  • Guidelines for the use and interpretation of assays for monitoring autophagy

    • In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field.

  • Prevalence and influencing factors of wheeze and asthma among preschool children in Urumqi city : a cross-sectional survey

    • To investigate the prevalence and indoor environmental influencing factors of wheeze and asthma among preschool children in Urumqi, Xinjiang, China to provide a strong basis for prevention and control. In August 2019, a cross-sectional study involving 8153 preschool children was conducted in 60 kindergartens in Urumqi. The ALLHOME-2 questionnaire was used for childhood wheeze and asthma survey, and the dampness in buildings and health (DBH) questionnaire was used for the childhood home dwelling and living environment survey. Multivariate unconditional logistic regression was then used to analyze the potential influencing factors of childhood asthma and wheeze. The prevalence of wheeze and asthma in children was 4.7% and 2.0%, respectively. Multivariate unconditional logistic regression results suggested that ethnicity other than the Han Chinese (odds ratio (OR) 1.39, 95% confidence interval (CI) 1.05-1.84), caesarean section (OR 1.24, 95% CI 1.00-1.53), family history of asthma (OR 5.00, 95% CI 3.36-7.44), carpet or floor bedding at home (OR 1.40, 95% CI 1.05-1.87), purchasing new furniture in the mother's residence during pregnancy (OR 1.58, 95% CI 1.06-2.36), pet keeping in the residence at aged 0-1 year (OR 1.55, 95% CI 1.13-2.13), passive smoking by child in the current residence (OR 1.35, 95% CI 1.01-1.80), and having mould or hygroma in the child's residence at aged 0-1 year (OR 1.72, 95% CI 1.12-2.64) were risk factors for wheeze. In addition, Girls (OR 0.73, 95% CI 0.59-0.90) was a protective factor for wheeze. Caesarean section (OR 1.46, 95% CI 1.06-2.00), family history of asthma (OR 7.06, 95% CI 4.33-11.53), carpet or floor bedding at home (OR 2.20, 95% CI 1.50-3.23), and pet keeping in the residence at aged 0-1 year (OR 1.64, 95% CI 1.04-1.83) were risk factors for asthma, whereas Girls (OR 0.58, 95% CI 0.42-0.80) was a protective factor for asthma. This survey indicates that the purchase of new furniture, the placement of carpet or floor bedding in the child's residence, the pets keeping, room dampness or moldy phenomena, and passive smoking may all contribute to an elevated risk of wheeze or asthma in children.

  • Longitudinal patient-reported outcomes 1 year after thoracoscopic segmentectomy versus lobectomy for early-stage lung cancer: a multicentre, prospective cohort study protocol

    • Segmentectomy and lobectomy are the main surgical procedures for early-stage lung cancer. However, few studies have analysed patient-reported outcomes after segmentectomy versus lobectomy. This study aims to compare patient-reported outcomes—such as symptoms, daily functioning and quality of life—between thoracoscopic segmentectomy and lobectomy for early-stage lung cancer during the 1 year after surgery.Methods and analysisOverall, 788 newly diagnosed patients with early-stage lung cancer (tumour size ≤2 cm), who are scheduled to undergo thoracoscopic segmentectomy or lobectomy, will be recruited in this multicentre, prospective cohort study. The patients will receive standardised care after surgery. The Perioperative Symptom Assessment for Lung Surgery—a validated lung cancer surgery-specific scale—will be used to assess the symptoms and functions at baseline, at discharge and monthly after discharge for 1 year. The European Organization for Research and Treatment of Cancer Quality of Life Questionnaire Core 30 and Lung Cancer module 29 will be used to assess the patients’ quality of life at the same time points. The primary outcome will be the shortness of breath scores during the first year after thoracoscopic segmentectomy and lobectomy and will be compared using mixed-effects models. The secondary outcomes will include other symptoms, indicators of daily functioning, quality of life scores and traditional clinical outcomes. These will be compared using mixed-effects models and the Student’s t-test, non-parametric test or Χ2test. Propensity score matching will be used to ensure an even distribution of known confounders between the groups.Ethics and disseminationThe Ethics Committee for Medical Research and New Medical Technology of Sichuan Cancer Hospital approved this study (approval number: SCCHEC-02-2022-002). All participants will be instructed to provide informed consent. The manuscript is based on protocol version 3.0. The study results will be presented at medical conferences and published in peer-reviewed journals.Trial registration numberChiCTR2200060753.

    • Neuronal toxic pollutants in environment possess hazards to human health. It is essential to determine the causative neuronal toxicants in environmental samples. In the present study, viability of primary cultured cerebellar granule neurons (CGNs), combined with sample extraction, chemical fractionation and identification, was applied for screening acid-resistant neuronal toxic substances in environmental samples. River sediments and agricultural soils along the river near a brominated flame retardant (BFR) manufacturing plant in South China were collected to screen the key neuronal toxicants. The results indicated that the manufacturing plant was a source of neuronal toxicity risks. In the sediment and soil near the plant, one of the causative toxicants was identified as tris-(2,3-dibromopropyl) isocyanurate (TBC) using HPLC-MS/MS. In addition, an unknown chemical possibly causing significant neuronal toxicity was isolated from all the soil samples in the region.

