    • Despite intense interest, methods that provide enhanced sensitivity and specificity in parallel measurements of candidate protein biomarkers in numerous samples have been lacking. We present herein a multiplex proximity ligation assay with readout via realtime PCR or DNA sequencing (ProteinSeq). We demonstrate improved sensitivity over conventional sandwich assays for simultaneous analysis of sets of 35 proteins in 5 μl of blood plasma. Importantly, we observe a minimal tendency to increased background with multiplexing, compared to a sandwich assay, suggesting that higher levels of multiplexing are possible. We used ProteinSeq to analyze proteins in plasma samples from cardiovascular disease (CVD) patient cohorts and matched controls. Three proteins, namely P-selectin, Cystatin-B and Kallikrein-6, were identified as putative diagnostic biomarkers for CVD. The latter two have not been previously reported in the literature and their potential roles must be validated in larger patient cohorts. We conclude that ProteinSeq is promising for screening large numbers of proteins and samples while the technology can provide a much-needed platform for validation of diagnostic markers in biobank samples and in clinical use.

  • Subsequent Event Risk in Individuals With Established Coronary Heart Disease : Design and Rationale of the GENIUS-CHD Consortium

    • BACKGROUND: The Genetics of Subsequent Coronary Heart Disease (GENIUS-CHD) consortium was established to facilitate discovery and validation of genetic variants and biomarkers for risk of subsequent CHD events, in individuals with established CHD.METHODS: The consortium currently includes 57 studies from 18 countries, recruiting 185 614 participants with either acute coronary syndrome, stable CHD, or a mixture of both at baseline. All studies collected biological samples and followed-up study participants prospectively for subsequent events.RESULTS: Enrollment into the individual studies took place between 1985 to present day with a duration of follow-up ranging from 9 months to 15 years. Within each study, participants with CHD are predominantly of self-reported European descent (38%-100%), mostly male (44%-91%) with mean ages at recruitment ranging from 40 to 75 years. Initial feasibility analyses, using a federated analysis approach, yielded expected associations between age (hazard ratio, 1.15; 95% CI, 1.14-1.16) per 5-year increase, male sex (hazard ratio, 1.17; 95% CI, 1.13-1.21) and smoking (hazard ratio, 1.43; 95% CI, 1.35-1.51) with risk of subsequent CHD death or myocardial infarction and differing associations with other individual and composite cardiovascular endpoints.CONCLUSIONS: GENIUS-CHD is a global collaboration seeking to elucidate genetic and nongenetic determinants of subsequent event risk in individuals with established CHD, to improve residual risk prediction and identify novel drug targets for secondary prevention. Initial analyses demonstrate the feasibility and reliability of a federated analysis approach. The consortium now plans to initiate and test novel hypotheses as well as supporting replication and validation analyses for other investigators.

  • Reduced intensity conditioning increases risk of severe cGVHD : identification of risk factors for cGVHD in a multicenter setting

    • Chronic graft-versus-host disease (cGVHD) remains a major cause of morbidity and mortality after allogeneic hematopoietic stem cell transplantation (HSCT). Aim is to identify risk factors for the development of cGVHD in a multicenter setting. Patients transplanted between 2000 and 2006 were analyzed (n = 820). Donors were HLA-identical siblings (57%), matched unrelated donors (30%), and HLA-A, B or DR antigen mismatched (13%). Reduced intensity conditioning (RIC) was given to 65% of patients. Overall incidence of cGVHD was 46% for patients surviving more than 100 days after HSCT (n = 747). Older patient age [HR 1.15, p < 0.001], prior acute GVHD [1.30, p = 0.024], and RIC [1.36, p = 0.028] increased overall cGVHD. In addition, RIC [4.85, p < 0.001], prior aGVHD [2.14, p = 0.001] and female donor to male recipient [1.80, p = 0.008] increased the risk of severe cGVHD. ATG had a protective effect for both overall [0.41, p < 0.001] and severe cGVHD [0.20, p < 0.001]. Relapse-free survival (RFS) was impaired in patients with severe cGVHD. RIC, prior aGVHD, and female-to-male donation increase the risk of severe cGVHD. ATG reduces the risk of all grades of cGVHD without hampering RFS. GVHD prophylaxis may be tailored according to the risk profile of patients.

    • Although transplant practices have changed over the last decades, no information is available on trends in incidence and outcome of chronic graft-versus-host disease (cGVHD) over time. This study used the central database of the Center for International Blood and Marrow Transplant Research (CIBMTR) to describe time trends for cGVHD incidence, nonrelapse mortality, and risk factors for cGVHD. The 12-year period was divided into 3 intervals, 1995 to 1999, 2000 to 2003, and 2004 to 2007, and included 26,563 patients with acute leukemia, chronic myeloid leukemia, and myelodysplastic syndrome. Multivariate analysis showed an increased incidence of cGVHD in more recent years (odds ratio = 1.19, P < .0001), and this trend was still seen when adjusting for donor type, graft type, or conditioning intensity. In patients with cGVHD, nonrelapse mortality has decreased over time, but at 5 years there were no significant differences among different time periods. Risk factors for cGVHD were in line with previous studies. This is the first comprehensive characterization of the trends in cGVHD incidence and underscores the mounting need for addressing this major late complication of transplantation in future research.

