    • Amphiphilic polymeric nanocarriers have attained immense attention for transporting drugs and other bioactive species to the living systems owing to their high loading capacity and efficient internalization. To avoid the side-effects of undesired interactions within the biological system, the use of biocompatible building blocks is most crucial in preparing polymeric amphiphiles. The excellent biocompatibility and multivalency of both glycerol and triglycerol make them suitable monomers to construct macromolecular skeletons. Besides these, easy availability and remarkable aqueous solubility further enlarge their use in synthetic chemistry and give ample possibilities for creating fascinating entities for practical applications. The conversion of glycerol into azido-glycerol and azido-triglycerol further helped in differential functionalization of their polymers with various hydrophobic and hydrophilic groups in different ratios thereby assisting in tuning the amphiphilicity of resulting functionalized polymers. Herein, we review the enzymatic synthesis of glycerol, azido-glycerol and azido-triglycerol based amphiphilic polymeric architectures as nanocarriers for various bio-active species. Enzymatically synthesized linear copolymers synthesized by selective esterification of primary hydroxyl groups of glycerol and its derivatives with suitable diacids/diesters are explored in this review.

    • Insecticide resistance jeopardizes the prevention of infectious diseases such as malaria and dengue fever by vector control of disease-transmitting mosquitoes. Effective new insecticidal compounds with minimal adverse effects on humans and the environment are therefore urgently needed. Here, we explore noncovalent inhibitors of the well-validated insecticidal target acetylcholinesterase (AChE) based on a 4-thiazolidinone scaffold. The 4-thiazolidinones inhibit AChE1 from the mosquitoes Anopheles gambiae and Aedes aegypti at low micromolar concentrations. Their selectivity depends primarily on the substitution pattern of the phenyl ring; halogen substituents have complex effects. The compounds also feature a pendant aliphatic amine that was important for activity; little variation of this group is tolerated. Molecular docking studies suggested that the tight selectivity profiles of these compounds are due to competition between two binding sites. Three 4-thiazolidinones tested for in vivo insecticidal activity had similar effects on disease-transmitting mosquitoes despite a 10-fold difference in their in vitro activity.

  • PKS 1830-211: OH and HI at z = 0.89 and the first MeerKAT UHF spectrum

    • The Large Survey Project (LSP) "MeerKAT Absorption Line Survey"(MALS) is a blind H I 21 cm and OH 18 cm absorption line survey in the L- and UHF-bands, primarily designed to better determine the occurrence of atomic and molecular gas in the circumgalactic and intergalactic medium, and its redshift evolution. Here we present the first results using the UHF band obtained towards the strongly lensed radio source PKS 1830-211, revealing the detection of absorption produced by the lensing galaxy. With merely 90 min of data acquired on-source for science verification and processed using the Automated Radio Telescope Imaging Pipeline (ARTIP), we detect in absorption the known H I 21 cm and OH 18 cm main lines at z = 0.89 at an unprecedented signal-to-noise ratio (4000 in the continuum, in each 6 km s-1 wide channel). For the first time we report the detection of OH satellite lines at z = 0.89, which until now have not been detected at z > 0.25. We decompose the OH lines into a thermal and a stimulated contribution, where the 1612 and 1720 MHz lines are conjugate. The total OH 1720 MHz emission line luminosity is 6100 L⊙. This is the most luminous known 1720 MHz maser line and is also among the most luminous of the OH main line megamasers. The absorption components of the different images of the background source sample different light paths in the lensing galaxy, and their weights in the total absorption spectrum are expected to vary in time on daily and monthly time scales. We compare our normalized spectra with those obtained more than 20 years ago, and find no variation. We interpret the absorption spectra with the help of a lens galaxy model derived from an N-body hydrodynamical simulation, with a morphology similar to its optical HST image. The resulting absorption lines depend mainly on the background continuum and the radial distribution of the gas surface density for each atomic and molecular species. We show that it is possible to reproduce the observations assuming a realistic spiral galaxy disk without invoking any central gas outflows. However, there are distinct and faint high-velocity features in the ALMA millimeter absorption spectra that most likely originate from high-velocity clouds or tidal features. These clouds may contribute to broaden the H I and OH spectra.

