    • The maximum limits of the Eurasian ice sheets during four glaciations have been reconstructed: (1) the Late Saalian (> 140 ka), (2) the Early Weichselian (100-80 ka), (3) the Middle Weichselian (60-50 ka) and (4) the Late Weichselian (25-15 ka). The reconstructed ice limits are based on satellite data and aerial photographs combined with geological field investigations in Russia and Siberia, and with marine seismic- and sediment core data. The Barents-Kara Ice Sheet got progressively smaller during each glaciation, whereas the dimensions of the Scandinavian Ice Sheet increased. During the last Ice Age the Barents-Kara Ice Sheet attained its maximum size as early as 90-80,000 years ago when the ice front reached far onto the continent. A regrowth of the ice sheets occurred during the early Middle Weichselian, culminating about 60-50,000 years ago. During the Late Weichselian the Barents-Kara Ice Sheet did not reach the mainland east of the Kanin Peninsula, with the exception of the NW fringe of Taimyr. A numerical ice-sheet model, forced by global sea level and solar changes, was run through the full Weichselian glacial cycle. The modeling results are roughly compatible with the geological record of ice growth, but the model underpredicts the glaciations in the Eurasian Arctic during the Early and Middle Weichselian. One reason for this is that the climate in the Eurasian Arctic was not as dry then as during the Late Weichselian glacial maximum.

    • Mercury stable isotopic compositions were determined for marine sediments from eight locations in the Arctic Ocean Basin. Mass dependent fractionation (MDF) and mass independent fractionation (MIF) of Hg stable isotopes were recorded across a variety of depositional environments, water depths, and stratigraphic ages. delta(202) Hg (MDF) ranges from -2.34% to -0.78%; Delta(199) Hg (MIF) from -0.18% to +0.12%; and Delta(201) Hg (MIF) from -0.29% to + 0.05% for the complete data set (n = 33). Holocene sediments from the Chukchi Sea and Morris Jesup Rise record the most negative Delta(199) Hg values, while Pleistocene sediments from the Central Arctic Ocean record the most positive Delta(199) Hg values. The most negative delta(202) Hg values are recorded in Pleistocene sediments. Eocene sediments (Lomonosov Ridge) show some overlap in their Hg isotopic compositions with Quaternary sediments, with a sample of the Arctic Ocean PETM (56 Ma) most closely matching the average Hg isotopic composition of Holocene Arctic marine sediments. Collectively, these data support a terrestrially-dominated Hg source input for Arctic Ocean sediment through time, although other sources, as well as influences of sea ice, atmospheric mercury depletion events (AMDEs), and anthropogenic Hg (in core top samples) on Hg isotopic signatures must also be considered.

  • Lack of HIN-1 methylation in BRCAl-linked and "BRCA1-like" breast tumors

    • We recently identified a candidate tumor suppressor gene, HIN-1, that is silenced due to methylation in the majority of sporadic breast carcinomas and is localized to 5q33-qter, an area frequently lost in BRCA1 tumors and thought to harbor a BRCA1 modifier gene. To establish whether germ-line mutations in HIN-1 may influence breast cancer risk, we sequenced the HIN-1 coding region in 10 familial breast cancer patients with positive logarithm of the odds scores of at least one of the markers flanking HIN-1. We also sequenced the HIN-1 coding region in 15 BRCA1 and 35 sporadic breast tumors to determine whether HIN-1 is the target of the frequent 5q loss in BRCA1 tumors. No sequence alterations were found in any of the cases analyzed. However, analysis of HIN-1 promoter methylation status revealed that in striking contrast to sporadic cases, there is a nearly complete lack of HIN-1 methylation in BRCA1 tumors (P < 0.0001). Sporadic breast tumors with a "BRCA1-like" histopathological phenotype also demonstrated significantly lower frequency of HIN-1 promoter methylation (P = 0.01) compared with other cancer types, and there was also a difference among tumors based on their estrogen receptor and HER2 status (P = 0.006), suggesting that HIN-1 methylation patterns are associated with specific breast cancer subtypes.

    • Sediment core HLY0503-8JPC raised by the HOTRAX'05 expedition from the Mendeleev Ridge was analyzed for multiple lithological, paleontological. and stable-isotopic proxies to reconstruct paleoceanographic conditions in the western Arctic Ocean during the Late Quaternary. The core, extensively sampled in the upper 5 m, reveals pronounced changes in sedimentary environments during the ca. 250 kyr interval encompassing Marine Isotopic Stages (MIS) 1 to 7. An estimated average resolution of 500 yr/sample, at least for the last glacial cycle including the last interglacial, provides more detail than seen in other sedimentary records from the western Arctic Ocean. The age control is provided by C-14 and amino acid racemization measurements on planktonic foraminifers and correlations with the stratigraphy developed for the central Lomonosov Ridge and with glacial events at the Eurasian Arctic margin. Cyclic variations in lithology combined with foraminiferal abundance and stable-isotopic composition indicate profound changes in hydrographic and depositional environments between interglacial-type and glacial-type periods apparently reflecting a combination of 100-kyr and precessional time scales. This periodicity is complicated by abrupt iceberg- and/or meltwater-discharge events with variable (Laurentide vs. Eurasian) provenance. The proxy record from the interval identified as the last interglacial (MIS 5e), which may aid in understanding the future state of the Arctic Ocean, indicates low ice conditions and possibly enhanced stratification of the water column.

    • Seafloor mapping of the central Lomonosov Ridge using a multibeam echo-sounder during the Beringia/Healy–Oden Trans-Arctic Expedition (HOTRAX) 2005 shows that a channel across the ridge has a substantially shallower sill depth than the 2500 m indicated in present bathymetric maps. The multibeam survey along the ridge crest shows a maximum sill depth of about 1870 m. A previously hypothesized exchange of deep water from the Amundsen Basin to the Makarov Basin in this area is not confirmed. On the contrary, evidence of a deep-water flow from the Makarov to the Amundsen Basin was observed, indicating the existence of a new pathway for Canadian Basin Deep Water toward the Atlantic Ocean. Sediment data show extensive current activity along the ridge crest and along the rim of a local Intra Basin within the ridge structure.

    • A study of three long cores from the outer shelf and continental slope north of Alaska in the Arctic Ocean indicate that localized drift deposits occur here with sedimentation rates of more than 1.5 m/kyr during the Holocene. Currents in this area average about 5–20 cm/s but can reach 100 cm/s and these velocities transport the sediment found in these cores primarily as intermittent suspended load. These high accumulation sediments form levee-like deposits associated with margins of canyons cutting across the shelf and slope. Unlike most textural investigations of Arctic sediment that focus on the coarser ice-rafted detritus (IRD), this paper focuses on the > 95% of the sediment, which is finer than 45 μm. The mean size of this fraction varies between 6 and 15 μm in Holocene sediments from the Chukchi–Alaskan shelf and slope with the higher values closer to shore. Analysis of detailed size distributions of these Holocene deposits are compared to 34 sediment samples collected from sea ice across the Arctic Ocean and to Holocene sediment from central Arctic Ocean cores and indicate that similar textural parameters occur in all of these sediments. Principal components of these size distributions indicate that sea ice is an important link between the shelves and the central Arctic. Factor scores indicate nearly identical components in the clay and fine silt size fractions but very different components in the coarse silt for sea ice sediment and central Arctic ridge sediments compared to shelf and continental slope deposits. Sea ice must contribute to sedimentation in both of these Arctic regions, but bottom currents dominate in the slope region, forming drift deposits.
