    • This work describes new electronics for the EX-OGAM2 (HP-Ge detector array) and NEDA (BC501A-based neutron detector array). A new digitizing card with high resolution has been designed for gamma-ray and neutron spectroscopy experiments. The higher bandwidth requirement of the NEDA signals, together with the necessity for accuracy, require a high sampling rate in order to preserve the shape for real-time Pulse Shape Analysis (PSA). The PSA is of paramount importance for the NEDA to discriminate between neutrons and gamma-ray signals. Both high resolution and high speed parameters are often difficult to achieve in a single electronic unit. These constraints, together with the need to build new digitizing electronics to improve performance and flexibility of signal analysis in nuclear physics experiments, led to the development a new FADC mezzanine card. In this work, the design and development are described, including the characterization procedure and the preliminary measurement results.

    • Characterization of spatial patterns of functional microbial communities could facilitate the understanding of the relationships between the ecology of microbial communities, the biogeochemical processes they perform and the corresponding ecosystem functions. Because of the important role the ammonia-oxidizing bacteria (AOB) and archaea (AOA) have in nitrogen cycling and nitrate leaching, we explored the spatial distribution of their activity, abundance and community composition across a 44-ha large farm divided into an organic and an integrated farming system. The spatial patterns were mapped by geostatistical modeling and correlations to soil properties and ecosystem functioning in terms of nitrate leaching were determined. All measured community components for both AOB and AOA exhibited spatial patterns at the hectare scale. The patchy patterns of community structures did not reflect the farming systems, but the AOB community was weakly related to differences in soil pH and moisture, whereas the AOA community to differences in soil pH and clay content. Soil properties related differently to the size of the communities, with soil organic carbon and total nitrogen correlating positively to AOB abundance, while clay content and pH showed a negative correlation to AOA abundance. Contrasting spatial patterns were observed for the abundance distributions of the two groups indicating that the AOB and AOA may occupy different niches in agro-ecosystems. In addition, the two communities correlated differently to community and ecosystem functions. Our results suggest that the AOA, not the AOB, were contributing to nitrate leaching at the site by providing substrate for the nitrite oxidizers.     • Patients with peritoneal metastasis from colorectal cancer (PMCRC) may have a chance of cure when treated with cytoreductive surgery (CRS) combined with heated intraperitoneal chemotherapy (HIPEC)1–5.Choice of chemotherapy for HIPEC has been based on knowledge of its systemic effects, pharmacokinetics, technical feasibility, hyperthermic efficacy enhancement, and tolerance6–8. Selection of cancer drugs for treatment based on phenotypical assessment of patient cancer cell drug sensitivity ex vivo is one approach to personalized cancer treatment. One technique for this is the fluorometric microculture cytotoxicity assay (FMCA) that has been used in drug development and for the development of personalized cancer medicine9–16.This study investigated whether ex vivo assessment of drug sensitivity by the FMCA provides predictive information in terms of peritoneal recurrence-free survival (PRFS) and overall survival (OS) in patients treated with CRS and HIPEC for isolated PMCRC.

  • Evaluating the utilisation of industrial excess heat from an energy systems perspective

    • Sweden aims to achieve climate neutrality by 2045. The need to immediately reduce greenhouse gas emissions in order to achieve climate targets affects industry directly. The pulp and paper sector is responsible for more than 50% of industrial energy use in Sweden. Increased energy efficiency is expected to contribute significantly to the reduction of primary energy use. The recovery and utilisation of industrial excess heat (IEH) has been identified as an important potential contribution to energy efficiency in industry. Previous research based on top-down studies has estimated the availability of IEH for entire sectors, and bottom-up results for many case studies are available. While top-down studies lack detailed information on the profile of the excess heat available, bottom-up studies have limited coverage. Detailed information about excess heat amounts and temperature levels is required for the assessment of the potential of the various heat recovery technologies that are available. The aim of this thesis is to present, in a series of steps, methods to systematically analyse an industrial process to obtain a detailed profile of the excess heat available under various process conditions, to aggregate results that can be generalised to whole industrial sectors, and to obtain IEH recovery potentials using different technologies. The assessment of the utilisation options for IEH recovery is complemented with an analysis of system aspects that could affect profitability and global greenhouse gas (GHG) emissions. An energy-targeting procedure combined with optimisation has been applied to six case studies of kraft pulp and paper mills in Sweden. This method obtained IEH profiles that were used in a regression analysis to estimate the IEH availability and electricity generation potentials from low and medium temperature IEH using organic Rankine cycles (ORC). A comparison of profitability and global GHG emissions between ORC electricity generation using IEH and small-scale combined heat and electricity (CHP) production is presented for three energy markets.The results show that there is a potential to increase electricity generation from low and medium temperature IEH by 7–9% in the kraft mills in Sweden, depending on the level of process integration considered. The utilisation of low and medium temperature IEH for electricity generation has the potential to reduce global GHG emissions in all the energy-market scenarios considered, but if biomass is considered a limited resource, district heating (DH) deliveries can achieve higher global GHG reductions. ORC electricity generation from low and medium temperature IEH is economically viable and showed overall better profitability and GHG emissions reductions than small-scale CHP using ORCs. The economic feasibility of ORC electricity generation is less affected by external conditions and uncertainties than direct DH deliveries.

