  • Genome-wide association analysis identifies three new susceptibility loci for childhood body mass index.

    • A large number of genetic loci are associated with adult body mass index. However, the genetics of childhood body mass index are largely unknown. We performed a meta-analysis of genome-wide association studies of childhood body mass index, using sex- and age-adjusted standard deviation scores. We included 35 668 children from 20 studies in the discovery phase and 11 873 children from 13 studies in the replication phase. In total, 15 loci reached genome-wide significance (P-value < 5 × 10(-8)) in the joint discovery and replication analysis, of which 12 are previously identified loci in or close to ADCY3, GNPDA2, TMEM18, SEC16B, FAIM2, FTO, TFAP2B, TNNI3K, MC4R, GPR61, LMX1B and OLFM4 associated with adult body mass index or childhood obesity. We identified three novel loci: rs13253111 near ELP3, rs8092503 near RAB27B and rs13387838 near ADAM23. Per additional risk allele, body mass index increased 0.04 Standard Deviation Score (SDS) [Standard Error (SE) 0.007], 0.05 SDS (SE 0.008) and 0.14 SDS (SE 0.025), for rs13253111, rs8092503 and rs13387838, respectively. A genetic risk score combining all 15 SNPs showed that each additional average risk allele was associated with a 0.073 SDS (SE 0.011, P-value = 3.12 × 10(-10)) increase in childhood body mass index in a population of 1955 children. This risk score explained 2% of the variance in childhood body mass index. This study highlights the shared genetic background between childhood and adult body mass index and adds three novel loci. These loci likely represent age-related differences in strength of the associations with body mass index.

  • Testing the epicPCR workflow to reveal eukaryote-prokaryote interactions at single-cell resolution

    • Physical interactions between microeukaryotes and their prokaryotic counterparts abound in nature but are laborious to characterize due to their large number and microscopic dimension. Emulsion, paired-isolation and concatenation PCR (epicPCR) has the potential to uncover these interactions at a large scale while maintaining the resolution of individual cells. In this approach, single eukaryotic cells are trapped inside polyacrylamide beads together with their physically associated bacterial partners. This is followed by molecular barcoding of taxonomic marker genes of both parts in a compartmentalized manner. In this study, we report the first iteration in the evaluation of this workflow when adapted to recover eukaryote-prokaryote associations via SSU rRNA gene linkage. Predatory associations between two cultured mixotrophic flagellates and their co-cultured bacterial cohort were our target. Three sets of experiments were carried out, in which the model flagellates were 1) encapsulated on their own, 2) with a bacterial community assembled artificially or 3) with a wastewater sample. In all cases, the most frequent associations between either of the mixotrophs and bacterial constituents were those that involved the most abundant bacterial taxa in the samples. This was most evident in experiment from the second set, where the strongest associations corresponded to those between the eukaryotes and the most abundant members of the mock bacterial community. This result points to the loss of single cell resolution at some point during the protocol. We hypothesize that the erosion of the compartmentalization principle might arise from two sources. First, the extreme polydispersity in droplet size of the polymerizing emulsion, combined with much smaller cell size and much higher population density of the bacteria relative to the eukaryotes, can cause random co-encapsulation of cells that are not physically attached. Second, the design of the barcoding reaction as implemented here might be prone to generate a large number of non-barcoded fragments susceptible to be spuriously tagged during nested PCR, ruining the signature of individual separation. Although technical limitations exist, avenues for further development remain open. The vast exploratory potential of the epicPCR technique justifies further research to overcome these technical constrains.

    • Three short-alkyl-chain-modified [DBU][TFSI] ionic liquids (ILs) were synthesized and utilized in electrets. The electrets were prepared by mixing a UV-curable polymer with the ionic liquids followed by polymerization while applying an external electric field, thus forming spatially separated anions and cations in the proximity of opposing surfaces of the composite slabs. The immobilized surplus surface charge was measured by periodically engaging the electret with a metal counter electrode plate and detecting the displacement current using a charge amplifier. The results show that electrets based on polymerized [DBU][TFSI] ILs have a separated surface charge density of up to 64 nC × cm−2, which equals an energy harvesting density of 7.0 nJ × cm−2. Control measurements repeated after a few days to assess the stability and reproducibility of the systems showed that while charge separation reverses over time to some extent, the polymerized ionic liquid samples are resilient to exposure to atmospheric conditions and could be utilized in this type of energy harvesting scheme.

