    • In this paper we present a systematic study of the morphology and composition of supported lipid bilayers (SLBs) formed by vesicle fusion using a wide variety of surface sensitive techniques that give information about the lateral as well as vertical structure and bilayer fluidity. SLBs of 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (POPC) and 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC) mixtures at five different bulk vesicle compositions were formed in such a way that the phase separation boundaries were crossed. For all compositions studied, the SLBs were systematically enriched with POPC compared to the nominal vesicle composition. Nevertheless, gel-fluid domain coexistence was observed for SLB compositions in which phase separation was expected based on the bulk phase diagram. The probable causes for the compositional difference in the SLBs are discussed in terms of the phase behaviour of the mixture and its effect on the membrane formation process by vesicle fusion.

  • Agreement between Family Members and the Physicians View in the ICU Environment: Personal Experience as a Factor Influencing Attitudes towards Corresponding Hypothetical Situations

    • Background: It is not known whether intensive care unit (ICU) patients family members realistically assess patients health status. Objectives: The aim was to investigate the agreement between family and intensivists assessment concerning changes in patient health, focusing on family members resilience and their perceptions of decision making. Methods: For each ICU patient, withdrawal criteria were assessed by intensivists while family members assessed the patients health development and completed the Connor-Davidson Resilience Scale and the Self-Compassion Scale. Six months after ICU discharge, follow-up contact was established, and family members gave their responses to two hypothetical scenarios. Results: 162 ICU patients and 189 family members were recruited. Intensivists decisions about whether a patient met the withdrawal criteria had 75,9% accuracy for prediction of survival. Families assessments were statistically independent of intensivists opinions, and resilience had a significant positive effect on the probability of agreement with intensivists. Six months after discharge, family members whose relatives were still alive were significantly more likely to consider that the family or patient themselves should be involved in decision-making. Conclusions: Resilience is related to an enhanced probability of agreement of the family with intensivists perceptions of patients health progression. Family attitudes in hypothetical scenarios were found to be significantly affected by the patients actual health progression.

  • Molecular basis for SNX-BAR-mediated assembly of distinct endosomal sorting tubules

    • Sorting nexins (SNXs) are regulators of endosomal sorting. For the SNX-BAR subgroup, a Bin/Amphiphysin/Rvs (BAR) domain is vital for formation/stabilization of tubular subdomains that mediate cargo recycling. Here, by analysing the in vitro membrane remodelling properties of all 12 human SNX-BARs, we report that some, but not all, can elicit the formation of tubules with diameters that resemble sorting tubules observed in cells. We reveal that SNX-BARs display a restricted pattern of BAR domain-mediated dimerization, and by resolving a 2.8 angstrom structure of a SNX1-BAR domain homodimer, establish that dimerization is achieved in part through neutralization of charged residues in the hydrophobic BAR-dimerization interface. Membrane remodelling also requires functional amphipathic helices, predicted to be present in all SNX-BARs, and the formation of high order SNX-BAR oligomers through selective 'tip-loop' interactions. Overall, the restricted and selective nature of these interactions provide a molecular explanation for how distinct SNX-BAR-decorated tubules are nucleated from the same endosomal vacuole, as observed in living cells. Our data provide insight into the molecular mechanism that generates and organizes the tubular endosomal network.
