent tumor growth.

  • Long-term ambient air pollution and coronary atherosclerosis : results from the Swedish SCAPIS study

    • Background and aims: Despite firm evidence for an association between long-term ambient air pollution exposure and cardiovascular morbidity and mortality, results from epidemiological studies on the association between air pollution exposure and atherosclerosis have not been consistent. We investigated associations between long-term low-level air pollution exposure and coronary atherosclerosis.Methods: We performed a cross-sectional analysis in the large Swedish CArdioPulmonary bioImaging Study (SCAPIS, n = 30 154), a random general population sample. Concentrations of total and locally emitted particulate matter <2.5 μm (PM2.5), <10 μm (PM10), and nitrogen oxides (NOx) at the residential address were modelled using high-resolution dispersion models. We estimated associations between air pollution exposures and segment involvement score (SIS), coronary artery calcification score (CACS), number of non-calcified plaques (NCP), and number of significant stenoses, using ordinal regression models extensively adjusted for potential confounders.Results: Median 10-year average PM2.5 exposure was 6.2 μg/m3 (range 3.5–13.4 μg/m3). 51 % of participants were women and 51 % were never-smokers. None of the assessed pollutants were associated with a higher SIS or CACS. Exposure to PM2.5 was associated with NCP (adjusted OR 1.34, 95 % CI 1.13, 1.58, per 2.05 μg/m3). Associations with significant stenoses were inconsistent.Conclusions: In this large, middle-aged general population sample with low exposure levels, air pollution was not associated with measures of total burden of coronary atherosclerosis. However, PM2.5 appeared to be associated with a higher prevalence of non-calcified plaques. The results suggest that increased risk of early-stage atherosclerosis or rupture, but not increased total atherosclerotic burden, may be a pathway for long-term air pollution effects on cardiovascular disease.