  • Examining the generalizability of research findings from archival data

    • This initiative examined systematically the extent to which a large set of archival research findings generalizes across contexts. We repeated the key analyses for 29 original strategic management effects in the same context (direct reproduction) as well as in 52 novel time periods and geographies; 45% of the reproductions returned results matching the original reports together with 55% of tests in different spans of years and 40% of tests in novel geographies. Some original findings were associated with multiple new tests. Reproducibility was the best predictor of generalizability-for the findings that proved directly reproducible, 84% emerged in other available time periods and 57% emerged in other geographies. Overall, only limited empirical evidence emerged for context sensitivity. In a forecasting survey, independent scientists were able to anticipate which effects would find support in tests in new samples.

  • PHIP - a novel candidate breast cancer susceptibility locus on 6q14.1

    • Most non-BRCA1/2 breast cancer families have no identified genetic cause. We used linkage and haplotype analyses in familial and sporadic breast cancer cases to identify a susceptibility locus on chromosome 6q. Two independent genome-wide linkage analysis studies suggested a 3 Mb locus on chromosome 6q and two unrelated Swedish families with a LOD > 2 together seemed to share a haplotype in 6q14.1. We hypothesized that this region harbored a rare high-risk founder allele contributing to breast cancer in these two families. Sequencing of DNA and RNA from the two families did not detect any pathogenic mutations. Finally, 29 SNPs in the region were analyzed in 44,214 cases and 43,532 controls from BCAC, and the original haplotypes in the two families were suggested as low-risk alleles for European and Swedish women specifically. There was also some support for one additional independent moderate-risk allele in Swedish familial samples. The results were consistent with our previous findings in familial breast cancer and supported a breast cancer susceptibility locus at 6q14.1 around the PHIP gene.

    • We are just starting to understand the physical processes driving the dramatic change in cosmic star formation rate between z ∼ 2 and the present day. A quantity directly linked to star formation is the molecular gas density, which should be measured through independent methods to explore variations due to cosmic variance and systematic uncertainties. We use intervening CO absorption lines in the spectra of mm-bright background sources to provide a census of the molecular gas mass density of the Universe. The data used in this work are taken from ALMACAL, a wide and deep survey utilizing the ALMA calibrator archive. While we report multiple Galactic absorption lines and one intrinsic absorber, no extragalactic intervening molecular absorbers are detected. However, due to the large redshift path surveyed (z = 182), we provide constraints on the molecular column density distribution function beyond z ∼ 0. In addition, we probe column densities of N(H2) > 1016 atoms cm−2, 5 orders of magnitude lower than in previous studies. We use the cosmological hydrodynamical simulation IllustrisTNG to show that our upper limits of ρ(H2) 108.3 M Mpc−3 at 0 < z ≤ 1.7 already provide new constraints on current theoretical predictions of the cold molecular phase of the gas. These results are in agreement with recent CO emission-line surveys and are complementary to those studies. The combined constraints indicate that the present decrease of the cosmic star formation rate history is consistent with an increasing depletion of molecular gas in galaxies compared to z ∼ 2.

  • Whole-genome Linkage Analysis and Sequence Analysis of Candidate Loci in Familial Breast Cancer

    • Background: Known breast cancer-predisposing genes account for fewer than 25% of all familial breast cancer cases and further studies are required to find the remaining high-and moderate-risk genes. We set-out to couple linkage analysis using microsatellite marker data and sequence analysis of linked regions in 13 non-BRCA1/2 families in order to find novel susceptibility loci and high-penetrant genes. Materials and Methods: Genotyping with 540 fluorescently-labeled microsatellite markers located on the 23 chromosomes at 7.25 cM resolution was used for primary linkage analysis and an additional 40 markers were used for fine-mapping of loci with a logarithm of odds (LOD) or heterogeneity LOD (HLOD) score greater than one. Whole-exome sequencing data of 28 members from all 13 families were used for the bioinformatics sequence analysis on the linked regions of these families. Results: Linkage analysis identified three loci on chromosome 18q as a putative region of interest (overall LOD=1, HLOD=1.2). Sequencing analysis of the three linked regions on 18q and mutation prediction algorithms did reveal three probable damaging variants. Conclusion: Overall, our study identified three weakly linked loci on 18q and three probable damaging variants of interest in the 13 families with breast cancer.

