    • Motivation: The BioTIME database contains raw data on species identities and abundances in ecological assemblages through time. These data enable users to calculate temporal trends in biodiversity within and amongst assemblages using a broad range of metrics. BioTIME is being developed as a community-led open-source database of biodiversity time series. Our goal is to accelerate and facilitate quantitative analysis of temporal patterns of biodiversity in the Anthropocene. Main types of variables included: The database contains 8,777,413 species abundance records, from assemblages consistently sampled for a minimum of 2 years, which need not necessarily be consecutive. In addition, the database contains metadata relating to sampling methodology and contextual information about each record. Spatial location and grain: BioTIME is a global database of 547,161 unique sampling locations spanning the marine, freshwater and terrestrial realms. Grain size varies across datasets from 0.0000000158 km(2) (158 cm(2)) to 100 km(2) (1,000,000,000,000 cm(2)). Time period and grainBio: TIME records span from 1874 to 2016. The minimal temporal grain across all datasets in BioTIME is a year. Major taxa and level of measurement: BioTIME includes data from 44,440 species across the plant and animal kingdoms, ranging from plants, plankton and terrestrial invertebrates to small and large vertebrates.

  • Biallelic variants in HPDL cause pure and complicated hereditary spastic paraplegia

    • Human 4-hydroxyphenylpyruvate dioxygenase-like (HPDL) is a putative iron-containing non-heme oxygenase of unknown specificity and biological significance. We report 25 families containing 34 individuals with neurological disease associated with biallelic HPDL variants. Phenotypes ranged from juvenile-onset pure hereditary spastic paraplegia to infantile-onset spasticity and global developmental delays, sometimes complicated by episodes of neurological and respiratory decompensation. Variants included bona fide pathogenic truncating changes, although most were missense substitutions. Functionality of variants could not be determined directly as the enzymatic specificity of HPDL is unknown; however, when HPDL missense substitutions were introduced into 4-hydroxyphenylpyruvate dioxygenase (HPPD, an HPDL orthologue), they impaired the ability of HPPD to convert 4-hydroxyphenylpyruvate into homogentisate. Moreover, three additional sets of experiments provided evidence for a role of HPDL in the nervous system and further supported its link to neurological disease: (i) HPDL was expressed in the nervous system and expression increased during neural differentiation; (ii) knockdown of zebrafish hpdl led to abnormal motor behaviour, replicating aspects of the human disease; and (iii) HPDL localized to mitochondria, consistent with mitochondrial disease that is often associated with neurological manifestations. Our findings suggest that biallelic HPDL variants cause a syndrome varying from juvenile-onset pure hereditary spastic paraplegia to infantile-onset spastic tetraplegia associated with global developmental delays. © 2021 The Author(s).

    • Mutations in the mitochondrial or nuclear genomes are associated with a diverse group of human disorders characterized by impaired mitochondrial respiration. Within this group, an increasing number of mutations have been identified in nuclear genes involved in mitochondrial RNA biology. The TEFM gene encodes the mitochondrial transcription elongation factor responsible for enhancing the processivity of mitochondrial RNA polymerase, POLRMT. We report for the first time that TEFM variants are associated with mitochondrial respiratory chain deficiency and a wide range of clinical presentations including mitochondrial myopathy with a treatable neuromuscular transmission defect. Mechanistically, we show muscle and primary fibroblasts from the affected individuals have reduced levels of promoter distal mitochondrial RNA transcripts. Finally, tefm knockdown in zebrafish embryos resulted in neuromuscular junction abnormalities and abnormal mitochondrial function, strengthening the genotype-phenotype correlation. Our study highlights that TEFM regulates mitochondrial transcription elongation and its defect results in variable, tissue-specific neurological and neuromuscular symptoms.

  • Evaluating Nature-Based Solutions for Water Management in Peri-Urban Areas

    • The term nature-based solutions (NBS) has gained traction in recent years and has been applied in many settings. There are few comprehensive assessment frameworks available that can guide NBS planning and implementation while at the same time capturing the short- and long-term impacts and benefits of the NBS. Here a recently presented framework, which builds on the theory of change and was developed to assess NBS at different phases of the project cycle, was applied to seven diverse case studies. The case studies addressed water quality and quantity issues in peri-urban areas across the global north and south. Framework indicators covering the sustainability dimensions (environmental, social and economic) were assessed at three stages of the framework: context, process and results. The work sought to investigate the following research objectives: (1) Can this framework be robust and yet flexible enough to be applied across a diverse selection of NBS projects that are at different phases of the project cycle and address different kinds of water challenges within varied ecological, social and economic contexts? (2) Is it possible to draw generalisations from a comparative analysis of the application of the framework to the case studies? Results showed that the framework was able to be applied to the case studies; however, their diversity showed that NBS projects designed in one context, for a specific purpose in a specific location, can not necessarily be transferred easily to another location. There were several process-based indicators that were universally significant for the case studies, including expertise, skills and knowledge of the involved actors, roles and responsibilities of involved actors and political support. The result-based indicators were case study-specific when environmental indicators were case study-specific, and important social indicators were environmental identity and recreational values. Overall, the use of the framework benefits the recognition of the implementation’s advances, such as the change in context, the processes in place and the results obtained.

