  • Aspen pectate lyase PtxtPL1-27 mobilizes matrix polysaccharides from woody tissues and improves saccharification yield

    • Background: Wood cell walls are rich in cellulose, hemicellulose and lignin. Hence, they are important sources of renewable biomass for producing energy and green chemicals. However, extracting desired constituents from wood efficiently poses significant challenges because these polymers are highly cross-linked in cell walls and are not easily accessible to enzymes and chemicals. Results: We show that aspen pectate lyase PL1-27, which degrades homogalacturonan and is expressed at the onset of secondary wall formation, can increase the solubility of wood matrix polysaccharides. Overexpression of this enzyme in aspen increased solubility of not only pectins but also xylans and other hemicelluloses, indicating that homogalacturonan limits the solubility of major wood cell wall components. Enzymatic saccharification of wood obtained from PL1-27-overexpressing trees gave higher yields of pentoses and hexoses than similar treatment of wood from wild-type trees, even after acid pretreatment. Conclusions: Thus, the modification of pectins may constitute an important biotechnological target for improved wood processing despite their low abundance in woody biomass.

  • Structural and Physical Effects of Carbon Nanofillers in Thermoplastic and Thermosetting Polymer Systems

    • Ever since the discovery of carbon nano materials like carbon nanotube (CNT) and graphene, this class of materials has gained significant attention due to their exotic properties. The principle idea of my present research project is to understand the novel improvements induced in polymer matrices with inclusion of the nanofillers. This thesis is thematically divided into three parts. In the first part we introduce principle materials that we use for preparation of composites. Methods of nanofiller preparation and different nanocomposites as previously reported in literature are discussed to formulate the basis of our study. Different dispersion techniques are discussed which facilitate uniform nanofiller distribution. A variety of experimental methods are described which were employed to investigate the structure and properties of the composites. In the second part we discuss in details polyamide-12 (PA12) composites using CNT and graphene as fillers. A marked improvement is recorded in the toughness of the films with incorporation of CNT, dispersed in PA12 using a surfactant. Electrical percolation is also achieved in the otherwise insulating matrix. With PA-12 fibers we explored the effect of fiber processing and CNT incorporation in the mechanical properties. Extensive wide angle x-ray diffraction was carried out to interpret the structural modifications brought about by CNT in the matrix. The final part of the thesis deals with a thermosetting polymer, epoxy composites. CNT, Graphene and also a mixture of the two nanofillers were used as reinforcing agents. Appreciable improvement was recorded in the mechanical properties, electrical and thermal conductivity of the composites. Detailed optical and electron microscopy was carried out to get a vivid idea of the micro-structure and dispersion. The presented work demonstrates the significant ability of carbon nanofillers to reinforce polymer matrices enhancing their mechanical, electrical and thermal properties and opening a wide horizon for a variety of applications.

  • Suppression of xylan endotransglycosylase PtxtXyn10A affects cellulose microfibril angle in secondary wall in aspen wood

    • Certain xylanases from family GH10 are highly expressed during secondary wall deposition, but their function is unknown. We carried out functional analyses of the secondary-wall specific PtxtXyn10A in hybrid aspen (Populus tremulaxtremuloides).PtxtXyn10A function was analysed by expression studies, overexpression in Arabidopsis protoplasts and by downregulation in aspen.PtxtXyn10A overexpression in Arabidopsis protoplasts resulted in increased xylan endotransglycosylation rather than hydrolysis. In aspen, the enzyme was found to be proteolytically processed to a 68kDa peptide and residing in cell walls. Its downregulation resulted in a corresponding decrease in xylan endotransglycosylase activity and no change in xylanase activity. This did not alter xylan molecular weight or its branching pattern but affected the cellulose-microfibril angle in wood fibres, increased primary growth (stem elongation, leaf formation and enlargement) and reduced the tendency to form tension wood. Transcriptomes of transgenic plants showed downregulation of tension wood related genes and changes in stress-responsive genes. The data indicate that PtxtXyn10A acts as a xylan endotransglycosylase and its main function is to release tensional stresses arising during secondary wall deposition. Furthermore, they suggest that regulation of stresses in secondary walls plays a vital role in plant development.