    • Genetic variants within nearly 1000 loci are known to contribute to modulation of blood lipid levels. However, the biological pathways underlying these associations are frequently unknown, limiting understanding of these findings and hindering downstream translational efforts such as drug target discovery.To expand our understanding of the underlying biological pathways and mechanisms controlling blood lipid levels, we leverage a large multi-ancestry meta-analysis (N=1,654,960) of blood lipids to prioritize putative causal genes for 2286 lipid associations using six gene prediction approaches. Using phenome-wide association (PheWAS) scans, we identify relationships of genetically predicted lipid levels to other diseases and conditions. We confirm known pleiotropic associations with cardiovascular phenotypes and determine novel associations, notably with cholelithiasis risk. We perform sex-stratified GWAS meta-analysis of lipid levels and show that 3-5% of autosomal lipid-associated loci demonstrate sex-biased effects. Finally, we report 21 novel lipid loci identified on the X chromosome. Many of the sex-biased autosomal and X chromosome lipid loci show pleiotropic associations with sex hormones, emphasizing the role of hormone regulation in lipid metabolism.Taken together, our findings provide insights into the biological mechanisms through which associated variants lead to altered lipid levels and potentially cardiovascular disease risk.

  • Common data elements and data management : Remedy to cure underpowered preclinical studies

    • Lack of translation of data obtained in preclinical trials to clinic has kindled researchers to develop new methodologies to increase the power and reproducibility of preclinical studies. One approach relates to harmonization of data collection and analysis, and has been used for a long time in clinical studies testing anti-seizure drugs. EPITARGET is a European Union FP7-funded research consortium composed of 18 partners from 9 countries. Its main research objective is to identify biomarkers and develop treatments for epileptogenesis. As the first step of harmonization of procedures between laboratories, EPITARGET established working groups for designing project-tailored common data elements (CDEs) and case report forms (CRFs) to be used in data collection and analysis. Eight major modules of CRFs were developed, presenting >1000 data points for each animal. EPITARGET presents the first single-project effort for harmonization of preclinical data collection and analysis in epilepsy research. EPITARGET is also anticipating the future challenges and requirements in a larger-scale preclinical harmonization of epilepsy studies, including training, data management expertise, cost, location, data safety and continuity of data repositories during and after funding period, and incentives motivating for the use of CDEs.

    • The duration of pregnancy is influenced by fetal and maternal genetic and non-genetic factors. Here we report a fetal genome-wide association meta-analysis of gestational duration, and early preterm, preterm, and postterm birth in 84,689 infants. One locus on chromosome 2q13 is associated with gestational duration; the association is replicated in 9,291 additional infants (combined P=3.96×10-14). Analysis of 15,588 mother-child pairs shows that the association is driven by fetal rather than maternal genotype. Functional experiments show that the lead SNP, rs7594852, alters the binding of the HIC1 transcriptional repressor. Genes at the locus include several interleukin 1 family members with roles in pro-inflammatory pathways that are central to the process of parturition. Further understanding of the underlying mechanisms will be of great public health importance, since giving birth either before or after the window of term gestation is associated with increased morbidity and mortality.

  • Adult congenital heart disease training in Europe: current status, disparities and potential solutions.

    • This study aimed to determine the status of training of adult congenital heart disease (ACHD) cardiologists in Europe.A questionnaire was sent to ACHD cardiologists from 34 European countries.Representatives from 31 of 34 countries (91%) responded. ACHD cardiology was recognised by the respective ministry of Health in two countries (7%) as a subspecialty. Two countries (7%) have formally recognised ACHD training programmes, 15 (48%) have informal (neither accredited nor certified) training and 14 (45%) have very limited or no programme. Twenty-five countries (81%) described training ACHD doctors 'on the job'. The median number of ACHD centres per country was 4 (range 0-28), median number of ACHD surgical centres was 3 (0-26) and the median number of ACHD training centres was 2 (range 0-28). An established exit examination in ACHD was conducted in only one country (3%) and formal certification provided by two countries (7%). ACHD cardiologist number versus gross domestic product Pearson correlation coefficient=0.789 (p<0.001).Formal or accredited training in ACHD is rare among European countries. Many countries have very limited or no training and resort to 'train people on the job'. Few countries provide either an exit examination or certification. Efforts to harmonise training and establish standards in exit examination and certification may improve training and consequently promote the alignment of high-quality patient care.