  • Levelling-up rhodolith-bed science to address global-scale conservation challenges

    • Global marine conservation remains fractured by an imbalance in research efforts and policy actions, limiting progression towards sustainability. Rhodolith beds represent a prime example, as they have ecological importance on a global scale, provide a wealth of ecosystem functions and services, including biodiversity provision and potential climate change mitigation, but remain disproportionately understudied, compared to other coastal ecosystems (tropical coral reefs, kelp forests, mangroves, seagrasses). Although rhodolith beds have gained some recognition, as important and sensitive habitats at national/regional levels during the last decade, there is still a notable lack of information and, consequently, specific conservation efforts. We argue that the lack of information about these habitats, and the significant ecosystem services they provide, is hindering the development of effective conservation measures and limiting wider marine conservation success. This is becoming a pressing issue, considering the multiple severe pressures and threats these habitats are exposed to (e.g., pollution, fishing activities, climate change), which may lead to an erosion of their ecological function and ecosystem services. By synthesizing the current knowledge, we provide arguments to highlight the importance and urgency of levelling-up research efforts focused on rhodolith beds, combating rhodolith bed degradation and avoiding the loss of associated biodiversity, thus ensuring the sustainability of future conservation programs.

  • The inter-individual relationship between training status and activity pattern during small-sided and full-sized games in professional male football players

    • Objectives: We analysed the inter-individual relationship between training status and activity pattern in small-sided games (SSG) and full-sized games (FG). Methods: Fourteen Italian elite male senior football players were monitored during four training sessions incorporating SSG, and six FG, during the pre-season. Baseline measurements included, countermovement jump (CMJ), 5-m sprint time and Yo-Yo intermittent recovery test level 1 distance (YYIR1). Activity pattern was collected via 15-Hz global positioning system with 100-Hz accelerometer integrated. Results: Maximum accelerations (Accmax) and maximum decelerations (Decmax) showed moderate correlations (P < 0.05; r = 0.49 and 0.52) between SSG and FG. YYIR1 correlated with total distance covered (TD) in both SSG and FG (P < 0.05; r = 0.42 to 0.57) and with high-intensity distance (HIR) during FG (P < 0.05; r = 0.46 ± 0.38). CMJ showed positive correlations with TD, Accmax, Decmax, total accelerations (Acctot) and total decelerations (Dectot) during FG (P < 0.05; r = 0.40 ± 0.38, 0.41 ± 0.40, 0.46 ± 0.38, 0.65 ± 0.29, 0.57 ± 0.33). Multiple regression analysis revealed Decmax during SSG, as significant predictor of physical performance during FG, explaining a significant amount of variation in HIR, very-high speed running, Accmax and Decmax (P < 0.05; R2 = 0.49, 0.44, 0.26 and 0.49) during FG. Conclusions: SSG seems more effective to enhance muscular efforts occurring during FG, such as accelerations and decelerations, rather than speed patterns. Specifically, Decmax distance imposed during SSG was identified as the predictor of players’ movement patterns during FG. Thus, importance of eccentric strength (i.e. imposed when decelerating) to achieve football-specific fitness is emphasised. These findings may assist coaches and any football practitioners for pre-season programming. © 2017, © 2017 Informa UK Limited, trading as Taylor & Francis Group.

  • Futures for invasive alien species management : using bottom-up innovations to envision positive systemic change

    • Invasive alien species (IAS) pose a key threat to biodiversity, the economy and human well-being, and continue to increase in abundance and impact worldwide. Legislation and policy currently dominate the global agenda for IAS, although translation to localised success may be limited. This calls for a wider range of responses to transform IAS management. An under-appreciated strategy to achieve success may come from bottom-up, experimental innovations (so-called “seeds”), which offer alternative visions of what may be possible for IAS management in the future. We present an application of a participatory process that builds on such innovations to create alternative visions of the future, with actionable pathways to guide change. Through a series of workshops with practitioners and academics, we used this process to explore alternative positive futures for IAS management in South Africa. We then identified a set of domains of change, that could enable these visions to be actioned by appropriate stakeholders. The domains of change highlight the social–ecological nature of the IAS sector, with interconnected actions needed in financial, cultural, social, technological and governance spheres. Key domains identified were the need to shift mindsets and values of society regarding IAS, as well as the need for appropriate and functional financing. This participatory futuring process offers a way to interrogate and scale bottom-up innovations, thereby creating optimism and allowing stakeholders to engage constructively with the future. This represents an important step in fostering the potential of bottom-up innovations to transform IAS management.