  • Structure of cellulose and microcrystalline cellulose from various wood species, cotton and flax studied by X-ray scattering

    • The structure of microcrystalline cellulose (MCC) made by mild acid hydrolysis from cotton linter, flax fibres and sulphite or kraft cooked wood pulp was studied and compared with the structure of the starting materials. Crystallinities and the length and the width of the cellulose crystallites were determined by wide-angle X-ray scattering and the packing and the cross-sectional shape of the microfibrils were determined by small-angle X-ray scattering. The morphological differences were studied by scanning electron microscopy. A model for the changes in microfibrillar structure between native materials, pulp and MCC samples was proposed. The results indicated that from softwood or hardwood pulp, flax cellulose and cotton linter MCC with very similar nanostructures were obtained with small changes in reaction conditions. The crystallinity of MCC samples was 54-65%. The width and the length of the cellulose crystallites increased when MCC was made. For example, between cotton and cotton MCC the width increased from 7.1 nm to 8.8 nm and the length increased from 17.7 nm to 30.4 nm. However, the longest crystallites were found in native spruce wood (35-36 nm).

    • The impact of drying on the structure of the never-dried hardwood cell wall was studied at nanometer level by means of wide- and small-angle X-ray scattering (WAXS, SAXS), and at micrometer level by X-ray microtomography (mu CT). Never-dried silver birch, European aspen and hybrid aspen samples were measured by WAXS in situ during drying in air. The samples included juvenile and mature wood, as well as normal and tension wood to allow comparison of the effects of different matrix compositions and microfibril angles. The deformations of cellulose crystallites and amorphous components of the cell wall were detected as changes in the cellulose reflections 200 and 004 and amorphous halo in the WAXS patterns. Especially, the width of the reflection 004, corresponding to the cellulose chain direction, increased due to drying in all the samples, indicating an increase of strain and disorder of the chains. Also, the cellulose unit cell shrank 0.2-0.3% during drying in this direction in all the samples except in hybrid aspen tension wood. According to the SAXS results of silver birch, the distance between microfibrils decreased during drying. It was detected by mu CT that the mean cross-sectional maximum width of the parenchymatous rays decreased from that of never-dried to air-dried birch by roughly 16%.

    • A bifunctional protein composed of a highly negatively charged oyster shell protein and a chitin-binding domain enabled the formation of biohybrid materials through non-covalent surface modification of chitin nanofibres. The results demonstrate that specific biomolecular interactions offer a route for the formation of biosynthetic materials.

    • The role of xylan as a limiting factor in the enzymatic hydrolysis of cellulose was studied by hydrolysing nanocellulose samples prepared by mechanical fibrillation of birch pulp with varying xylan content. Analyzing the nanocelluloses and their hydrolysis residues with dynamic FT-IR spectroscopy revealed that a certain fraction of xylan remained tightly attached to cellulose fibrils despite partial hydrolysis of xylan with xylanase prior to pulp fibrillation and that this fraction remained in the structure during the hydrolysis of nanocellulose with cellulase mixture as well. Thus, a loosely bound fraction of xylan was predicted to have been more likely removed by purified xylanase. The presence of loosely bound xylan seemed to limit the hydrolysis of crystalline cellulose, indicated by an increase in cellulose crystallinity and by preserved crystal width measured with wide-angle X-ray scattering. Removing loosely bound xylan led to a proportional hydrolysis of xylan and cellulose with the cellulase mixture.

