  • Comparing and visualizing titanium implant integration in rat bone using 2D and 3D techniques

    • The aim was to compare the osseointegration of grit-blasted implants with and without a hydrogen fluoride treatment in rat tibia and femur, and to visualize bone formation using state-of-the-art 3D visualization techniques. Grit-blasted implants were inserted in femur and tibia of 10 Sprague-Dawley rats (4 implants/rat). Four weeks after insertion, bone implant samples were retrieved. Selected samples were imaged in 3D using Synchrotron Radiation-based CT (SRCT). The 3D data was quantified and visualized using two novel visualization techniques, thread fly-through and 2D unfolding. All samples were processed to cut and ground sections and 2D histomorphometrical comparisons of bone implant contact (BIC), bone area (BA), and mirror image area (MI) were performed. BA values were statistically significantly higher for test implants than controls (p<0.05), but BIC and MI data did not differ significantly. Thus, the results partly indicate improved bone formation at blasted and hydrogen fluoride treated implants, compared to blasted implants. The 3D analysis was a valuable complement to 2D analysis, facilitating improved visualization. However, further studies are required to evaluate aspects of 3D quantitative techniques, with relation to light microscopy that traditionally is used for osseointegration studies.   • Improved Quantification of Bone Remodelling by Utilizing Fuzzy Based Segmentation

    • We present a novel fuzzy theory based method for the segmentation of images required in histomorphometrical investigations of bone implant integration. The suggested method combines discriminant analysis classification controlled by an introduced uncertainty measure, and fuzzy connectedness segmentation method, so that the former is used for automatic seeding of the later. A thorough evaluation of the proposed segmentation method is performed. Comparison with previously published automatically obtained measurements, as well as with manually obtained ones, is presented. The proposed method improves the segmentation and, consequently, the accuracy of the automatic measurements, while keeping advantages with respect to the manual ones, by being fast, repeatable, and objective.

    • In this paper we present an automatic method for measurements of bone area and bone-implant contact length around the implant in histological images. The method is then compared to the time consuming and subjective manual method performed in the light-microscope. Significant difference in the length values of the estimation of the bone-implant contact were observed, revealing greater numbers for the automatic method compared to the manual. These differences are most likely due to staining artifacts. However, the area measurements of the automatic method correspond well with the manual method. Furthermore, this paper also gives a brief description of the initial investigation of the use of tomography imaging methods to quantify bone remodeling around implants. The synchrotron radiation based microtomography technique (SRµCT) seems to be suitable for an subsequent analysis of bone tissue integration.

To automatize the quantification, Cuanto, asoftware for measurements of bone area and estimation ofbone-implant contact length in histological images of boneimplantsamples has been developed. The quantificationresult of the software is compared to manual measurements;area measurements correspond well with the manualquantification whereas significant differences in the lengthestimation is observed. The possibility of zooming in downto cell-level when quantifying manually is believed to renderthe discrepancy.