  • Large-Scale Gene-Centric Meta-analysis across 32 Studies Identifies Multiple Lipid Loci

    • Genome-wide association studies (GWASs) have identified many SNPs underlying variations in plasma-lipid levels. We explore whether additional loci associated with plasma-lipid phenotypes, such as high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), total cholesterol (TC), and triglycerides (TGs), can be identified by a dense gene-centric approach. Our meta-analysis of 32 studies in 66,240 individuals of European ancestry was based on the custom similar to 50,000 SNP genotyping array (the ITMAT-Broad-CARe array) covering similar to 2,000 candidate genes. SNP-lipid associations were replicated either in a cohort comprising an additional 24,736 samples or within the Global Lipid Genetic Consortium. We identified four, six, ten, and four unreported SNPs in established lipid genes for HDL-C, LDL-C, TC, and TGs, respectively. We also identified several lipid-related SNPs in previously unreported genes: DGAT2, HCAR2, GPIHBP1, PPARG, and FTO for HDL-C; SOCS3, APOH, SPTY2D1, BRCA2, and VLDLR for LDL-C; SOCS3, UGT1A1, BRCA2, UBE3B, FCGR2A, CHUK, and INSIG2 for TC; and SERPINF2, C4B, GCK, GATA4, INSR, and LPAL2 for TGs. The proportion of explained phenotypic variance in the subset of studies providing individual-level data was 9.9% for HDL-C, 9.5% for LDL-C, 10.3% for TC, and 8.0% for TGs. This large meta-analysis of lipid phenotypes with the use of a dense gene-centric approach identified multiple SNPs not previously described in established lipid genes and several previously unknown loci. The explained phenotypic variance from this approach was comparable to that from a meta-analysis of GWAS data, suggesting that a focused genotyping approach can further increase the understanding of heritability of plasma lipids.

    • Möjligheter att åstadkomma elenergibesparingar inom skogsindustrin genom modifiering av luftade dammar till anaerob-aerob drift har studerats i laboratorieskala. I tre parallella system behandlades avloppsvatten från tillverkning av såväl oblekt som blekt sulfatmassa samt avloppsvatten från CTMP-tillverkning. I samtliga 7 genomförda försök var den totala uppehållstiden 7 dygn. Behandlingen genomfördes vid rumstemperatur (20-22ºC) efter pH-justering, men utan tillsats av närsalter. Inverkan av avloppsvattnets uppehållstid under anaeroba betingelser liksom inverkan av slamrecirkulering undersöktes. Uppehållstiden i det anaeroba steget var härvid antingen 2,3 eller 4,6 dygn, dvs antingen 33 eller 66% av dammens totalvolym utgjorde ett anaerobt behandlingssteg. Ett kontrollförsök drevs under betingelser motsvarande de som upprätthålls sommartid i konventionella luftade dammar, dvs med hela volymen luftad. Försöksresultaten visar att för avloppsvatten från såväl oblekt som blekt sulfatmassa kan samma eller bättre behandlingsresultat erhållas vid övergång från helt anaerob till kombinerad anaerob-aerob drift

The output voltage from the generator fluctuates both inamplitude and frequency and must therefore be converted to enablegrid connection. In order to study the voltage conversion, themeasure station was fitted with a six pulse diode rectifierand a capacitive filter during the autumn of 2006. The WEC is based on a permanent magnet linear generator located on the sea floor driven by a point absorber. A measuring station has been installed on a nearby island where all measurements and experiments on the WEC have been carried out. The output voltage from the generator fluctuates both in amplitude and frequency and must therefore be converted to enable grid connection. In order to study the voltage conversion, the measuring station was fitted with a six pulse diode rectifier and a capacitive filter during the autumn of 2006. The object of this paper is to present a detailed description of the Lysekil research site. Special attention will be given to the power absorption by the generator when it is connected to a nonlinear load.

  • Downstream passage of diadromous fish in Swedish rivers

    • Downstream passage problems for diadromous fish are rarely addressed in regulated rivers in northern Europe. Some attempts to decrease the mortality among downstream moving fish are now tested in southern Sweden, ranging from the use of trash gates totesting of low-sloped fine-spaced racks. This presentation describes the efficiency of twosuch measures at hydroelectric facilities in the River Ätran. The targeted species were Atlantic salmon, anadromous brown trout and European eel. Smolt (N=66, 2007), kelt (N=20, 2007) and silver eels (N=170, 2007-2008) were radio-tagged and tracked inpassing the facilities.An open trash gate proved to have a very low efficiency for smolt (7%) and most individuals passed through the racks and turbines. The efficiency was intermediate for kelt (40%) and several individuals died on the trash racks or remained upstream until theend of the study. The route seeking time was limited for smolts, but substantial for kelts. Since no control groups were used the extent of the delays could not be quantified. The overall passage success was 50% for kelts and 85% for smolts, illustrating the need for a more efficient measure for kelts in particular.The passage for silvers eels of the two lower most hydroelectric plants (HEPs) in the River Ätran revealed a 28% passage success at HEP 2 (N=50), with a 23,5 m head and Francis turbines. The corresponding success for the control group released downstream ofthe HEP was 92% (N=50). The following year the old rack (20 mm / 43°) was replaced by a new rack (18 mm / 35°) with exits at the surface. With the new rack in place 80% of the eels released upstream were successfully diverted and caught, resulting in a totalsuccess of 90% (compared to 28% with the old rack). The success at HEP 1 was 76% (N=58), but no individuals entered the open trash gate. The success rate was somewhat uncertain because of the short distance to sea and the possibility for injured (but not dead) individuals to reach sea but not surviving in the long run