    • This dissertation presents new work and results in the flow of complex fluids and experimental methodologies for their investigation. Plate-like colloidal particles of kaolinite and nickel hydroxide are studied. A study of lamellar fragments and their mixture with the nickel hydroxide particles is also presented. The lamellar fragments are self-assembled structures of surfactant molecules that approximate disks. Particles are seen to align with their large faces parallel to the flow direction under shear and elongational strains. Order parameters have been calculated to quantify the extent of preferential alignment and direction of orientation. The experimental data are supported by comparisons with finite-element fluid mechanics calculations that provide estimates of the flow patterns and the strain rates. Elongational strain rates in the range of 5 − 20 s−1 are required to induce a high degree of alignment with the various sizes of the particles whereas about two to three order of magnitude higher shear strain rates are required. The combination of both elongational and shear strain is an effective means to provide a uniform alignment. Comparison of the Peclet numbers calculated for both the shear and elongational flow are presented and this explains that alignment occurs when the energy per particle of the strain is larger than the thermal energy. Mixtures have shown complex behavior: significant changes in the structure are observed that are not seen to the same extent in samples at rest. X-ray diffraction and small-angle neutron scattering techniques are used to characterize the samples and determine the structure in flowing systems. Laboratory X-ray diffraction can be used to characterize dispersed samples. The combination of dynamic light scattering and X-ray diffraction was used to estimate the thickness of the stabilizing layers of the polymer on the colloidal particles. Scattering of synchrotron radiation and neutrons are powerful complementary techniques to provide information about flow and the potential to apply them to systems that are beyond the scope of simple simulations has been demonstrated.

  • Binding of Levosimendan, a Calcium Sensitizer, to Cardiac Troponin C

    • Levosimendan is an inodilatory drug that mediates its cardiac effect by the calcium sensitization of contractile proteins. The target protein of levosimendan is cardiac troponin C (cTnC). In the current work, we have studied the interaction of levosimendan with Ca2+-saturated cTnC by heteronuclear NMR and small angle x-ray scattering. A specific interaction between levosimendan and the Ca2+-loaded regulatory domain of recombinant cTnCC35S was observed. The changes in the NMR spectra of the N-domain of full-length cTnCC35S, due to the binding of levosimendan to the primary site, were indicative of a slow conformational exchange. In contrast, no binding of levosimendan to the regulatory domain of cTnCA-Cys, where all the cysteine residues are mutated to serine, was detected. Moreover, it was shown that levosimendan was in fast exchange on the NMR time scale with a secondary binding site in the C-domain of both cTnCC35S and cTnCA-Cys. The small angle x-ray scattering experiments confirm the binding of levosimendan to Ca2+-saturated cTnC but show no domain-domain closure. The experiments were run in the absence of the reducing agent dithiothreitol and the preservative sodium azide (NaN 3), since we found that levosimendan reacts with these chemicals, commonly used for preparation of NMR protein samples.

  • Structure of oak wood from the Swedish warship Vasa revealed by X-ray scattering and microtomography

    • The degradation of oak wood of the historical warship Vasa was studied, focusing on cellular structure by X-ray microtomography (mu CT) and on the nanostructure of the cell wall by wide- and small-angle X-ray scattering (WAXS, SAXS). Solid samples [polyethylene glycol (PEG)-, impregnated and PEG-extracted] were submitted to X-ray analysis and the results compared to those of recent oak. The cellular structure of the Vasa oak was surprisingly well preserved at the micrometer level, according to the mu CT images. As revealed by WAXS, the fraction of crystalline cellulose was lower in the Vasa samples compared with recent oak, but the average length and width of cellulose crystallites (25 +/- 2 nm and 3.0 +/- 0.1 nm, respectively), and the mean microfibril angles (4-9 degrees), showed no significant differences. Accordingly, the crystalline parts of cellulose microfibrils are well preserved in the Vasa oak. The SAXS results indicated a declined short-range order between the cellulose microfibrils and a higher porosity of the Vasa oak compared with recent oak, which may be explained by modification of the hemicellulose-lignin matrix.
