  • Global dataset of soil organic carbon in tidal marshes

    • Tidal marshes store large amounts of organic carbon in their soils. Field data quantifying soil organic carbon (SOC) stocks provide an important resource for researchers, natural resource managers, and policy-makers working towards the protection, restoration, and valuation of these ecosystems. We collated a global dataset of tidal marsh soil organic carbon (MarSOC) from 99 studies that includes location, soil depth, site name, dry bulk density, SOC, and/or soil organic matter (SOM). The MarSOC dataset includes 17,454 data points from 2,329 unique locations, and 29 countries. We generated a general transfer function for the conversion of SOM to SOC. Using this data we estimated a median (± median absolute deviation) value of 79.2 ± 38.1 Mg SOC ha−1 in the top 30 cm and 231 ± 134 Mg SOC ha−1 in the top 1 m of tidal marsh soils globally. This data can serve as a basis for future work, and may contribute to incorporation of tidal marsh ecosystems into climate change mitigation and adaptation strategies and policies.

  • Adipose lipid turnover and long-term changes in body weight

    • The worldwide obesity epidemic(1) makes it important to understand how lipid turnover (the capacity to store and remove lipids) regulates adipose tissue mass. Cross-sectional studies have shown that excess body fat is associated with decreased adipose lipid removal rates(2,3). Whether lipid turnover is constant over the life span or changes during long-term weight increase or loss is unknown. We determined the turnover of fat cell lipids in adults followed for up to 16 years, by measuring the incorporation of nuclear bomb test-derived C-14 in adipose tissue triglycerides. Lipid removal rate decreases during aging, with a failure to reciprocally adjust the rate of lipid uptake resulting in weight gain. Substantial weight loss is not driven by changes in lipid removal but by the rate of lipid uptake in adipose tissue. Furthermore, individuals with a low baseline lipid removal rate are more likely to remain weight-stable after weight loss. Therefore, lipid turnover adaptation might be important for maintaining pronounced weight loss. Together these findings identify adipose lipid turnover as an important factor for the long-term development of overweight/obesity and weight loss maintenance in humans.

  • The Age of Olfactory Bulb Neurons in Humans

    • Continuous turnover of neurons in the olfactory bulb is implicated in several key aspects of olfaction. There is a dramatic decline postnatally in the number of migratory neuroblasts en route to the olfactory bulb in humans, and it has been unclear to what extent the small number of neuroblasts at later stages contributes new neurons to the olfactory bulb. We have assessed the age of olfactory bulb neurons in humans by measuring the levels of nuclear bomb test-derived C-14 in genomic DNA. We report that C-14 concentrations correspond to the atmospheric levels at the time of birth of the individuals, establishing that there is very limited, if any, postnatal neurogenesis in the human olfactory bulb. This identifies a fundamental difference in the plasticity of the human brain compared to other mammals.

  • The search for disks or planetary objects around directly imaged companions : a candidate around DH Tauri B

    • Context. In recent decades, thousands of substellar companions have been discovered with both indirect and direct methods of detection. While the majority of the sample is populated by objects discovered using radial velocity and transit techniques, an increasing number have been directly imaged. These planets and brown dwarfs are extraordinary sources of information that help in rounding out our understanding of planetary systems.Aims. In this paper, we focus our attention on substellar companions detected with the latter technique, with the primary goal of investigating their close surroundings and looking for additional companions and satellites, as well as disks and rings. Any such discovery would shed light on many unresolved questions, particularly with regard to their possible formation mechanisms.Methods. To reveal bound features of directly imaged companions, whether for point-like or extended sources, we need to suppress the contribution from the source itself. Therefore, we developed a method based on the negative fake companion technique that first estimates the position in the field of view (FoV) and the flux of the imaged companion with high precision, then subtracts a rescaled model point spread function (PSF) from the imaged companion, using either an image of the central star or another PSF in the FoV. Next it performs techniques, such as angular differential imaging, to further remove quasi-static patterns of the star (i.e., speckle contaminants) that affect the residuals of close-in companions.Results. After testing our tools on simulated companions and disks and on systems that were chosen ad hoc, we applied the method to the sample of substellar objects observed with SPHERE during the SHINE GTO survey. Among the 27 planets and brown dwarfs we analyzed, most objects did not show remarkable features, which was as expected, with the possible exception of a point source close to DH Tau B. This candidate companion was detected in four different SPHERE observations, with an estimated mass of ~1MJup, and a mass ratio with respect to the brown dwarf of 1∕10. This binary system, if confirmed, would be the first of its kind, opening up interesting questions for the formation mechanism, evolution, and frequency of such pairs. In order to address the latter, the residuals and contrasts reached for 25 companions in the sample of substellar objects observed with SPHERE were derived. If the DH Tau Bb companion is real, the binary fraction obtained is ~7%, which is in good agreement with the results obtained for field brown dwarfs.Conclusions. While there may currently be many limitations affecting the exploration of bound features to directly imaged exoplanets and brown dwarfs, next-generation instruments from the ground and space (i.e., JWST, ELT, and LUVOIR) will be able to image fainter objects and, thus, drive the application of this technique in upcoming searches for exo-moons and circumplanetary disks.

    • Stem cells generate neurons in discrete regions in the postnatal mammalian brain. However, the extent of neurogenesis in the adult human brain has been difficult to establish. We have taken advantage of the integration of (14)C, generated by nuclear bomb tests during the Cold War, in DNA to establish the age of neurons in the major areas of the human cerebral neocortex. Together with the analysis of the neocortex from patients who received BrdU, which integrates in the DNA of dividing cells, our results demonstrate that, whereas nonneuronal cells turn over, neurons in the human cerebral neocortex are not generated in adulthood at detectable levels but are generated perinatally.

  • Flow Cytometry of Mouse and Human Adipocytes for the Analysis of Browning and Cellular Heterogeneity

    • Adipocytes, once considered simple lipid-storing cells, are rapidly emerging as complex cells with many biologically diverse functions. A powerful high-throughput method for analyzing single cells is flow cytometry. Several groups have attempted to analyze and sort freshly isolated adipocytes; however, using an adipocyte-specific reporter mouse, we demonstrate that these studies fail to detect the majority of white adipocytes. We define critical settings required for adipocyte flow cytometry and provide a rigid strategy for analyzing and sorting white and brown adipocyte populations. The applicability of our protocol is shown by sorting mouse adipocytes based on size or UCP1 expression and demonstrating that a subset of human adipocytes lacks the beta(2)-adrenergic receptor, particularly in the insulin-resistant state. In conclusion, the present study confers key technological insights for analyzing and sorting mature adipocytes, opening up numerous downstream research applications.
