    • New strategies for prevention and treatment of type 2 diabetes (T2D) require improved insight into disease etiology. We analyzed 386,731 common single-nucleotide polymorphisms (SNPs) in 1464 patients with T2D and 1467 matched controls, each characterized for measures of glucose metabolism, lipids, obesity, and blood pressure. With collaborators (FUSION and WTCCC/UKT2D), we identified and confirmed three loci associated with T2D - in a noncoding region near CDKN2A and CDKN2B, in an intron of IGF2BP2, and an intron of CDKAL1 - and replicated associations near HHEX and in SLC30A8 found by a recent whole-genome association study. We identified and confirmed association of a SNP in an intron of glucokinase regulatory protein (GCKR) with serum triglycerides. The discovery of associated variants in unsuspected genes and outside coding regions illustrates the ability of genome-wide association studies to provide potentially important clues to the pathogenesis of common diseases.

  • The Development and Evaluation of an Animated Video for Pre- and Postoperative Instructions for Patients with Osteoarthritis : A Design Science Research Approach

    • Osteoarthritis (OA) is a condition in the hip or knee joints that develops during a long period of time and sometimes needs hip or knee joint replacement surgery when pain gets too intense for the patient. This paper describes how an animated video for pre- and postoperative instructions for patients with osteoarthritis was designed. The design science research (DSR) approach was followed by creating a web-based animated video. The web-based animated video is used to support surgical departments with education for patients suffering from OA. In the web-based animated video, information about OA surgical treatment and its pre- and post-arrangements was included. The relevance, the rigor, and the design cycles were focused on, with some iterations of and improvements in the animations. Even after implementation, there was a feedback-loop with comments from the surgeons and their patients. Moreover, as more departments will use the web-based animated video, they want to make their special mark on it, so that further changes will be made. This paper presents the design and successful implementation of an animated video for pre- and postoperative instructions for patients with osteoarthritis, tightly linked to the patient journey and the workflow of healthcare professionals. The animated video serves not only as a tool to improve care but also as a basis for further scientific research studies.

    • In Sweden, opinions have long been divided on the meaning of the agency concept of authority and its basis. Due to a number of judgments from the Swedish Supreme Court, the discussion has been given renewed relevance. The dissertation investigates the question of the origin of authority. By using a legal dogmatic method, relevant sources are identified and analyzed in order to determine the content of current law.The dissertation first describes the development of authority in Swedish law and the purpose and function of authority rules. For there to be an act of authorization, it is required that the person who performs a legal act does so in the name of another. However, the exact meaning of acting in the name of another has proved to be far from obvious. The concept is central to the act of authorization, which is why it is also defined within the framework of this dissertation.The concept of declaration of intent, which plays a central role in the creation of authority, is also discussed. This is because there are divided opinions as to whether there are any subjective requirements on the principal or recipient for a declaration of intent to be considered to exist.Finally, the grounds for authority that have been identified in the dissertation are presented by analyzing the potential grounds identified within the framework of the dissertation.

  • Double-bunches for two-color soft X-ray free-electron laser at the MAX IV Laboratory

    • The ability to generate two-color free-electron laser (FEL) radiation enables a wider range of user experiments than just single-color FEL radiation. There are different schemes for generating the two colors, the original being to use a single bunch and two sets of undulators with different K-parameters. An alternative scheme was recently shown, where two separate bunches in the same RF bucket are used for lasing at different wavelengths in a single set of undulators. We here investigate the feasibility of accelerating and compressing a double-bunch time structure generated in the photocathode electron gun for subsequent use in a soft X-ray FEL at the MAX IV Laboratory.

    • Beam-driven plasma-wakefield acceleration is an acceleration scheme promising accelerating fields of at least two to three orders of magnitude higher than in conventional radiofrequency accelerating structures. The scheme relies on using a charged particle bunch (driver) to drive a non-linear plasma wake, into which a second bunch (witness) can be injected at an appropriate distance behind the first, yielding a substantial energy gain of the witness bunch particles. This puts very special demands on the machine providing the particle beam. In this article, we use simulations to show that, if driver-witness-bunches can be generated in the photocathode electron gun, the MAX IV Linear Accelerator could be used for plasma-wakefield acceleration.

