  • Man-made Mediterranean temporary ponds as a tool for amphibian conservation

    • Mediterranean temporary ponds (MTPs) are crucial breeding sites for local amphibians; a faunal group in decline in the Mediterranean mainly due to wetland destruction. Although the disappearance of lentic habitats in other regions of the world has been ameliorated by the creation and restoration of wetlands, these tactics remain untested in Mediterranean wetlands. To evaluate the amphibian colonization dynamics of artificial MTPs in the north-eastern Iberian Peninsula, we monitored two artificial MTPs (one in a high-diversity area and the other in a low-diversity area) over 2 years. We selected reference wetlands at each area, and amphibian communities of man-made MTPs were compared to those of reference wetlands. We found that, beginning in the first year; artificial MTPs were a valuable habitat for regional amphibian fauna. Posteriorly, we focused on the reproductive success of the natterjack toad, Bufo calamita. Using a drift fence equipped with pit-fall traps, we measured the abundance of both reproductive adults entering the pond and post-metamorphic individuals exiting the pond. Using mark-recapture methods, we analyzed the time and energy costs associated with reproduction in the artificial MTPs. We observed a highly variable colonization dynamics between sites, between hydroperiods, and within hydroperiods, and these dynamics often depended on meteorological factors. Our results suggest that even at a short timescale, native amphibian species such as B. calamita can benefit from wetland creation in the Mediterranean region.

  • Antimicrobial peptide and sequence variation along a latitudinal gradient in two anurans

    • Background While there is evidence of both purifying and balancing selection in immune defense genes, large-scale genetic diversity in antimicrobial peptides (AMPs), an important part of the innate immune system released from dermal glands in the skin, has remained uninvestigated. Here we describe genetic diversity at three AMP loci (Temporin, Brevinin and Palustrin) in two ranid frogs (Rana arvalis and R. temporaria) along a 2000 km latitudinal gradient. We amplified and sequenced part of the Acidic Propiece domain and the hypervariable Mature Peptide domain ( 150-200 bp) in the three genes using Illumina Miseq and expected to find decreased AMP genetic variation towards the northern distribution limit of the species similarly to studies on MHC genetic patterns. Results We found multiple loci for each AMP and relatively high gene diversity, but no clear pattern of geographic genetic structure along the latitudinal gradient. We found evidence of trans-specific polymorphism in the two species, indicating a common evolutionary origin of the alleles. Temporin and Brevinin did not form monophyletic clades suggesting that they belong to the same gene family. By implementing codon evolution models we found evidence of strong positive selection acting on the Mature Peptide. We also found evidence of diversifying selection as indicated by divergent allele frequencies among populations and high Theta k values. Conclusion Our results suggest that AMPs are an important source of adaptive diversity, minimizing the chance of microorganisms developing resistance to individual peptides.

  • Major Histocompatibility Complex Variation and Haplotype Associated Survival in Response to Experimental Infection of Two Bd-GPL Strains Along a Latitudinal Gradient

    • While both innate and adaptive immune system mechanisms have been implicated in resistance against the chytrid fungus Batrachochytrium dendrobatidis (Bd), studies on the role of specific MHC haplotypes on Bd infection are rare. Here, we studied variation in MHC Class IIB loci in the common toad Bufo bufo along a latitudinal gradient across Sweden. In general, Swedish toad populations had few MHC Class IIB haplotypes and MHC diversity declined from south (13 haplotypes) to the north (four haplotypes). The low diversity may compromise the ability of northern populations to fight emerging disease, such as Bd. In a laboratory experiment, we infected newly metamorphosed toads with two strains of the Global Pandemic Lineage of the fungus (Bd-GPL) and compared survival with sham controls. Bd-infected toads had lower survival compared to controls. Moreover, survival was dependent on the Bd-strain and northern toads had lower Bd-mediated survival than southern individuals. MHC diversity was lower in northern toads. All northern experimental animals were monomorphic for a single MHC haplotype, whereas we found seven different haplotypes in southern experimental animals. In southern toads, survival was dependent on both Bd-strain and MHC haplotype suggesting differential infection dynamics depending on both Bd-strain and host immune system characteristics.

