  • Impact of protease and lipase activities on quality of Swedish raw milk

    • An increasing demand for dairy products with long shelf-life is foreseen. A limiting factor for maintaining high quality of these products is the activity of spoilage enzymes during storage. Lipase and protease activities in Swedish raw milk at farm and dairy level were investigated, analysing milk samples from three geographical regions and two seasons. Lipase activity in milk at farm level was affected by regional and seasonal variations, whereas at dairy level only season had an effect. Lipase activity was positively correlated with ionic calcium. For protease activity, no effect of either region or season was seen. Degradation products, e.g., free fatty acids, peptides and plasmin proteolysis products, varied differently between season and geographical origin at both farm and dairy level. The results indicate that lipase and protease activities are important for raw milk quality, while ionic calcium might be a future indicator for milk fat stability.

  • The collection of narratives on strategies, ethics and public response of COVID-19 national vaccination process written by members of scholarly association Navigating Knowledge Landscapes Network

    • The data collected in this dataset are narratives exploring public policies, attitudes, individual behaviors, and the collective experiences of the involved communities regarding vaccination during the COVID-19 pandemic. The narratives were written by the members of the interdisciplinary research network Navigating Knowledge Landscapes (NKL; http://knowledge-landscapes.hiim.hr/). The members of the network are scholars belonging to different research disciplines and the aim of the network is to explore and discuss the individual aspects of citizens’ navigation of (new) knowledge in the digital society. An invitation to participate in this study was sent to members of the network on May 5, 2021. First responses were received on May 13, 2021, while the last response (including updated versions) was received on July 21, 2021. In total, 25 contributions were received from 27 scholars representing 17 countries (some scholars co-authored their contributions). Each of them described strategies, ethics, and public response of COVID-19 national vaccination process in their country.

  • Sleep Satisfaction May Modify the Association between Metabolic Syndrome and BMI, Respectively, and Occupational Stress in Japanese Office Workers

    • The association between obesity and psychological stress is ambiguous. The aim is to investigate the association between metabolic syndrome (MetS) and body mass index (BMI), respectively, with occupational stress among Japanese office workers. The study is a secondary analysis of the intervention group from a randomized controlled trial. There are 167 participants included in the analysis. Occupational stress is self-reported using the Brief Job Stress Questionnaire (BJSQ). BMI and the classification of MetS/pre-MetS was based on the participants’ annual health check-up data. The primary exposure is divided into three groups: no MetS, pre-MetS, and MetS in accordance with Japanese guidelines. The secondary exposure, BMI, remains as a continuous variable. Multiple linear regression is implemented. Sensitivity analyses are stratified by sleep satisfaction. Pre-MetS is significantly associated with occupational stress (7.84 points; 95% CI: 0.17, 15.51). Among participants with low sleep satisfaction, pre-MetS (14.09 points; 95% CI: 1.71, 26.48), MetS (14.72 points; 95% CI: 0.93, 28.51), and BMI (2.54 points; 95% CI: 0.05, 4.99) are all significantly associated with occupational stress. No significant associations are observed in participants with high sleep satisfaction. The findings of this study indicate that sleep satisfaction may modify the association between MetS and BMI, respectively, and occupational stress.

    • The role of exosomes in cancer can be inferred from the observation that they transfer tumor cell derived genetic material and signalling proteins, resulting in e.g. increased tumor angiogenesis and metastasis. However, the membrane transport mechanisms and the signalling events involved in the uptake of these virus-like particles remain ill-defined. We now report that internalization of exosomes derived from glioblastoma (GBM) cells involves nonclassical, lipid raft-dependent endocytosis. Importantly, we show that the lipid raft associated protein caveolin-1 (CAV1), in analogy with its previously described role in virus uptake, negatively regulates the uptake of exosomes. We find that exosomes induce the phosphorylation of several downstream targets known to associate with lipid rafts as signalling and sorting platforms, such as extracellular signal-regulated kinase-1/2 (ERK1/2) and heat shock protein 27 (HSP27). Interestingly, exosome uptake appears dependent on unperturbed ERK1/2-HSP27 signalling, and ERK1/2 phosphorylation is under negative influence by CAV1 during internalization of exosomes. These findings significantly advance our general understanding of exosome-mediated uptake and offer potential strategies for how this pathway may be targeted through modulation of CAV1 expression and ERK1/2 signaling.

