    • The sensitive detection and quantification of DNA targets in the food industry and in environmental and clinical settings are issues of utmost importance in ensuring contamination-free food, monitoring the environment, and battling disease. Selective probes coupled with powerful amplification techniques are therefore of major interest. In this study, we set out to create an integrated microchemical chip that benefits from microfluidic chip technology in terms of sensitivity and a strong detection methodology provided jointly by padlock probes and rolling circle amplification (RCA). Here, we have integrated padlock probes and RCA into a microchip. The chip uses solid phase capture in a microchannel to enable washing cycles and decrease analytical area, and employs on-bead RCA for single-molecule amplification and detection. We investigated the effects of reagent concentration and amount of padlock probes, and demonstrated the feasibility of detecting Salmonella.

  • Visual acuity and the risk of cycling injuries : register-based cohort study from adolescence to middle-age

    • BACKGROUND: Cycling is increasingly encouraged in many countries as an inexpensive and healthy choice of transportation. Operating any vehicle on the road requires high visual acuity, but few studies to our knowledge have examined the association between vision and cycling injuries.METHODS: We examined whether poorer visual acuity is associated with increased risk of fatal and non-fatal cycling injuries. We used prospectively recorded register data for 691,402 men born between 1970 and 1992 in Sweden. We followed these men from an average age of 18 years, when visual acuity was assessed during the conscription assessment, to age 45 at the latest. We identified fatal and non-fatal cycling and car injuries using Patient and Cause of Death registers. Cox regression models were used to estimate hazard ratios (HR) and 95% confidence intervals (95%CI).RESULTS: Based on visual acuity for the eye with the best vision, moderately impaired acuity 0.9 to 0.6 when wearing refractive correction was associated with increased risk for cycling injuries (HR=1.44, 95%CI 1.16-1.79) compared with unimpaired vision (uncorrected visual acuity 1.0) and after adjustment for a wide range of potential confounders. This association remained consistent across various sensitivity analyses. Visual acuity was not associated with car injury risk.CONCLUSIONS: In this cohort study, poorer vision was specifically associated with a higher rate of cycling injuries.

    • In bandwidth abundant optical networks it is important to develop design strategies that are not only cost effective but also able to account for the inherent characteristics of the optical transmission medium responsible for the signal quality degradation. With this objective in mind, the paper proposes a design algorithm able to determine an appropriate placement of regenerators in Grouped Routing Entity (GRE)-based optical networks. The objective of the algorithm is to guarantee a certain signal quality level to all connections while minimizing the number of fibers and switching ports required in the network The proposed design strategy relies on a physical-layer impairment model, specifically introduced in the paper for this purpose, able to estimate the signal quality of an optical path in a GRE-based optical network Simulation results indicate that compared with a single layer optical path network the proposed design algorithm can reduce at least 87% of the switch ports while the increment in the number of fibers is lower than 10%.

    • The design and testing of different optical heads were performed to evaluate the pitch deviation of a diffrac-tion scale grating with a small diffraction angle. Based on the proposed pitch deviation evaluation method employing optical angle sensors based on laser au-tocollimation, a modified optical head with position -sensitive detectors (PSDs) is first designed and con-structed by following the conventional optical config-uration. Owing to the small angle of diffraction of the first-order diffracted beams, the modified optical head has a large working distance, resulting in poor sensor stability. Therefore, a novel and compact optical head employing a pair of small prisms is designed and de-veloped to shorten the working distance of the opti-cal head. An additional modification was also made to the developed compact optical head in such a way that collimator objectives (COs) in the laser autocollima-tion units are removed to improve the sensor sensitiv-ity. Experimental comparisons were conducted using the three types of optical heads to verify the feasibility of the developed optical angle sensor with PSDs.

    • This Swedish register-based cohort study examined whether asthma, hay fever and allergic dermatitis in late adolescence identified in the early 1970s are associated with antidepressant medication in middle age, between 2006 and 2009. After adjustment for childhood and adulthood sociodemographic characteristics, psychological, cognitive and physical function, and comorbidity, the magnitude of the associations diminished for asthma, while hay fever and atopic dermatitis retained associations. Hay fever and atopic dermatitis in adolescence have potentially important implications for future mental health, while asthma may already have influenced an individual's ability to cope with stress by late adolescence.

  • Lactobacilli as a Vector for Delivery of Nanobodies against Norovirus Infection

    • Passive administration of neutralizing antibodies (Abs) is an attractive strategy for the control of gastrointestinal infections. However, an unanswered practical concern is the need to assure the stability of sufficient amounts of orally administered neutralizing Abs against intestinal pathogens (e.g., norovirus) in the harsh environment of the gastrointestinal tract. To this end, we expressed a single-domain Ab (VHH, nanobody) against norovirus on the cell surface of Lactobacillus, a natural and beneficial commensal component of the gut microbiome. First, we used intestinal epithelial cells generated from human induced pluripotent stem cells to confirm that VHH 1E4 showed neutralizing activity against GII.17 norovirus. We then expressed VHH 1E4 as a cell-wall–anchored form in Lactobacillus paracasei BL23. Flow cytometry confirmed the expression of VHH 1E4 on the surface of lactobacilli, and L. paracasei that expressed VHH 1E4 inhibited the replication of GII.17 norovirus in vitro. We then orally administered VHH 1E4-expressing L. paracasei BL23 to germ-free BALB/c mice and confirmed the presence of lactobacilli with neutralizing activity in the intestine for at least 10 days after administration. Thus, cell-wall-anchored VHH-displaying lactobacilli are attractive oral nanobody deliver vectors for passive immunization against norovirus infection.

    • Polycyclic aromatic hydrocarbons (PAHs) contain less than or similar to 20% of the carbon in the interstellar medium. They are potentially produced in circumstellar environments (at temperatures greater than or similar to 1000 kelvin), by (similar to 10 kelvin) interstellar clouds, or by processing of carbon-rich dust grains. We report isotopic properties of PAHs extracted from samples of the asteroid Ryugu and the meteorite Murchison. The doubly-C-13 substituted compositions (Delta 2x(13)C values) of the PAHs naphthalene, fluoranthene, and pyrene are 9 to 51 parts per thousand higher than values expected for a stochastic distribution of isotopes. The Delta 2x(13)C values are higher than expected if the PAHs formed in a circumstellar environment, but consistent with formation in the interstellar medium. By contrast, the PAHs phenanthrene and anthracene in Ryugu samples have Delta 2x(13)C values consistent with formation by higher-temperature reactions.
