    • Uterine leiomyomata (UL) are the most common tumours of the female genital tract and the primary cause of surgical removal of the uterus. Genetic factors contribute to UL susceptibility. To add understanding to the heritable genetic risk factors, we conduct a genome-wide association study (GWAS) of UL in up to 426,558 European women from FinnGen and a previous UL meta-GWAS. In addition to the 50 known UL loci, we identify 22 loci that have not been associated with UL in prior studies. UL-associated loci harbour genes enriched for development, growth, and cellular senescence. Of particular interest are the smooth muscle cell differentiation and proliferation-regulating genes functioning on the myocardin-cyclin dependent kinase inhibitor 1A pathway. Our results further suggest that genetic predisposition to increased fat-free mass may be causally related to higher UL risk, underscoring the involvement of altered muscle tissue biology in UL pathophysiology. Overall, our findings add to the understanding of the genetic pathways underlying UL, which may aid in developing novel therapeutics.

    • Air pollution has been suggested to be associated with depression. However, current evidence is conflicting, and no study has considered different sources of ambient particulate matter with an aerodynamic diameter below 2.5 µm (PM2.5). We evaluated the associations of long-term exposure to PM2.5 from road traffic and residential wood combustion with the prevalence of depression in the Helsinki region, Finland.MethodsWe conducted a cross-sectional analysis based on the Helsinki Capital Region Environmental Health Survey 2015–2016 (N=5895). Modelled long-term outdoor concentrations of PM2.5 were evaluated using high-resolution emission and dispersion modelling on an urban scale and linked to the home addresses of study participants. The outcome was self-reported doctor-diagnosed or treated depression. We applied logistic regression and calculated the OR for 1 μg/m3 increase in PM2.5, with 95% CI. Models were adjusted for potential confounders, including traffic noise and urban green space.ResultsOf the participants, 377 reported to have been diagnosed or treated for depression by a doctor. Long-term exposure to PM2.5 from road traffic (OR=1.23, 95% CI 0.86 to 1.73; n=5895) or residential wood combustion (OR=0.78, 95% CI 0.43 to 1.41; n=5895) was not associated with the prevalence of depression. The estimates for PM2.5 from road traffic were elevated, but statistically non-significant, for non-smokers (OR=1.38, 95% CI 0.94 to 2.01; n=4716).ConclusionsWe found no convincing evidence of an effect of long-term exposure to PM2.5 from road traffic or residential wood combustion on depression.

  • Options-Based Security-Oriented Framework for Addressing Uncerainty Issues in IT Security

    • Continuous development and innovation in Information Technology introduces novel configuration methods, software development tools and hardware components. This steady state of flux is very desirable as it improves productivity and the overall quality of life in societies. However, the same phenomenon also gives rise to unseen threats, vulnerabilities and security concerns that are becoming more critical with the passage of time. As an implication, technological progress strongly impacts organizations’ existing information security methods, policies and techniques, making obsolete existing security measures and mandating reevaluation, which results in an uncertain IT infrastructure. In order to address these critical concerns, an options-based reasoning borrowed from corporate finance is proposed and adapted for evaluation of security architecture and decision- making to handle them at organizational level. Options theory has provided significant guidance for uncertainty management in several domains, such as Oil & Gas, government R&D and IT security investment projects. We have applied options valuation technique in a different context to formalize optimal solutions in uncertain situations for three specific and identified uncertainty issues in IT security. In the research process, we formulated an adaptation model for expressing options theory in terms useful for IT security which provided knowledge to formulate and propose a framework for addressing uncertainty issues in information security. To validate the efficacy of this proposed framework, we have applied this approach to the SHS (Spridnings- och Hämtningssystem) and ESAM (E-Society) systems used in Sweden. As an ultimate objective of this research, we intend to develop a solution that is amenable to automation for the three main problem areas caused by technological uncertainty in information security: i) dynamically changing security requirements, ii) externalities caused by a security system, iii) obsoleteness of evaluation. The framework is general and capable of dealing with other uncertainty management issues and their solutions, but in this work we primarily deal with the three aforementioned uncertainty problems. The thesis presents an in-depth background and analysis study for a proposed options-based security-oriented framework with case studies for SHS and ESAM systems. It has also been assured that the framework formulation follows the guidelines from industry best practices criteria/metrics. We have also proposed how the whole process can be automated as the next step in development.

  • Phage Biocontrol of Pseudomonas aeruginosa in Water

    • Bacteriophage control of harmful or pathogenic bacteria has aroused growing interest, largely due to the rise of antibiotic resistance. The objective of this study was to test phages as potential agents for the biocontrol of an opportunistic pathogen Pseudomonas aeruginosa in water. Two P. aeruginosa bacteriophages (vB_PaeM_V523 and vB_PaeM_V524) were isolated from wastewater and characterized physically and functionally. Genomic and morphological characterization showed that both were myoviruses within the Pbunavirus genus. Both had a similar latent period (50-55 min) and burst size (124-134 PFU/infected cell), whereas there was variation in the host range. In addition to these environmental phages, a commercial Pseudomonas phage, JG003 (DSM 19870), was also used in the biocontrol experiments. The biocontrol potential of the three phages in water was tested separately and together as a cocktail against two P. aeruginosa strains; PAO1 and the environmental strain 17V1507. With PAO1, all phages initially reduced the numbers of the bacterial host, with phage V523 being the most efficient (>2.4 log(10) reduction). For the environmental P. aeruginosa strain (17V1507), only the phage JG003 caused a reduction (1.2 log(10)) compared to the control. The cocktail of three phages showed a slightly higher decrease in the level of the hosts compared to the use of individual phages. Although no synergistic effect was observed in the host reduction with the use of the phage cocktail, the cocktail-treated hosts did not appear to acquire resistance as rapidly as hosts treated with a single phage. The results of this study provide a significant step in the development of bacteriophage preparations for the control of pathogens and harmful microbes in water environments.
