  • Robust estimation of bacterial cell count from optical density
  • 2020
  • Ingår i: Communications Biology. - : Springer Science and Business Media LLC. - 2399-3642. ; 3:1
  • Tidskriftsartikel (refereegranskat)abstract
    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

  • Review of the European project - Impact of Accelerator-Based Technologies on Nuclear Fission Safety (IABAT)
  • 2001
  • Ingår i: Progress in nuclear energy (New series). - 0149-1970 .- 1878-4224. ; 38:1-2, s. 135-151
  • Tidskriftsartikel (refereegranskat)abstract
    • The IABAT project - Impact of Accelerator Based Technologies on Nuclear Fission Safety - started in 1996 in the frame of 4(th) Framework Programme of the European Union, R&D specific programme Nuclear fission safety 1994-1998, area A.2 Exploring innovative approaches/Fuel cycle concepts, as one of the first common European activities in ADS. The project was completed October 31, 1999. The overall objective of the IABAT project has been a preliminary assessment of the potential of Accelerator-Driven Systems (ADS) for transmutation of nuclear waste and for nuclear energy production with minimum waste generation. Moreover, more specific topics related to nuclear data and code development for ADS have been studied in more detail. Four ADSs have been studied for different fuel/coolant combinations: liquid metal coolant and solid fuel, liquid metal coolant and dispersed fuel, and fast and thermal molten salt systems. Target studies comprised multiple target solutions and radiation damage problems in a target environment. In a tool development part of the project a methodology of subcriticality monitoring has been developed based on Feynman-alpha and Rossi-alpha methods. Moreover, a new Monte-Carlo burnup code taking full advantage of continuous neutron cross-section data has been developed and benchmarked. Impact on the risk from high-level waste repositories fi om radiotoxicity reduction using ADS has been assessed giving no crystal-clear benefits of ADS for repository radiotoxicity reduction but concluding some important prerequisites for effective transmutation. In proliferation studies important differences between critical reactors and ADS have been underlined and non-proliferation measures have been proposed. In assessment of accelerator technology costing models have been created that allow the circular and linear accelerator options to be compared and the effect of parameter variations examined. The calculations reported show that cyclotron systems would be more economical, due mainly to the advantage of the cost of RF power supplies. However, the accelerator community regards with skepticism the possibility of transporting and extracting more than a 10mA beam current from a 1GeV cyclotron and therefore technical factors may limit the application of cyclotrons. Finally, this review summarizes development of nuclear data in the energy region between 20 Mev and 150 MeV. Neutron and proton transport data files for Fe, Ni, Pb, Th, U-238 and Pu-239 have been created. The high-energy part of the data files consists completely of results from model calculations, which are benchmarked against the available experimental data. Although there is obviously future work left regarding fine-tuning of several parts of the data files, the representation of nuclear reaction information up to 150 MeV is already better than can be attained with intranuclear cascade codes.

  • Addressing the challenges of E-cigarette safety profiling by assessment of pulmonary toxicological response in bronchial and alveolar mucosa models
  • 2020
  • Ingår i: Scientific reports. - : Springer Science and Business Media LLC. - 2045-2322. ; 10:1, s. 20460-
  • Tidskriftsartikel (refereegranskat)abstract
    • Limited toxicity data on electronic cigarette (ECIG) impede evidence-based policy recommendations. We compared two popular mixed fruit flavored ECIG-liquids with and without nicotine aerosolized at 40 W (E-smoke) with respect to particle number concentrations, chemical composition, and response on physiologically relevant human bronchial and alveolar lung mucosa models cultured at air–liquid interface. E-smoke was characterized by significantly increased particle number concentrations with increased wattage (25, 40, and 55 W) and nicotine presence. The chemical composition of E-smoke differed across the two tested flavors in terms of cytotoxic compounds including p-benzoquinone, nicotyrine, and flavoring agents (for example vanillin, ethyl vanillin). Significant differences in the expression of markers for pro-inflammation, oxidative stress, tissue injury/repair, alarm anti-protease, anti-microbial defense, epithelial barrier function, and epigenetic modification were observed between the flavors, nicotine content, and/ or lung models (bronchial or alveolar). Our findings indicate that ECIG toxicity is influenced by combination of multiple factors including flavor, nicotine content, vaping regime, and the region of respiratory tree (bronchial or alveolar). Toxic chemicals and flavoring agents detected in high concentrations in the E-smoke of each flavor warrant independent evaluation for their specific role in imparting toxicity. Therefore, multi-disciplinary approaches are warranted for comprehensive safety profiling of ECIG.

  • Generation of Bis-cationic heterocyclic inhibitors of Bacillus subtilis HPr kinase/phosphatase from a ditopic dynamic combinatorial library
  • 2003
  • Ingår i: Journal of Medicinal Chemistry. - : American Chemical Society (ACS). - 0022-2623 .- 1520-4804. ; 46:26, s. 5803-5811
  • Tidskriftsartikel (refereegranskat)abstract
    • Ditopic dynamic combinatorial. libraries were generated and screened toward inhibition of the bifunctional enzyme HPr kinase/phosphatase from Bacillus subtilis. The libraries were composed of all possible combinations resulting from the dynamic interconversion of 16 hydrazides and five monoaldehyde or dialdehyde building blocks, resulting in libraries containing up to 440 different constituents. Of all possible acyl hydrazones formed, active compounds containing two terminal cationic heterocyclic recognition groups separated by a spacer of appropriate structure could be rapidly identified using a dynamic deconvolution procedure. Thus, parallel testing of sublibraries where one specific component was excluded basically revealed all the essential components. A potent ditopic inhibitor, based on 2-amino-benzimidazole, was identified from the process.

  • Mapping pathological (99m)Tc-d,l-hexamethylpropylene amine oxime uptake in Alzheimer's disease and frontal lobe dementia with SPECT
  • 2001
  • Ingår i: Dementia and geriatric cognitive disorders. - : S. Karger AG. - 1420-8008 .- 1421-9824. ; 12:3, s. 177-184
  • Tidskriftsartikel (refereegranskat)abstract
    • Seventeen patients with probable Alzheimer’s disease (AD), 7 patients with frontal lobe dementia (FLD) and 19 control subjects (NOR) were examined by 99mTc-<i>d,l</i>- hexamethylpropylene amine oxime (99mTc-HMPAO) SPECT. Images were standardised in the same 3D space and averaged within each group. After normalisation, the three sets of images were analysed in all cerebral lobes, hippocampus, thalamus and basal ganglia. In AD, the 99mTc-HMPAO uptake values were significantly reduced, as compared to NOR, in the parietal, temporal and insular lobes. In patients with FLD, the uptake was altered in all lobes with the exception of the parietal lobe. The uptake in the nucleus caudatus decreased significantly in both AD and FLD as compared to NOR. The uptake in the anterior cingulate cortex was significantly reduced in FLD. Subtraction images highlighted all significantly decreased areas. In conclusion, standardising SPECT in a common space and subtracting data from a control group improves the visual interpretation of images. In this study, the typical temporo-parietal and fronto-parietal 99mTc-HMPAO uptake reductions were found in AD and FLD, respectively. The uptake in the nucleus caudatus was found to decrease significantly in AD and FLD and the one in the anterior cingulate cortex was reduced in FLD.
