    • The Internet of Things (IoT) and the Internet of Services (IoS) are two well-knownexemplars of the emerging ‘Internet variants’. These variants will be tightlyinterwoven yet specific with respect to the supporting technologies needed. Thepresent paper discusses the five variants identified as essential by the authors: IoT,IoS, Internet-of-Humans, Internet-of-Crowds, and Internet-of-Clouds. For eachvariant, a non-comprehensive set of research challenges is cited and related to thestate of the art and to ongoing projects of the lab.

    • We undertook a meta-analysis of six Crohn's disease genome-wide association studies (GWAS) comprising 6,333 affected individuals (cases) and 15,056 controls and followed up the top association signals in 15,694 cases, 14,026 controls and 414 parent-offspring trios. We identified 30 new susceptibility loci meeting genome-wide significance (P < 5 × 10⁻⁸). A series of in silico analyses highlighted particular genes within these loci and, together with manual curation, implicated functionally interesting candidate genes including SMAD3, ERAP2, IL10, IL2RA, TYK2, FUT2, DNMT3A, DENND1B, BACH2 and TAGAP. Combined with previously confirmed loci, these results identify 71 distinct loci with genome-wide significant evidence for association with Crohn's disease.

  • MINDS. The Detection of 13 CO 2 with JWST-MIRI Indicates Abundant CO 2 in a Protoplanetary Disk

    • We present JWST-MIRI Medium Resolution Spectrometer (MRS) spectra of the protoplanetary disk around the low-mass T Tauri star GW Lup from the MIRI mid-INfrared Disk Survey Guaranteed Time Observations program. Emission from 12CO213CO2, H2O, HCN, C2H2, and OH is identified with 13CO2 being detected for the first time in a protoplanetary disk. We characterize the chemical and physical conditions in the inner few astronomical units of the GW Lup disk using these molecules as probes. The spectral resolution of JWST-MIRI MRS paired with high signal-to-noise data is essential to identify these species and determine their column densities and temperatures. The Q branches of these molecules, including those of hot bands, are particularly sensitive to temperature and column density. We find that the 12CO2 emission in the GW Lup disk is coming from optically thick emission at a temperature of ∼400 K. 13CO2 is optically thinner and based on a lower temperature of ∼325 K, and thus may be tracing deeper into the disk and/or a larger emitting radius than 12CO2. The derived N CO 2 / N H 2 O ratio is orders of magnitude higher than previously derived for GW Lup and other targets based on Spitzer-InfraRed-Spectrograph data. This high column density ratio may be due to an inner cavity with a radius in between the H2O and CO2 snowlines and/or an overall lower disk temperature. This paper demonstrates the unique ability of JWST to probe inner disk structures and chemistry through weak, previously unseen molecular features.

  • Optimization of sulfuric acid-assisted glycerol pretreatment of sugarcane bagasse

    • A combined pretreatment of sugarcane bagasse with glycerol and sulfuric acid was investigated based on a central composite rotatable experimental design. The following factors were varied: temperature (150–199°C), time (0.69–2.3 h), H2SO4 concentration (0.0–1.1%), and glycerol concentration (55.4–79.6%). Xylans and lignin were considerably solubilized during pretreatment. Xylan solubilization, ranging between 6% and 94%, increased significantly with the increase of temperature, time, and H2SO4 concentration and dropped with the increase of glycerol amount. Glycerol restricted the solubilization and full hydrolysis of xylans and the degradation of xylose. Lignin solubilization (20.6–49.4%) increased with the increase of all the experimental factors. Cellulose recovery, which was generally high, increased with the increasing of glycerol concentration and declined at high levels of the other factors. Recoveries above 97% were achieved at low H2SO4 concentration and high glycerol load, whereas the lowest value (83.4%) was achieved in the longest-lasting experiment. The models based on the experimental results predicted the maximal lignin solubilization at 187.7°C, 2.3 h, 79.6% glycerol, and 0.64% H2SO4, whereas the highest yield of enzymatic hydrolysis can be expected at 194.1°C, 1.67 h, 79.6% glycerol, and 1.1% H2SO4. The optimal conditions were confirmed in control experiments. The synergistic effect of sulfuric acid and glycerol on the enzymatic hydrolysis of cellulose was demonstrated.

