    • This deliverable provides an architectural strategy and overall system design forthe DySCAS middleware system as a first step towards a complete systemspecification. The DySCAS Basic Architecture captures both the systemconceptualization and an initial function-level outline without consideringimplementation and technology details. It defines the middleware system in termsof its application and operational contexts, expected features, middleware servicesthat group functions, behaviours, as well as the structuring and implementationdecisions that are most crucial to satisfy the given set of requirements. In thisdocument, we also outline some fundamental strategies for binding themiddleware components to the target platforms and infrastructures and forhandling potential errors at both initialization-time and rum-time, while taking thetechnology support and domain needs into consideration.This deliverable also covers the rationale behind the architecture solutions andprovides a generic framework that relates architecture principles and styles, wellknownmechanisms, and reference models to the expected functionalities andqualities of DySCAS. The aim is to consolidate the proposed solutions and also tofacilitate the communications between DySCAS partners and to third-parties.

  • Genome-wide RNAi Screen Identifies Cohesin Genes as Modifiers of Renewal and Differentiation in Human HSCs

    • To gain insights into the regulatory mechanisms of hematopoietic stem cells (HSCs), we employed a genome-wide RNAi screen in human cord-blood derived cells and identified candidate genes whose knockdown maintained the HSC phenotype during culture. A striking finding was the identification of members of the cohesin complex (STAG2, RAD21, STAG1, and SMC3) among the top 20 genes from the screen. Upon individual validation of these cohesin genes, we found that their knockdown led to an immediate expansion of cells with an HSC phenotype in vitro. A similar expansion was observed in vivo following transplantation to immunodeficient mice. Transcriptome analysis of cohesin-deficient CD34(+) cells showed an upregulation of HSC-specific genes, demonstrating an immediate shift toward a more stem-cell-like gene expression signature upon cohesin deficiency. Our findings implicate cohesin as a major regulator of HSCs and illustrate the power of global RNAi screens to identify modifiers of cell fate.

    • Embedded systems, with their tight technology integration, andmultiple requirements andstakeholders, are characterized by tightly interrelated processes, information and tools.Embedded systems will as a consequence be described by multiple, heterogeneous andinterrelated descriptions such as for example requirements documents, design and analysismodels, software and hardware descriptions. We refer to a system designed this way as amulti-view (MV) system.The main contribution of this paper is a characterization of model-based approachesto MV systems. The characterization takes three main perspectives: semantic relationsbetween views (content), relations over time (process), and manipulation of views (operations).We complement these perspectives by investigating MV system challenges and bya survey of related approaches. The characterization aims to provide a basis for a betterunderstanding, design and implementation of MV systems, and thereby to overcome thecurrent fragmented points of view on integrated multi-view modeling (MVM).

    • In this report, we present a concept for generic multi-viewed component models based on modular  and (de)composable viewpoints.   By decomposing systems not only along component but also viewpoint borders, a family of multi-viewed component models is proposed. Semantic relations between the viewpoints are modeled in order to be able to ensure e.g. consistency between separate but semantically partly  overlapping views.  We also characterize different types of relations between viewpoints in order to see how the choice of viewpoints in the component model influences the complexity of multi-viewed components. Finally we provide an example and describe how the concepts presented can be applied in practice.

    • The complexity of automated driving poses challenges for providing safety assurance. Focusing on the architecting of an Autonomous Driving Intelligence (ADI), i.e. the computational intelligence, sensors and communication needed for high levels of automated driving, we investigate so called safety supervisors that complement the nominal functionality. We present a problem formulation and a functional architecture of a fault-tolerant ADI that encompasses a nominal and a safety supervisor channel. We then discuss the sources of hazardous events, the division of responsibilities among the channels, and when the supervisor should take over. We conclude with identified directions for further work.

    • Embedded and Cyber-Physical Systems education faces several challenges as well as opportunities as every-“thing” becomes connected, and as technology development accelerates. Initiatives such as CDIO, as well as several other academic and industry initiatives to create new CPS programs illustrate strong interests and awareness of these challenges. We provide an overview of foreseen educational needs, existing state of the art in education and an analysis of the subject of CPS with the purpose of understanding the implications for education. The investigation points to key issues in curriculum design regarding balancing depth and breadth, theory and practices, academic and industrial needs, and core technical skills with complementary skills. Curricula in CPS could, if the right balance is achieved, educate CPS engineers of the future that are “ready to engineer”. We conclude by synthesizing high level guidelines in terms of strategies and considerations for CPS curriculum development.

  • Scientific evidence of sustainable plant disease protection strategies for oats in Sweden: a systematic map

    • Background: Oat (Avena sativa L.) is an important cereal crop for livestock feed and human consumption. The largest oat-producing countries are located in the Northern Hemisphere with Sweden as the tenth largest producer. Oat production is challenged by different diseases that can lead to significant yield reductions and impaired grain quality. The use of efficient and sustainable plant protection management is of great economic and ecological importance. The systematic map in this study aims to provide a knowledge base inventory and to identify areas that need to be researched in the future in terms of plant disease management for more sustainable oat production.Methods: Literature searches were conducted in both academic bibliographic databases and relevant online sources of grey literature. A time-span restriction of 40 years (1978-2018) was applied to the searches. English was used in all searches, and Swedish, Norwegian and Danish languages were used in the grey literature searches. The screening process, which followed a protocol with eligibility criteria, was conducted at three levels: title, abstract and full text. Metadata incorporating bibliographic information, study location, climatic zone, disease name, the common and scientific names of the disease-causing organism, pathogen type, intervention and management methods, diseased plant part, plant stage, and outcome were extracted from the studies and included in the systematic map. The systematic map findings are visualized in figures and tables and described. All included studies can be found in a searchable database.Review findings: A total of 58 eligible articles, most (n = 51) from scientific journals published in English, were included in the systematic map. A majority of the studies were conducted in the Northern Hemisphere in temperate climatic zones, where most of the world's oats are produced. The earliest article was published in 1980, followed by an oscillating temporal distribution of articles over the following years. By country, Canada had the highest number of articles, and by region, Europe had the highest number. Fungi were the most studied pathogen type, and a total of 16 different diseases were reported. Fusarium head blight (Fusarium spp.) and crown rust (Puccinia coronata) were the most studied diseases. In total, 17 different intervention management approaches for controlling the diseases were analyzed in the articles, with cultivar resistance and pesticide application as the most studied methods.Conclusion: The map highlights the low quantity of available relevant field research on oat disease management. To our knowledge, this is the first systematic map of crop protection. This map provides a database of scientific literature that can be used to develop sustainable disease management strategies. The method used in this study has great potential and can also be used to benefit other crops. Research is often based on the availability of funding, and this map could be a useful tool for researchers and funding organizations to identify relevant research topics that need to be further studied. In addition, this systematic map offers a useful tool for field-based advisors in providing scientifically relevant crop protection strategies for farmers.