    • Existing techniques for injecting probes into running code are limited: they either fail to support probing arbitrary locations, or to support scalable, rapid toggling of probes. We introduce a new technique on x86-64, called instruction punning, which allows scalable probes at any instruction. The key idea is that when we inject a jump instruction, the relative address of the jump serves simultaneously as data and as an instruction sequence. We show that this approach achieves probe invocation overheads of only a few dozen cycles, as well as low probe activation/deactivation costs, even when all threads in the system are both invoking probes and toggling them. Unlike competing systems, the latent overhead when probes are deactivated is exactly zero.

  • Serology assessment of antibody response to SARS-CoV-2 in patients with COVID-19 by rapid IgM/IgG antibody test

    • The coronavirus disease 2019 (COVID-19) pandemic has created a global health- and economic crisis. Detection of antibodies to severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) which causes COVID-19 by serological methods is important to diagnose a current or resolved infection. In this study, we applied a rapid COVID-19 IgM/IgG antibody test and performed serology assessment of antibody response to SARS-CoV-2. In PCR-confirmed COVID-19 patients (n = 45), the total antibody detection rate is 92% in hospitalized patients and 79% in non-hospitalized patients. The total IgM and IgG detection is 63% in patients with <2 weeks from disease onset; 85% in non-hospitalized patients with >2 weeks disease duration; and 91% in hospitalized patients with >2 weeks disease duration. We also compared different blood sample types and suggest a higher sensitivity by serum/plasma over whole blood. Test specificity was determined to be 97% on 69 sera/plasma samples collected between 2016-2018. Our study provides a comprehensive validation of the rapid COVID-19 IgM/IgG serology test, and mapped antibody detection patterns in association with disease progress and hospitalization. Our results support that the rapid COVID-19 IgM/IgG test may be applied to assess the COVID-19 status both at the individual and at a population level.

    • We present a Functional Compute Language (FCL) for low-levelGPU programming. FCL is functional in style, which allows foreasy composition of program fragments and thus easy prototypingand a high degree of code reuse. In contrast with projects such asFuthark, Accelerate, Harlan, Nessie and Delite, the intention is notto develop a language providing fully automatic optimizations, butinstead to provide a platform that supports absolute control of theGPU computation and memory hierarchies. The developer is thusrequired to have an intimate knowledge of the target platform, as isalso required when using CUDA/OpenCL directly.FCL is heavily inspired by Obsidian. However, instead of relyingon a multi-staged meta-programming approach for kernel generationusing Haskell as meta-language, FCL is completely selfcontained,and we intend it to be suitable as an intermediate languagefor data-parallel languages, including data-parallel parts ofhigh-level array languages, such as R, Matlab, and APL.We present a type-system and a dynamic semantics suitablefor understanding the performance characteristics of both FCL andObsidian-style programs. Our aim is that FCL will be useful as aplatform for developing new parallel algorithms, as well as a target languagefor various code-generators targeting GPU hardware.

    • Bone mineral density (BMD) is the most widely used predictor of fracture risk. We performed the largest meta-analysis to date on lumbar spine and femoral neck BMD, including 17 genome-wide association studies and 32,961 individuals of European and east Asian ancestry. We tested the top BMD-associated markers for replication in 50,933 independent subjects and for association with risk of low-trauma fracture in 31,016 individuals with a history of fracture (cases) and 102,444 controls. We identified 56 loci (32 new) associated with BMD at genome-wide significance (P < 5 × 10(-8)). Several of these factors cluster within the RANK-RANKL-OPG, mesenchymal stem cell differentiation, endochondral ossification and Wnt signaling pathways. However, we also discovered loci that were localized to genes not known to have a role in bone biology. Fourteen BMD-associated loci were also associated with fracture risk (P < 5 × 10(-4), Bonferroni corrected), of which six reached P < 5 × 10(-8), including at 18p11.21 (FAM210A), 7q21.3 (SLC25A13), 11q13.2 (LRP5), 4q22.1 (MEPE), 2p16.2 (SPTBN1) and 10q21.1 (DKK1). These findings shed light on the genetic architecture and pathophysiological mechanisms underlying BMD variation and fracture susceptibility.