  • Air monitoring of polychlorinated biphenyls, polybrominated diphenyl ethers and organochlorine pesticides in West Antarctica during 2011-2017 : Concentrations, temporal trends and potential sources

    • Annual air samples were collected at various sites in the Fildes Peninsula, West Antarctica from December 2010 to January 2018 using XAD-2 resin passive air samplers to investigate concentrations, temporal trends and potential sources of persistent organic pollutants (POPs) in Antarctic air. Relatively low concentrations of polychlorinated biphenyls (PCBs) (Σ19PCBs: 1.5-29.7 pg/m3), polybrominated diphenyl ethers (PBDEs) (Σ12PBDEs: 0.2-2.9 pg/m3) and organochlorine pesticides (OCPs) (Σ13OCPs: 101-278 pg/m3) were found in the atmosphere of West Antarctica. PCB-11, BDE-47 and hexachlorobenzene (HCB) were the predominant compounds in the atmosphere. The concentrations of PCBs, HCHs, DDTs and endosulfans were found to show decreasing temporal trends, whereas uniform temporal trends were observed for HCB. The atmospheric half-life values for PCBs, HCHs, DDTs and endosulfans in Antarctic air were estimated for the first time, using regressions of the natural logarithm of the concentrations versus the number of years, obtaining the values of 2.0, 2.0, 2.4 and 1.2 year, respectively. An increasing ratio of α-HCH/γ-HCH indicated long residence time for α-HCH and possible transformation of γ-HCH to α-HCH in the atmosphere. The ratios of p,p'-DDT/p,p'-DDE were mostly lower than unity in this study, which could be attributed to aged sources. It was found that long-range atmospheric transport was still considered to be the main contributing factor to the atmospheric levels of the POPs in West Antarctica whereas the contribution of human activities at the Chinese Great Wall Station was minor. The results of this study give a view on the most recent temporal trends and provide new insights regarding the occurrence of various POPs in the Antarctic atmosphere.

  • Observations of Kelvin-Helmholtz Waves in the Earth's Magnetotail Near the Lunar Orbit

    • Kelvin‐Helmholtz waves (KHWs), which have been widely observed at the magnetopause in the region near the Earth, play an essential role in the transport of solar wind plasma and energy into the magnetosphere under dominantly northward interplanetary magnetic field (IMF) conditions. In this study, we present simultaneous observations of KHWs under the northward IMF observed by both the Acceleration, Reconnection, Turbulence, and Electrodynamics of Moon's Interaction with the Sun (ARTEMIS) spacecraft in the Earth's magnetotail around the lunar orbit (at X ~ −50RE, Y ~ 30RE, dusk side) and the Geotail in the near‐Earth space (at X ~ −5RE, Y ~ −10RE, dawn side). The KHWs are quantitatively characterized by their dominant period, phase velocity, and wavelength, utilizing wavelet analysis and an approximation of their center‐of‐mass velocity. Our results suggest that the phase velocity and spatial scale of KHWs may increase as they propagate along the boundary layer toward the tail. Alternatively, the differences between the ARTEMIS and Geotail observations may indicate the possibility of dawn‐dusk asymmetry in the excited KHWs in this study. Our results strongly evidence the existence of the development of KHWs in terms of their wave frequency and scale size in the magnetotail and provide insight to the time evolution of KHWs along the magnetopause.

  • Characterization of a trichome-specific promoter of the aldehyde dehydrogenase 1 (ALDH1) gene in Artemisia annua

    • Artemisinin is a frequently used anti-malaria drug extracted from glandular trichomes (GSTs) in Artemisia annua L. In this study, we report on the characterization of the promoter of aldehyde dehydrogenase 1 (ALDH1) involved in the biosynthesis of artemisinin. A 1620-bp promoter fragment was cloned upstream of the ALDH1 start codon. Putative regulatory cis-acting elements are predicted by software, revealing that this gene is affected by complex factors. The activity of the ALDH1 promoter was analyzed using a reporter gene GUS. GUS expression showed a spatial difference in leaves at different ages. In young leaves, GUS staining was exclusively discovered in GSTs. In older leaves, both GSTs and T-shaped trichomes (TSTs) showed GUS signals. Only TSTs showed GUS staining in lower leaves. No GUS staining was detected in the bottom leaves. The result demonstrates that the ALDH1 promoter is trichome-specific. The RT-Q-PCR analysis revealed that both wild-type and recombinant promoters showed similar activity in A. annua. After application of exogenous 100 μM methyl jasmonate, 100 μM gibberellin and 100 μM salicylic acid separately, the transcript levels were increased significantly, indicating that ALDH1 may play an important role in the response to hormones in A. annua.

  • Transpolar arc observation after solar wind entry into the high-latitude magnetosphere

    • Recently, Cluster observations have revealed the presence of new regions of solar wind plasma entry at the high-latitude magnetospheric lobes tailward of the cusp region, mostly during periods of northward interplanetary magnetic field. In this study, observations from the Global Ultraviolet Imager (GUVI) experiment on board the TIMED spacecraft and Wideband Imaging Camera imager on board the IMAGE satellite are used to investigate a possible link between solar wind entry and the formation of transpolar arcs in the polar cap. We focus on a case when transpolar arc formation was observed twice right after the two solar wind entry events were detected by the Cluster spacecraft. In addition, GUVI and IMAGE observations show a simultaneous occurrence of auroral activity at low and high latitudes after the second entry event, possibly indicating a two-part structure of the continuous band of the transpolar arc.