  • Det nya hastighetsgränssystemet : regionala beslutsfattares syn, problembilder och förväntningar

    • Under 2008 och 2009 inför Vägverket nya hastighetsgränser baserade på ett mer flexibelt tiostegsystem. Det innebär att dagens hastighetsgränser kompletteras med 40, 60, 80, 100 och 120 km/tim. Hastigheten ska successivt anpassas efter hur säker vägen är och den grad av våld som människokroppen tål vid ett misstag, till exempel en avåkning. Syftet med föreliggande studie är att ge en samlad beskrivning och analys av regionala aktörers syn på det nya hastighetsgränssystemet, dvs. deras problembilder, förväntningar på vad systemet ska kunna åstadkomma samt syn på hur införandeprocessen hittills har genomförts. De förändringar som har behandlats i studien är de som ingår i etapp 1 av införandet av det nya systemet, dvs. förändringar av hastighetsgränser på det nationella stamvägnätet. Studiens data har samlats in genom intervjuer med representanter för remissinstanserna, dvs. kommuner, regionförbund/regionala samverkansorgan, länspolismyndigheter och länsstyrelser. För att få en spridning av intervjuerna över landet genomfördes, i samråd med Vägverket, intervjuerna i tre olika regioner: Skåne, Västra Götaland och Västerbotten. I varje region genomfördes ca sju intervjuer och totalt består materialet således av 21 genomförda intervjuer. Förutom intervjumaterialet har också samtliga skriftliga remissyttranden som lämnades in i de tre regionerna i samband med etapp 1 använts som material i studien. Resultaten visar att det finns en betydande ovana bland respondenterna att hantera och diskutera omprövningar av gällande hastighetsgränser, vilket medförde svårigheter för respondenterna att bilda sig en uppfattning om syftet med omprövningen. Denna problematik sammanhänger med intervjupersonernas uppfattning att det nya systemet drivs igenom utan en tydlig koppling till åtgärdsplaneringen. En klar majoritet av de som har intervjuats anser inte att det är motiverat att sänka hastighetsgränserna för att öka trafiksäkerheten. Framkomlighet och regional utveckling ska prioriteras genom att förbättra vägnätets standard och höja hastighetsgränserna. För kommuner och regionala samverkansorgan är frågan om hastighetsgränser en politisk fråga beroende på de negativa konsekvenserna för framkomlighet, pendling och regional utveckling som sänkningar av hastighetsgränser anses leda till. Därför efterlyses en bredare förankringsprocess för kommande etapper än den som tillämpades vid etapp 1. Därmed skulle möjligheterna öka för kommuner, regionförbund och andra aktörer att påverka besluten i en riktning som är i linje med de egna intressena om hög framkomlighet och regional utveckling.

  • Long-term glycemic variability and the risk of mortality in diabetic patients receiving peritoneal dialysis.