    • Climate change and the induced environmental disturbances is one of the major threats that have a strong impact on bacterial communities in the Antarctic environment. To cope with the persistent extreme environment and inhospitable conditions, psychrophilic bacteria are thriving and displaying striking adaptive characteristics towards severe external factors including freezing temperature, sea ice, high radiation and salinity which indicates their potential in regulating climate change’s environmental impacts. The review illustrates the different adaptation strategies of Antarctic microbes to changing climate factors at the structural, physiological and molecular level. Moreover, we discuss the recent developments in “omics” approaches to reveal polar “blackbox” of psychrophiles in order to gain a comprehensive picture of bacterial communities. The psychrophilic bacteria synthesize distinctive cold-adapted enzymes and molecules that have many more industrial applications than mesophilic ones in biotechnological industries. Hence, the review also emphasizes on the biotechnological potential of psychrophilic enzymes in different sectors and suggests the machine learning approach to study cold–adapted bacteria and engineering the industrially important enzymes for sustainable bioeconomy.

  • A qualitative study on antibiotic use and animal health management in smallholder dairy farms of four regions of India

    • BackgroundAntimicrobial resistance (AMR) is a major global public health issue. In India, access to medicines is poorly regulated and therefore antibiotics in dairy cattle are commonly used by farmers without consulting with veterinarians. This study was conducted to understand practices and knowledge related to antibiotic use and AMR among dairy farmers and veterinary professionals in selected urban and peri-urban areas of India.MethodsA total of 28 focus group discussions with farmers and 53 interviews with veterinary professionals were carried out.ResultsMastitiswas identified as the main animal health challenge. Antibiotic consultation behavior of farmers depended on the availability of veterinarians. Except in Bangalore, farmers were found to often treat animals on their own. They were found unaware of the concept of AMR, but knew the importance of vaccination. Veterinarians included in the study had a good understanding of antibiotics, AMR, and zoonotic diseases.ConclusionThe knowledge level and practices observed in the study related to the use/abuse of antibiotics can potentially increase the risk of development of AMR and its transfer in the community. Our findings can help support AMR – mitigation efforts in the country, including the design of better policies on antibiotic use in dairy.

  • Comparing the Effectiveness of Different Approaches to Raise Awareness About Antimicrobial Resistance in Farmers and Veterinarians of India

    • BackgroundAntimicrobial resistance (AMR) is a global public health threat. The indiscriminate use of antibiotics in the animal health sector contributes to increasing rates of AMR and studies involving dairy farmers in India have found knowledge levels regarding antibiotics and AMR to be very low. The purpose of this study was to assess different methods to raise awareness and knowledge about AMR and antibiotic use among dairy farmers, paravets (veterinary assistants), and veterinarians. Materials and MethodsThe study was conducted in September-December of 2018 in some parts of Haryana, Assam, Karnataka, and West Bengal. It had two parts: an intervention meeting (September-October 2018) which consisted of focus group discussions (FGD) with farmers, key informant interviews (KII) with veterinary professionals along with distribution of information packages, and then a follow-up survey (November-December 2018). The villages were randomly allocated to either one of the four intervention approaches (1-FGD/KII and information package on AMR; 2-FGD/KI and information on animal health; 3- FGD/KII and information package on animal health plus information on AMR; or 4- only the FGD/KII). A follow-up survey was done to assess the effect of interventions. ResultsIn total, 274 dairy farmers and 51 veterinary professionals (21 veterinarians and 30 paravets) participated in the follow-up survey. Many of the farmers and veterinary professionals who participated in the follow-up survey had been part of the intervention meetings. The average knowledge score of farmers was 7.8. It was found that the knowledge score was higher amongst farmers who had participated in the intervention meetings (p < 0.05), had received intervention approach 2 (p = 0.03) or approach 3 (p = 0.01), and amongst female farmers (p = 0.03) compared to male. The veterinary professionals had good knowledge but lacked interest in training the farmers about antimicrobial resistance. ConclusionOur research demonstrated that a higher percentage of farmers and veterinary professionals who attended the intervention meeting had improved knowledge. Dairy farmers should be regularly educated on antibiotic usage and how to avoid misusing them. Also, veterinary experts should be provided with tools and strategies to educate farmers on the use of antimicrobials.