  • Can amphibians take the heat? : Vulnerability to climate warming in subtropical and temperate larval amphibian communities

    • Predicting the biodiversity impacts of global warming implies that we know where and with what magnitude these impacts will be encountered. Amphibians are currently the most threatened vertebrates, mainly due to habitat loss and to emerging infectious diseases. Global warming may further exacerbate their decline in the near future, although the impact might vary geographically. We predicted that subtropical amphibians should be relatively susceptible to warming-induced extinctions because their upper critical thermal limits (CTmax) might be only slightly higher than maximum pond temperatures (Tmax). We tested this prediction by measuring CTmax and Tmax for 47 larval amphibian species from two thermally distinct subtropical communities (the warm community of the Gran Chaco and the cool community of Atlantic Forest, northern Argentina), as well as from one European temperate community. Upper thermal tolerances of tadpoles were positively correlated (controlling for phylogeny) with maximum pond temperatures, although the slope was steeper in subtropical than in temperate species. CTmax values were lowest in temperate species and highest in the subtropical warm community, which paradoxically, had very low warming tolerance (CTmaxTmax) and therefore may be prone to future local extinction from acute thermal stress if rising pond Tmax soon exceeds their CTmax. Canopy-protected subtropical cool species have larger warming tolerance and thus should be less impacted by peak temperatures. Temperate species are relatively secure to warming impacts, except for late breeders with low thermal tolerance, which may be exposed to physiological thermal stress in the coming years.

    • The extension of road networks is considered one of the major factors affecting fauna survival. Roadkill has been documented widely and affects all taxonomic groups. Although roadkill is associated mainly with traffic density, some life-history traits of species and the area surrounding roads are expected to modify number of roadkills both taxonomically and geographically. Here we studied the number of roadkills of vertebrates in an extensive region in the northeastern Iberian Peninsula. We surveyed 820 km of 41 roads in two different seasons (spring and autumn), that differ in traffic intensity. In addition, we covered zones with distinct climatic characteristics and levels of protection of the surrounding habitats. Amphibians showed the highest number of roadkills whereas reptiles, birds and mammals had similar rates. General Linear Model tests showed no differences in roadkills by climatic region; however, differences in number of roadkills were linked to protection status, with the highest number of casualties in highly protected areas. Redundancy Analysis demonstrated that the number of amphibians and reptiles killed was associated with roads in highly protected areas whereas that of mammals and birds was linked to unprotected areas. Protected areas often receive many visitors, which in turn may increase wildlife casualties as a result of greater traffic density. We recommend that correction measures be taken to reduce the high number of vertebrate fauna killed along roads that cross protected areas.

    • Environmental induced developmental plasticity occurs in many organisms and it has been suggested to facilitate biological diversification. Here we use ranid frogs to examine whether morphological changes derived from adaptive developmental acceleration in response to pool drying within a species are mirrored by differences among populations and across species. Accelerated development in larval anurans under pool drying conditions is adaptive and often results in allometric changes in limb length and head shape. We examine the association between developmental rate and morphology within population, among populations in divergent environments, and among species inside the Ranidae frog family, combining experimental approaches with phylogenetic comparative analyses. We found that frogs reared under decreasing water conditions that simulated fast pool drying had a faster development rate compared to tadpoles reared on constant water conditions. This faster developmental rate resulted in different juvenile morphologies between the two pool drying conditions. The association between developmental rate and morphology found as a result of plasticity was not mirrored by differences among populations that differed in development, neither was it mirrored among species that differed in development rate. We conclude that morphological differences among populations and species were not driven by variation in developmental time per se. Instead, selective factors, presumably operating on locomotion and prey choice, seem to have had a stronger evolutionary effect on frog morphology than evolutionary divergences in developmental rate in the ranid populations and species studied.

