    • This paper presents the potential for the Swedish Laqua system to be used as a sustainable method for on-site landfill leachate management in Northern Ireland, specifically the potential to use locally sourced filter materials from Northern Ireland as part of the filter system. Four carbon containing ashes and four types of peat were tested over a 24 hours period by a shaking test with untreated landfill leachate. Considering the results of this screening test, and the economical and sustainable supply of filter materials, one combination of ash and peat was selected to be column tested. Column testing with artificial leachate containing 7 organic pollutants (3 PAHs and 4 PCBs) and 9 inorganic pollutants showed that locally sourced filter materials effectively removed both organic and inorganic pollutants. A subsequent column test with landfill leachate for 13 weeks demonstrated it was feasible to apply the Laqua system with economical locally sourced filter materials.

    • CD169(+) resident macrophages in lymph nodes of breast cancer patients are for unknown reasons associated with a beneficial prognosis. This contrasts CD169(+) macrophages present in primary breast tumors (CD169(+) TAMs), that correlate with a worse prognosis. We recently showed that these CD169(+) TAMs were associated with tertiary lymphoid structures (TLSs) and T-regs in breast cancer. Here, we show that CD169(+) TAMs can be monocyte-derived and express a unique mediator profile characterized by type I IFNs, CXCL10, PGE(2) and inhibitory co-receptor expression pattern. The CD169(+) monocyte-derived macrophages (CD169(+) Mo-M) possessed an immunosuppressive function in vitro inhibiting NK, T and B cell proliferation, but enhanced antibody and IL6 secretion in activated B cells. Our findings indicate that CD169(+) Mo-M in the primary breast tumor microenvironment are linked to both immunosuppression and TLS functions, with implications for future targeted Mo-M therapy.

    • Background Ex vivo gene therapy in the central nervous system (CNS) holds great promise for diseases such as the neurodegenerative disorders. However, achieving stable, long-term transgene expression in grafted cells has proven problematic. This study reports the establishment of an in vitro model of transgene down-regulation in cells grafted to the CNS using the immortalized neural progenitor cell lines HiB5 and RN33B. Methods Neural cell lines were transduced at 33 C with different GFP constructs, both viral and non-viral, containing either viral or non-viral promoters. Cell differentiation in vitro was obtained by culturing the cells at 37 C in serum-free defined media, which halts cell division, and GFP-expression was analysed by FACS. As early as day 3 of culture at 37degreesC, the transgene expression decreased markedly in most cell lines. To validate the assay, the same clones were grafted to the adult rat striatum and the down-regulation of GFP-expression was evaluated. Results The temporal pattern of down-regulation was found to be similar in vitro and in vivo. Using this assay, it was shown that addition of inhibitors of histone deacetylation, but not an inhibitor of DNA methylation, reversed the silencing of GFP in quiescent neural progenitors by up to 308% of control values. Conclusion These results suggest that the same mechanisms controlling gene transcription of the host cell's genome are active in controlling transgene expression and that this should be taken into account when constructing vectors for gene therapy. The assay reported in this study could be used as a screening method to evaluate new vectors. Copyright (C) 2002 John Wiley Sons, Ltd.

  • Recommendations for Next-Generation Ground Magnetic Perturbation Validation

    • Data-model validation of ground magnetic perturbation forecasts, specifically of the time rate of change of surface magnetic field, dB/dt, is a critical task for model development and for mitigation of geomagnetically induced current effects. While a current, community-accepted standard for dB/dt validation exists (Pulkkinen et al., 2013), it has several limitations that prevent more complete understanding of model capability. This work presents recommendations from the International Forum for Space Weather Capabilities Assessment Ground Magnetic Perturbation Working Team for creating a next-generation validation suite. Four recommendations are made to address the existing suite: greatly expand the number of ground observatories used, expand the number of events included in the suite from six to eight, generate metrics as a function of magnetic local time, and generate metrics as a function of activity type. For each of these, implementation details are explored. Limitations and future considerations are also discussed.