  • MINDS. JWST/MIRI Reveals a Dynamic Gas-rich Inner Disk inside the Cavity of SY Cha

    • SY Cha is a T Tauri star surrounded by a protoplanetary disk with a large cavity seen in the millimeter continuum but has the spectral energy distribution of a full disk. Here we report the first results from JWST/Mid-InfraRed Instrument (MIRI) Medium Resolution Spectrometer (MRS) observations taken as part of the MIRI mid-INfrared Disk Survey (MINDS) GTO Program. The much improved resolution and sensitivity of MIRI-MRS compared to Spitzer enables a robust analysis of the previously detected H2O, CO, HCN, and CO2 emission as well as a marginal detection of C2H2. We also report the first robust detection of mid-infrared OH and rovibrational CO emission in this source. The derived molecular column densities reveal the inner disk of SY Cha to be rich in both oxygen- and carbon-bearing molecules. This is in contrast to PDS 70, another protoplanetary disk with a large cavity observed with JWST, which displays much weaker line emission. In the SY Cha disk, the continuum, and potentially the line, flux varies substantially between the new JWST observations and archival Spitzer observations, indicative of a highly dynamic inner disk.

    • Obesity is globally prevalent and highly heritable, but its underlying genetic factors remain largely elusive. To identify genetic loci for obesity susceptibility, we examined associations between body mass index and ~2.8 million SNPs in up to 123,865 individuals with targeted follow up of 42 SNPs in up to 125,931 additional individuals. We confirmed 14 known obesity susceptibility loci and identified 18 new loci associated with body mass index (P < 5 × 10−8), one of which includes a copy number variant near GPRC5B. Some loci (at MC4R, POMC, SH2B1 and BDNF) map near key hypothalamic regulators of energy balance, and one of these loci is near GIPR, an incretin receptor. Furthermore, genes in other newly associated loci may provide new insights into human body weight regulation.

  • Plasma HDL cholesterol and risk of myocardial infarction : a mendelian randomisation study

    • BACKGROUND: High plasma HDL cholesterol is associated with reduced risk of myocardial infarction, but whether this association is causal is unclear. Exploiting the fact that genotypes are randomly assigned at meiosis, are independent of non-genetic confounding, and are unmodified by disease processes, mendelian randomisation can be used to test the hypothesis that the association of a plasma biomarker with disease is causal.METHODS: We performed two mendelian randomisation analyses. First, we used as an instrument a single nucleotide polymorphism (SNP) in the endothelial lipase gene (LIPG Asn396Ser) and tested this SNP in 20 studies (20,913 myocardial infarction cases, 95,407 controls). Second, we used as an instrument a genetic score consisting of 14 common SNPs that exclusively associate with HDL cholesterol and tested this score in up to 12,482 cases of myocardial infarction and 41,331 controls. As a positive control, we also tested a genetic score of 13 common SNPs exclusively associated with LDL cholesterol.FINDINGS: Carriers of the LIPG 396Ser allele (2·6% frequency) had higher HDL cholesterol (0·14 mmol/L higher, p=8×10(-13)) but similar levels of other lipid and non-lipid risk factors for myocardial infarction compared with non-carriers. This difference in HDL cholesterol is expected to decrease risk of myocardial infarction by 13% (odds ratio [OR] 0·87, 95% CI 0·84-0·91). However, we noted that the 396Ser allele was not associated with risk of myocardial infarction (OR 0·99, 95% CI 0·88-1·11, p=0·85). From observational epidemiology, an increase of 1 SD in HDL cholesterol was associated with reduced risk of myocardial infarction (OR 0·62, 95% CI 0·58-0·66). However, a 1 SD increase in HDL cholesterol due to genetic score was not associated with risk of myocardial infarction (OR 0·93, 95% CI 0·68-1·26, p=0·63). For LDL cholesterol, the estimate from observational epidemiology (a 1 SD increase in LDL cholesterol associated with OR 1·54, 95% CI 1·45-1·63) was concordant with that from genetic score (OR 2·13, 95% CI 1·69-2·69, p=2×10(-10)).INTERPRETATION: Some genetic mechanisms that raise plasma HDL cholesterol do not seem to lower risk of myocardial infarction. These data challenge the concept that raising of plasma HDL cholesterol will uniformly translate into reductions in risk of myocardial infarction.

  • The Mid-infrared Instrument for JWST and Its In-flight Performance

    • The Mid-Infrared Instrument (MIRI) extends the reach of the James Webb Space Telescope (JWST) to 28.5 μm. It provides subarcsecond-resolution imaging, high sensitivity coronagraphy, and spectroscopy at resolutions of λ/Δλ ∼ 100-3500, with the high-resolution mode employing an integral field unit to provide spatial data cubes. The resulting broad suite of capabilities will enable huge advances in studies over this wavelength range. This overview describes the history of acquiring this capability for JWST. It discusses the basic attributes of the instrument optics, the detector arrays, and the cryocooler that keeps everything at approximately 7 K. It gives a short description of the data pipeline and of the instrument performance demonstrated during JWST commissioning. The bottom line is that the telescope and MIRI are both operating to the standards set by pre-launch predictions, and all of the MIRI capabilities are operating at, or even a bit better than, the level that had been expected. The paper is also designed to act as a roadmap to more detailed papers on different aspects of MIRI.
