    • The 2016 European Vasculitis Society (EUVAS) meeting, held in Leiden, the Netherlands, was centered around phenotypic subtyping in antineutrophil cytoplasmic antibody (ANCA)–associated vasculitis (AAV). There were parallel meetings of the EUVAS petals, which here report on disease assessment; database; and long-term follow-up, registries, genetics, histology, biomarker studies, and clinical trials. Studies currently conducted will improve our ability to discriminate between different forms of vasculitis. In a project that involves the 10-year follow-up of AAV patients, we are working on retrieving data on patient and renal survival, relapse rate, the cumulative incidence of malignancies, and comorbidities. Across Europe, several vasculitis registries were developed covering over 10,000 registered patients. In the near future, these registries will facilitate clinical research in AAV on a scale hitherto unknown. Current studies on the genetic background of AAV will explore the potential prognostic significance of genetic markers and further refine genetic associations with distinct disease subsets. The histopathological classification of ANCA-associated glomerulonephritis is currently evaluated in light of data coming out of a large international validation study. In our continuous search for biomarkers to predict clinical outcome, promising new markers are important subjects of current research. Over the last 2 decades, a host of clinical trials have provided evidence for refinement of therapeutic regimens. We give an overview of clinical trials currently under development, and consider refractory vasculitis in detail. The goal of EUVAS is to stimulate ongoing research in clinical, serological, and histological management and techniques for patients with systemic vasculitis, with an outlook on the applicability for clinical trials.

  • Overview of European Audience Research. Research report from the COST Action IS0906 Transforming Audiences, Transforming Societies

    • The state of audience research in European countries is not yet properly charted due to the fragmented and varied nature of the field. Obtaining an overview was both an objective of the Action in its own right, and a precondition of other achievements. For the first meeting of the Working Groups (WG) in Lisbon (November 11-13, 2010), the Steering Group (SG) of the Action invited the members to submit short national essays that review existing and emerging audience research in their respective countries. The reviews could be theoretical, topical, methodological, empirical or institutional. Most of them were presented in the WG sessions of the Lisbon meeting. The SG requested some revisions or additional essays in order to better cover the countries for which the essays where too focused, or to cover the countries that were not represented at all. In total, 58 essays covering the 30 participating countries were delivered and are available on the Action website at: http://www.cost-transforming-audiences.eu. This research report brings together these 59 essays as well as a tentative synthesis in the concluding section.

    • Background: Since the publication of the European League Against Rheumatism (EULAR) recommendations for the management of large vessel vasculitis (LVV) in 2009, several relevant randomised clinical trials and cohort analyses have been published, which have the potential to change clinical care and therefore supporting the need to update the original recommendations. Methods: Using EULAR standardised operating procedures for EULAR-endorsed recommendations, the EULAR task force undertook a systematic literature review and sought opinion from 20 experts from 13 countries. We modified existing recommendations and created new recommendations. Results: Three overarching principles and 10 recommendations were formulated. We recommend that a suspected diagnosis of LVV should be confirmed by imaging or histology. High dose glucocorticoid therapy (40-60 mg/day prednisone-equivalent) should be initiated immediately for induction of remission in active giant cell arteritis (GCA) or Takayasu arteritis (TAK). We recommend adjunctive therapy in selected patients with GCA (refractory or relapsing disease, presence of an increased risk for glucocorticoid-related adverse events or complications) using tocilizumab. Methotrexate may be used as an alternative. Non-biological glucocorticoid-sparing agents should be given in combination with glucocorticoids in all patients with TAK and biological agents may be used in refractory or relapsing patients. We no longer recommend the routine use of antiplatelet or anticoagulant therapy for treatment of LVV unless it is indicated for other reasons. Conclusions: We have updated the recommendations for the management of LVV to facilitate the translation of current scientific evidence and expert opinion into better management and improved outcome of patients in clinical practice.

  • Generational Use of News Media in Estonia : Media Access, Spatial Orientations and Discursive Characteristics of the News Media

    • Contemporary media research highlights the importance of empirically analysing the relationships between media and age, changing user patterns over the life course, and generational experiences within media discourse beyond the widely hyped buzz terms such as the ‘digital natives’, ‘Google generation’, and other digitally and technologically capable generation groups. This doctoral thesis seeks to define the ‘repertoires’ of news media that different generations use to obtain topical information and create their ‘media space’. It contributes to the development of a framework within which to analyse generational features in news audiences by putting the main focus on the cultural view of generations. This perspective was first introduced by Karl Mannheim in 1928. Departing from his legacy, generations can be better conceived as social formations that are built on self-identification, rather than equally distributed cohorts. With the purpose of discussing the emergence of various ‘audiencing’ patterns from the perspectives of age, life course and generational identity, the thesis presents Estonia – a post-Soviet Baltic state – as an empirical example of a transforming society with a dynamic media landscape which is witnessing the expanding impact of new media and a shift to digitisation.The thesis is based on data from two nationally representative cross-section surveys on media use and media attitudes (conducted during the 2002-2012 period) and focus group discussions, that are used to map similarities and differences among five generation cohorts born between 1932 and 1997 with regard to the access and use of the established news media, thematic preferences and spatial orientations of media use, and discursive approach to news formats.The findings demonstrate remarkable differences between the cohorts, suggesting that they could be merged into three main groups that represent the prevailing types of relations with the news media. Yet, the study also reveals that attitudes and behaviour (including media behaviour), are not necessarily divided by year of birth, but are more and more dispersed along individualised interests and preferences.

  • Identification of LINE retrotransposons and long non-coding RNAs expressed in the octopus brain

    • Background Transposable elements (TEs) widely contribute to the evolution of genomes allowing genomic innovations, generating germinal and somatic heterogeneity, and giving birth to long non-coding RNAs (lncRNAs). These features have been associated to the evolution, functioning, and complexity of the nervous system at such a level that somatic retrotransposition of long interspersed element (LINE) L1 has been proposed to be associated to human cognition. Among invertebrates, octopuses are fascinating animals whose nervous system reaches a high level of complexity achieving sophisticated cognitive abilities. The sequencing of the genome of the Octopus bimaculoides revealed a striking expansion of TEs which were proposed to have contributed to the evolution of its complex nervous system. We recently found a similar expansion also in the genome of Octopus vulgaris. However, a specific search for the existence and the transcription of full-length transpositionally competent TEs has not been performed in this genus. Results Here, we report the identification of LINE elements competent for retrotransposition in Octopus vulgaris and Octopus bimaculoides and show evidence suggesting that they might be transcribed and determine germline and somatic polymorphisms especially in the brain. Transcription and translation measured for one of these elements resulted in specific signals in neurons belonging to areas associated with behavioral plasticity. We also report the transcription of thousands of lncRNAs and the pervasive inclusion of TE fragments in the transcriptomes of both Octopus species, further testifying the crucial activity of TEs in the evolution of the octopus genomes. Conclusions The neural transcriptome of the octopus shows the transcription of thousands of putative lncRNAs and of a full-length LINE element belonging to the RTE class. We speculate that a convergent evolutionary process involving retrotransposons activity in the brain has been important for the evolution of sophisticated cognitive abilities in this genus.