    • Limited data exist on training of European paediatric and adult congenital cardiologists.A structured and approved questionnaire was circulated to national delegates of Association for European Paediatric and Congenital Cardiology in 33 European countries.Delegates from 30 countries (91%) responded. Paediatric cardiology was not recognised as a distinct speciality by the respective ministry of Health in seven countries (23%). Twenty countries (67%) have formally accredited paediatric cardiology training programmes, seven (23%) have substantial informal (not accredited or certified) training, and three (10%) have very limited or no programme. Twenty-two countries have a curriculum. Twelve countries have a national training director. There was one paediatric cardiology centre per 2.66 million population (range 0.87-9.64 million), one cardiac surgical centre per 4.73 million population (range 1.63-10.72 million), and one training centre per 4.29 million population (range 1.63-10.72 million population). The median number of paediatric cardiology fellows per training programme was 4 (range 1-17), and duration of training was 3 years (range 2-5 years). An exit examination in paediatric cardiology was conducted in 16 countries (53%) and certification provided by 20 countries (67%). Paediatric cardiologist number is affected by gross domestic product (R2 = 0.41).Training varies markedly across European countries. Although formal fellowship programmes exist in many countries, several countries have informal training or no training. Only a minority of countries provide both exit examination and certification. Harmonisation of training and standardisation of exit examination and certification could reduce variation in training thereby promoting high-quality care by European congenital cardiologists.

    • In the present work electrically conductive, flexible, lightweight carbon sponge materials derived from open-pore structure melamine foams are studied and explored. Hydrophobic and hydrophilic surface properties - depending on the chosen treatment conditions - allow the separation and storage of liquid chemical compounds. Activation of the carbonaceous structures substantially increases the specific surface area from ,4 m2 g21 to ,345 m2 g21 , while retaining the original three-dimensional, open-pore structure suitable for hosting, for example, Ni catalyst nanoparticles. In turn the structure is rendered suitable for hydrogenating acetone to 2-propanol and methyl isobutyl ketone as well for growing hierarchical carbon nanotube structures used as electric double-layer capacitor electrodes with specific capacitance of ,40 F/g. Mechanical stress-strain analysis indicates the materials are super-compressible (.70% volume reduction) and viscoelastic with excellent damping behavior (loss of 0.69 6 0.07), while piezoresistive measurements show very high gauge factors (from ,20 to 50) over a large range of deformations. The cost-effective, robust and scalable synthesis - in conjunction with their fascinating multifunctional utility - makes the demonstrated carbon foams remarkable competitors with other three-dimensional carbon materials typically based on pyrolyzed biopolymers or on covalently bonded graphene and carbon nanotube frameworks.

    • Polylactic acid (PLA) is a biopolymer that has potential for use in food packaging applications; however, its low crystallinity and poor gas barrier properties limit its use. This study aimed to increase the understanding of the structure property relation of biopolymer blends and their nanocomposites. The crystallinity of the final materials and their effect on barrier properties was studied. Two strategies were performed: first, different concentrations of poly(hydroxybutyrate) (PHB; 10, 25, and 50 wt %) were compounded with PLA to facilitate the PHB spherulite development, and then, for further increase of the overall crystallinity, glycerol triacetate (GTA) functionalized chitin nano crystals (ChNCs) were added. The PLA:PHB blend with 25 wt % PHB showed the formation of many very small PHB spherulites with the highest PHB crystallinity among the examined compositions and was selected as the matrix for the ChNC nanocomposites. Then, ChNCs with different concentrations (0.5, 1, and 2 wt %) were added to the 75:25 PLA:PHB blend using the liquid-assisted extrusion process in the presence of GTA. The addition of the ChNCs resulted in an improvement in the crystallization rate and degree of PHB crystallinity as well as mechanical properties. The nanocomposite with the highest crystallinity resulted in greatly decreased oxygen (O) and carbon dioxide (CO2) permeability and increased the overall mechanical properties compared to the blend with GTA. This study shows that the addition ChNCs in PLA:PHB can be a possible way to reach suitable gas barrier properties for food packaging films.