    • It has been suggested that phenotypic plasticity can facilitate evolutionary diversification of organisms. If life-history and morphological diversification across a lineage is mirrored in diversification in the same traits due to phenotypic plasticity within a lineage it fulfils one of the expectations that are needed to support this diversification hypothesis. We carried out a laboratory study to examine development rate and morphology between and within populations of the parsley frog, Pelodytes punctatus. We found that frogs reared in the laboratory had a longer development time, relatively longer hind legs and relatively narrower heads under constant water level compared to those under decreasing water level simulating pool drying. This adaptive phenotypic plasticity response to pool drying was mirrored across populations because frogs from permanent waters had longer development times, relatively longer hind legs and relatively narrower heads compared to frogs from temporary waters. Hence the developmental and morphological plasticity observed within populations was also observed between populations as constitutive expressed traits. We suggest that the morphology pattern observed was driven by a common developmental process (time to metamorphosis), indicating that plasticity may contribute to evolutionary change, ultimately resulting in genetic accommodation of the morphological traits.

    • Ectotherm development rates often show adaptive divergence along climatic gradients, but the genetic basis for this variation is rarely studied. Here, we investigated the genetic basis for phenotypic variation in larval development in the moor frog Rana arvalis from five regions along a latitudinal gradient from Germany to northern Sweden. We focused on the C/EBP-1 gene, a transcription factor associated with larval development time. Allele frequencies at C/EBP-1 varied strongly among geographical regions. Overall, the distribution of alleles along the gradient was in concordance with the dual post-glacial colonization routes into Scandinavia, with a large number of alleles exclusively present along the southern colonization route. Only three of 38 alleles were shared between the routes. Analysis of contemporary selection on C/EBP-1 showed divergent selection among the regions, probably reflecting adaptation to the local environmental conditions, although this was especially strong between southern and northern regions coinciding also with lineages from different colonization routes. Overall, the C/EBP-1 gene has historically been under purifying selection, but two specific amino acid positions showed significant signals of positive selection. These positions showed divergence between southern and northern regions, and we suggest that they are functionally involved in the climatic adaptation of larval development. Using phenotypic data from a common garden experiment, we found evidence for specific C/EBP-1 alleles being correlated with larval development time, suggesting a functional role in adaptation of larval development to large-scale climatic variation.

    • The longitudinal movements and abiotic factors that determine the distribution of Pyrenean mountain newts (males, females, amplexus and metamorphic newts) were examined in a 1500 meters segment of a Pre-Pyrenean stream. Migration of adult newts was studied by means of capture-recapture techniques in the course of one year. Our results indicate that the mean distance the newts migrated per year was less than 50 meters and the population could be considered sedentary. No significant differences in longitudinal movement patterns between the sexes were observed, and movement patterns were more or less constant over time. Habitat variables determining newt abundance was estimated by means of a GLM. Our results indicate that the number of refugia (wood debris, stones and fissures) determines the distribution of newts. Larval abundance was correlated with stream-bed structure.

    • Stochastic effects from demographic processes and selection are expected to shape the distribution of genetic variation in spatially heterogeneous environments. As the amount of genetic variation is central for long‐term persistence of populations, understanding how these processes affect variation over large‐scale geographical gradients is pivotal. We investigated the distribution of neutral and putatively adaptive genetic variation, and reconstructed demographic history in the moor frog (Rana arvalis) using 136 individuals from 15 populations along a 1,700‐km latitudinal gradient from northern Germany to northern Sweden. Using double digest restriction‐site associated DNA sequencing we obtained 27,590 single nucleotide polymorphisms (SNPs), and identified differentiation outliers and SNPs associated with growing season length. The populations grouped into a southern and a northern cluster, representing two phylogeographical lineages from different post‐glacial colonization routes. Hybrid index estimation and demographic model selection showed strong support for a southern and northern lineage and evidence of gene flow between regions located on each side of a contact zone. However, patterns of past gene flow over the contact zone differed between neutral and putatively adaptive SNPs. While neutral nucleotide diversity was higher along the southern than the northern part of the gradient, nucleotide diversity in differentiation outliers showed the opposite pattern, suggesting differences in the relative strength of selection and drift along the gradient. Variation associated with growing season length decreased with latitude along the southern part of the gradient, but not along the northern part where variation was lower, suggesting stronger climate‐mediated selection in the north. Outlier SNPs included loci involved in immunity and developmental processes.
