    • The presence of elevated levels of naturally occurring arsenic in groundwater of Bangladesh, has severely impaired the decade long effort of providing safe water to nearly 98% of its population and putting an estimated 3 5 million people-nearly one fourth of the total population at risk. In order to address this problem, a project titled "Arsenic in tubewell (TW) water and health consequences in Matlab Upazila of Chandpur district (AsMat)" is being implemented jointly by ICDDR,B and BRAC. During this study. all the TWs in Matlab have been assigned unique identification numbers, with marked GPS coordinates, depth, and age. It is estimated that nearly 65% of the about 13,000 TWs in Matlab have As concentrations above the Bangladesh drinking water standard (50 mu g/L). In order to minimize arsenic exposure, a work to provide various alternate safe drinking water options to the exposed population has been initiated. As of March 2004, about 1047 different alternate safe water options, such as Pond Sand Filter (PSF), Rainwater Harvester (RWH) and different filters to remove arsenic as well as pathogenic bacteria, were distributed among the targeted exposed population in Matlab. To ensure sustainable use, the provided options were assessed based on community acceptability, technical viability, and financial viability.

  • Higher risk of hyperglycemia with greater susceptibility in females in chronic arsenic-exposed individuals in Bangladesh

    • Arsenic (As) toxicity and diabetes mellitus (DM) are emerging public health concerns worldwide. Although exposure to high levels of As has been associated with DM, whether there is also an association between low and moderate As exposure and DM remains unclear. We explored the dose-dependent association between As exposure levels and hyperglycemia, with special consideration of the impact of demographic variables, in 641 subjects from rural Bangladesh. The total study participants were divided into three groups depending on their levels of exposure to As in drinking water (low, moderate and high exposure groups). Prevalence of hyperglycemia, including impaired glucose tolerance (IGT) and DM was significantly associated with the subjects’ drinking water arsenic levels. Almost all exposure metrics (As levels in the subjects’ drinking water, hair and nails) showed dose-dependent associations with the risk of hyperglycemia, IGT and DM. Among the variables considered, sex, age, and BMI were found to be associated with higher risk of hyperglycemia, IGT and DM. In sex-stratified analyses, As exposure showed a clearer pattern of dose-dependent risk for hyperglycemia in females than males. Finally, drinking water containing low-to-moderate levels of As (50.01–150 μg/L) was found to confer a greater risk of hyperglycemia than safe drinking water (As ≤10 μg/L). Thus the results suggested that As exposure was dose-dependently associated with hyperglycemia, especially in females. © 2019 Elsevier B.V.

  • Complex Involvement of Interleukin-26 in Bacterial Lung Infection

    • Pneumonia is a global cause of mortality, and this provides a strong incentive to improve the mechanistic understanding of innate immune responses in the lungs. Here, we characterized the involvement of the cytokine interleukin (IL)-26 in bacterial lung infection. We observed markedly increased concentrations of IL-26 in lower airway samples from patients with bacterial pneumonia and these correlated with blood neutrophil concentrations. Moreover, pathogen-associated molecular patterns (PAMPs) from both Gram-negative and -positive bacteria increased extracellular IL-26 concentrations in conditioned media from human models of alveolar epithelial cells, macrophages, and neutrophils in vitro. Stimulation with IL-26 inhibited the inherent release of neutrophil elastase and myeloperoxidase in unexposed neutrophils. This stimulation also inhibited the expression of activity makers in neutrophils exposed to Klebsiella pneumoniae. In addition, priming of human lung tissue ex vivo with exogenous IL-26 potentiated the endotoxin-induced increase in mRNA for other cytokines involved in the innate immune response, including the master Th17-regulator IL-23 and the archetype inhibitory cytokine IL-10. Finally, neutralization of endogenous IL-26 clearly increased the growth of Klebsiella pneumoniae in the macrophage culture. These findings suggest that IL-26 is involved in bacterial lung infection in a complex manner, by modulating critical aspects of innate immune responses locally and systemically in a seemingly purposeful manner and by contributing to the killing of bacteria in a way that resembles an antimicrobial peptide. Thus, IL-26 displays both diagnostic and therapeutic potential in pneumonia and deserves to be further evaluated in these respects.

    • The levee effect refers to the paradox that the construction of a levee to protect from flooding might induce property owners to invest more in their property, increasing the potential damages should the levee breach. Thus, paradoxically, the levee might increase flood risk. The levee effect was observed for high-income countries. We analyze whether it can also be observed in a low-income country, Bangladesh. In the Jamuna floodplain different levels of flood protection have existed alongside each other since the 1960s, so their effects can be compared.

