    • METIS WP4 covers research activities in network-level aspects of the advancement of wireless network technologies towards the year 2020 and beyond. The aim is to develop novel network-level technology concepts to address the challenges foreseen in future scenarios with regard to interference, traffic and mobility management issues. Moreover, another task of this work package is to propose functional enablers which can support the above potential solutions.This document provides* a report of the ongoing progress in WP4 regarding the research topics agreed upon in IR 4.1,* a high level description of the proposed concepts and approaches adopted by different partners.More specifically, the document describes, first set of potential network-level solutions and presents some first research results in order to position them with regards to the state of the art approaches. It also gives an overview of research activities to be considered later in WP4.

In order not to constrain the potential solutions, the requirements are specified from an end-user perspective. The deliverable will not only serve as the guideline for the technical work and system concept design in METIS, but also can serve in external research communities to help to harmonize the work towards the future radio access system including the new generation system of 5G

  • The Potential for Treatment Shortening With Higher Rifampicin Doses : Relating Drug Exposure to Treatment Response in Patients With Pulmonary Tuberculosis

    • Background. Tuberculosis remains a huge public health problem and the prolonged treatment duration obstructs effective tuberculosis control. Higher rifampicin doses have been associated with better bactericidal activity, but optimal dosing is uncertain. This analysis aimed to characterize the relationship between rifampicin plasma exposure and treatment response over 6 months in a recent study investigating the potential for treatment shortening with high-dose rifampicin. Methods. Data were analyzed from 336 patients with pulmonary tuberculosis (97 with pharmacokinetic data) treated with rifampicin doses of 10, 20, or 35 mg/kg. The response measure was time to stable sputum culture conversion (TSCC). We derived individual exposure metrics with a previously developed population pharmacokinetic model of rifampicin. TSCC was modeled using a parametric time-to-event approach, and a sequential exposure-response analysis was performed. Results. Higher rifampicin exposures increased the probability of early culture conversion. No maximal limit of the effect was detected within the observed range. The expected proportion of patients with stable culture conversion on liquid medium at week 8 was predicted to increase from 39% (95% confidence interval, 37%-41%) to 55% (49%-61%), with the rifampicin area under the curve increasing from 20 to 175 mg/L.h (representative for 10 and 35 mg/kg, respectively). Other predictors of TSCC were baseline bacterial load, proportion of culture results unavailable, and substitution of ethambutol for either moxifloxacin or SQ109. Conclusions. Increasing rifampicin exposure shortened TSCC, and the effect did not plateau, indicating that doses >35 mg/kg could be yet more effective. Optimizing rifampicin dosage while preventing toxicity is a clinical priority.

    • Alkyl glycosides were synthesised from starch and alcohols using Aspergillus oryzae a-amylase as catalyst. In the degradation of starch by a-amylase, the alcohols competed with water as glycosyl acceptors. In the reaction with methanol, methyl maltoside and methyl maltotrioside were the main alcoholysis products. Conversion of 45 g/1 starch in 30% methanol resulted in a product mixture containing 26 mM maltooligosaccharides and 3.6 mM methyl glycosides. With ethanol, propanol and butanol, alkyl maltosides and alkyl maltotetraosides were detected, and with benzyl alcohol, benzyl glycosides having two, three or five glucose units were formed. No alcoholysis reaction occurred with hexanol or octanol. In conclusion, alpha-amylase is promising for the one-step synthesis of alkyl glycosides having more than one monosaccharide unit, which are difficult to synthesise in other ways.

  • Boundary Layer Parametrization in Numerical Weather Prediction Models

    • Numerical weather prediction (NWP) and climate models have shown to have a challenge to correctly simulate stable boundary layers and diurnal cycles. This aim of this study is to evaluate, describe and give suggestions for improvements of the descriptions of stable boundary layers in operational NWP models. Two papers are included. Paper I focuses on the description of the surface and the interactions between the surface and the boundary layer in COAMPSR, a regional NWP model. The soil parametrization showed to be of great importance to the structure of the boundary layer. Moreover, it showed also that a low frequency of radiation calculations caused a bias in received solar energy at the surface.In paper II, the focus is on the formulation of the turbulent transport in stable boundary layers. There, an implementation of a diffusion parametrization based on the amount of turbulent kinetic energy (TKE) is tested in a single column model (SCM) version of the global NWP model Integrated Forecast System (IFS). The TKE parametrization turned out to behave similarly as the currently operational diffusion parametrization in convective regimes and neutral regimes, but showed to be less diffusive in weakly stable and stable conditions. The formulations of diffusion also turned out to be very dependent on the length scale formulation. If the turbulence and the gradients of wind temperature and wind are weak, the magnitude of turbulence can enter an oscillating mode. This oscillation can be avoided with the use of a lower limit of the length scale.