    • The large amount of glucose in the dialysate used in peritoneal dialysis (PD) likely affects the glycemic control. The aim of this study was to investigate the association between HbA1c variability, as a measure of long-term glycemic variability, and the risk of all-cause mortality in diabetic patients with PD.325 patients with diabetes and ESRD were followed (2008-2018) in the Swedish Renal Registry. Patients were separated in seven groups according to level of HbA1c variability. The group with the lowest variability was denoted the reference. The ratio of the standard deviation (SD) to the mean of HbA1c, HbA1c (SD)/HbA1c (mean), i.e. the coefficient of variation (CV), was defined as HbA1c variability. Hazard ratios (HR) and 95% confidence intervals (CI) were examined using Cox regression analyses.During follow-up, 170 (52%) deaths occurred. The highest mortality was among patients with the second highest HbA1c variability, CV≥2.83 [n = 44 of which 68% patients died]. In the multivariate analyses where lowest HbA1c variability (CV≤0.51) was used as the reference group, HbA1c CV 2.83-4.60 (HR 3.15, 95% CI 1.78-5.55; p<0.001) and CV> 4.6 (HR 2.48, 95% CI 1.21-5.11; p = 0.014) were associated with increased risk of death.The high risk of all-cause mortality in patients with diabetes and PD increased significantly with elevated HbA1c variability, as measure of long-term glycemic control. This indicates that stable glycemia is associated with an improvement of survival; whereas more severe glycemic fluctuations, possibly caused by radical changes in dialysis regimes or peritonitis, are associated with a higher risk of mortality in diabetic patients with PD.

    • Knowledge about association between glycated hemoglobin (HbA1c) and risk of all-cause mortality in patients with diabetes mellitus on maintenance hemodialysis (HD)-treatment is sparse. The study aims to investigate association between HbA1c and all-cause mortality in patients with diabetes and maintenance HD-treatment, separately for two age groups- above and below 75years.2487 patients (mean age 66years, 66% men) were separated in two age groups: ≤75years (n=1810) and>75years (n=677) and followed up between 2008 and 2018. Hazard ratios (HR) and 95% confidence intervals (CI) for associations between HbA1c and all-cause mortality were calculated using Cox-regression-models.1295 (52%) patients died and 473 (70%) among the patients above 75years old. In the multivariate analysis, HbA1c5-6% was used as reference. In patients≤75years old, only increased HbA1c>9.7%, HR2.03(CI1.43-2.89) was associated with increased risk of all-cause mortality. In patients>75years, HbA1c≤5%, HR1.67(CI1.16-2.40); HbA1c6.9-7.8%, HR1.41(CI1.03-1.93) and HbA1c8.7-9.7%, HR1.79 (CI1.08-2.96) were associated with increased risk of all-cause mortality.We found a J-shaped association between HbA1c and mortality only in diabetic HD-patients>75years. This probably indicates that in an old population of diabetic HD-patients, both intensive glucose control and hyperglycemia could be harmful and associated with higher risk of death.

  • Sex-dependent role of microglia in disulfide high mobility group box 1 protein-mediated mechanical hypersensitivity

    • High mobility group box 1 protein (HMGB1) is increasingly regarded as an important player in the spinal regulation of chronic pain. Although it has been reported that HMGB1 induces spinal glial activation in a Toll-like receptor (TLR)4-dependent fashion, the aspect of sexual dimorphisms has not been thoroughly addressed. Here, we examined whether the action of TLR4-activating, partially reduced disulfide HMGB1 on microglia induces nociceptive behaviors in a sex-dependent manner. We found disulfide HMGB1 to equally increase microglial Iba1 immunoreactivity in lumbar spinal dorsal horn in male and female mice, but evoke higher cytokine and chemokine expression in primary microglial culture derived from males compared to females. Interestingly, TLR4 ablation in myeloid-derived cells, which include microglia, only protected male mice from developing HMGB1-induced mechanical hypersensitivity. Spinal administration of the glial inhibitor, minocycline, with disulfide HMGB1 also prevented pain-like behavior in male mice. To further explore sex difference, we examined the global spinal protein expression using liquid chromatography-mass spectrometry and found several antinociceptive and anti-inflammatory proteins to be upregulated in only male mice subjected to minocycline. One of the proteins elevated, alpha-1-antitrypsin, partially protected males but not females from developing HMGB1-induced pain. Targeting downstream proteins of alpha-1-antitrypsin failed to produce robust sex differences in pain-like behavior, suggesting that several proteins identified by liquid chromatography-mass spectrometry are required to modulate the effects. Taken together, the current study highlights the importance of mapping sex dimorphisms in pain mechanisms and point to processes potentially involved in the spinal antinociceptive effect of microglial inhibition in male mice.