    • Potentially toxic elements (PTEs) contamination in Buriganga river water, provoked by the point and non-point sources, has recently become a major health concern. Concentration, pollution degree, drinking and irrigation water potentiality, and associated health risks are the factors considered for this study. This study ascertained a mean pH, EC, TDS, and salinity value of 6.96, 86.25 mu S/cm, 51.86 mg/L, and 86.56%, respectively, revealing that it is safe for drinking and agricultural purposes. Furthermore, the mean value of major anion (NO-23: 1.16, SO-24 : 13.40, PO-34 : 0.66, Cl-1 : 14.71 mg/L) and major cations (Na+1 : 16.95, K+1 : 4.26, Ca+2 : 17.63, and Mg+2 : 5.47 mg/L) also estimated safe for human consumption and agricultural use. Trace element evaluation index (TEI) value was found to vary among the sites and ranging from 4.40 to 10.50, indicating moderate pollution and risk imposed by the single PTEs can be ranked as Mn>Fe>Pb>As>Cu>Zn. Nemerow index and comprehensive pollution index value stipulated an insignificant to moderate pollution by the studied elements. Consequently, no ecological risk was posed via elements studied in the proposed sites. The emerging sources for PTEs are confined to both anthropogenic and lithogenic derivations. Non-carcinogenic risk assessment stipulated that the river water was safe for adults and children as the estimated Hazard Quotient (HQ) and Hazard Index (HI) value was found below the threshold value (HQ/HI<1). Total carcinogenic risk was calculated at 7.44E-08 and 1.60E-08 for adults and children, respectively, specifying no risk, and water from the sites was evaluated as suitable for agricultural irrigation purposes on the basis of estimated indices.

    • Danio annulosus, new species, is described from a small pool below the Shuvolong Falls in the Kaptai Lake system in Bangladesh. It shares with chain danios (D. assamila, D. dangila, D. catenatus, D. concatenatus, and D. sysphigmatus) a colour pattern consisting of series of dark rings with light interspaces along the side, complete lateral line, 14 cir-cumpeduncular scales, a produced first ray in the pectoral fin, and a black humeral spot. It differs from other chain danios in possessing much shorter pectoral and pelvic fins, and a humeral spot that is slightly wider than deep instead of round or deeper than wide. The mitochondrial cytochrome c oxidase subunit I (COI) sequence separates D. annulosus from the most similar species, D. catenatus by a p-distance of 3.4%. Although recorded from only a single locality, Danio annulosus is expected to have a wider distribution in the Karnafuli River drainage

    • Four species of Devario are recorded from Bangladesh: D. aequipinnatus, D. anomalus, D. coxi, new species, and D. devario. Devario aequipinnatus has a wide distribution in northern India and Bangladesh. Devario coxi, from southeastern Bangladesh near Cox’s Bazar, differs from D. aequipinnatus in mtDNA (COI, p-distance 1.8%), colouration, proportional measurements, and meristics. The minor morphological differences and low frequency of overlapping meristics suggest relatively recent separation of D. coxi from other D. aequipinnatus. Devario anomalus occurs only in southeastern Bangladesh and is here reported from localities in addition to the type locality. It differs from the similar D. xyrops in adjacent Myanmar by slender body shape and by 2.3% p-distance in the COI gene. Specimens of D. anomalus from the Sangu River were found to have the mitochondrial genome of D. aequipinnatus from Bangladesh, but agree with other D. anomalus in the nuclear RAG1 gene. Devario devario has a wide distribution on the Indian Peninsula and border regions; in Bangladesh it is restricted in distribution to the Ganga, Brahmaputra, and Meghna drainages. Reports of D. assamensis and D. malabaricus from Bangladesh are misidentifications. Perilampus ostreographus M’Clelland, 1839, is tentatively synonymized with D. aequipinnatus. Phylogenetic analysis of 14 species of striped devarios based on the COI gene results in a polytomy with four unresolved clades. Devario deruptotalea from the Chindwin basin is the sister group of D. aequipinnatus+D. coxi. Devario devario is the sistergroup of D. xyrops+D. anomalus.

    • Laubuka tenella is a new species characterized by the colour pattern, consisting of short dark verticalbars anteriorly on the side, and a dark lateral band posteriorly on the side, combined with a relativelyshort pelvic fin and 29–30 lateral-line scales. It is separated from other   analysed by minimum9 % uncorrected p-distance in the mitochondrial COI gene. The type series is composed of specimens from small streams in the Cox’s Bazar District in Bangladesh (the type locality), and the Thandwe River drainage in western Myanmar. Laubuka brahmaputraensis is strongly indicated to be a junior synonymof L. laubuca, the second known species of Laubuka in Bangladesh. Eustira ceylonensis, currently in thes ynonymy of Devario malabaricus, is a valid species of Laubuka.

    • Osteobrama cotio is considered to be a widespread species in India and Bangladesh. Mitochondrial DNA (COI, 16S rRNA)shows that populations from the Meghna River, Karnafuli and Sangu Rivers, Narmada River, and Godavari River aregenetically distinct from each other. No morphological differences were found to separate Meghna and Karnafuli+Sangu populations, however. A putative new species, “Osteobrama serrata” has been described from the Barak River basin, statedto be distinguished from O. cotio by the presence of a serrated third dorsal-fin ray. The description of “O. serrata” doesnot fulfil requirements of the International Code of Zoological Nomenclature, (International Commission on ZoologicalNomenclature 1999) and the name is thus unavailable. Published DNA sequences of “Osteobrama serrata” are identicalto sequences of O. cotio from Bangladesh. As mentioned already in the original description, O. cotio has a serrated third dorsal-fin ray.