    • The effects of Collembola grazing activities on the ectomycorrhizal symbiosis were studied in laboratory microcosms. Pinus contorta seedlings in association with Paxillus involutus were grown in a sandy soil and the Collembola Onychiurus armatus was added in different densities. To study effects on nutrient uptake by the extramatrical mycorrhizal mycelium, cups containing 15NH4+ and phytin amended soil were evenly distributed in the microcosms. These cups were covered with a net that allowed the mycelium to penetrate but neither Collembola nor plant roots. Extramatrical hyphal growth was impeded at a high density of O. Armatus. Low densities of O. armatus increased the extramatrical hyphal growth, the colonization rate of side plants and the biomass of P. involutus. However, the amount of P. involutus on/in the plant roots was not affected. Thus, low densities of collembolans induced a shift towards a larger proportion of P. involutus growing extramatrically. The presence of O. armatus in low numbers enhanced the uptake and transfer of 15N by P. involvus to the plants by up to 76%. The Collembola population growth was not higher in mycorrhizal compared with non-mycorrhizal microcosms while nematode population size was reduced in mycorrhizal compared with non-mycorrhizal microcosms.

  • Soil Resources Influence Spatial Patterns of Denitrifying Communities at Scales Compatible with Land Management

    • Knowing spatial patterns of functional microbial guilds can increase our understanding of the relationships between microbial community ecology and ecosystem functions. Using geostatistical modeling to map spatial patterns, we explored the distribution of the community structure, size, and activity of one functional group in N cycling, the denitrifiers, in relation to 23 soil parameters over a 44-ha farm divided into one organic and one integrated crop production system. The denitrifiers were targeted by the nirS and nirK genes that encode the two mutually exclusive types of nitrite reductases, the cd(1) heme-type and copper reductases, respectively. The spatial pattern of the denitrification activity genes was reflected by the maps of the abundances of nir genes. For the community structure, only the maps of the nirS community were related to the activity. The activity was correlated with nitrate and dissolved organic nitrogen and carbon, whereas the gene pools for denitrification, in terms of size and composition, were influenced by the soil structure. For the nirS community, pH and soil nutrients were also important in shaping the community. The only unique parameter related to the nirK community was the soil Cu content. However, the spatial pattern of the nirK denitrifiers corresponded to the division of the farm into the two cropping systems. The different community patterns, together with the spatial distribution of the nirS/nirK abundance ratio, suggest habitat selection on the nirS-and nirK-type denitrifiers. Our findings constitute a first step in identifying niches for denitrifiers at scales relevant to land management.

    • Aims and objectives: To explore newly graduated nurses' strategies for, and experiences of, sleep problems and fatigue when starting shiftwork. A more comprehensive insight into nurses' strategies, sleep problems, fatigue experiences and contributing factors is needed to understand what support should be provided. Background: For graduate nurses, the first years of practice are often stressful, with many reporting high levels of burnout symptoms. Usually, starting working as a nurse also means an introduction to shiftwork, which is related to sleep problems. Sleep problems may impair stress management and, at the same time, stress may cause sleep problems. Previously, sleep problems and fatigue have been associated with burnout, poor health and increased accident risk. Design and Methods: Semi-structured interviews were conducted with nurses (N = 11) from four different Swedish hospitals, and qualitative inductive content analysis was used. The study was approved by the Regional Ethical Review Board in Stockholm. The COREQ checklist was followed. Results: Many nurses lacked effective strategies for managing sleep and fatigue in relation to shiftwork. Various strategies were used, of which some might interfere with factors regulating and promoting sleep such as the homeostatic drive. Sleep problems were common during quick returns, often due to difficulties unwinding before sleep, and high workloads exacerbated the problems. The described consequences of fatigue in a clinical work context indicated impaired executive and nonexecutive cognitive function. Conclusion: The findings indicate that supporting strategies and behaviours for sleep and fatigue in an intervention for newly graduated nurses starting shiftwork may be of importance to improve well-being among nurses and increase patient safety. Relevance to clinical practice: This study highlights the importance of addressing sleep and fatigue issues in nursing education and work introduction programmes to increase patient safety and improve well-being among nurses.
