  • DNA-streckkodning av marina växtplankton : Ett nytt verktyg i miljöövervakningen

    • Växtplankton utgör grunden i den marina näringsväven och används världen över för att bestämma miljöstatus i hav och sjöar. De ingår exempelvis som en kvalitetsfaktor i EU:s vattendirektiv och havsmiljödirektiv. Det finns långa tidsserier av växtplanktonövervakning där analyserna utförts med mikroskopi. Det sker nu en snabb internationell utveckling av DNA-metoder för att övervaka växtplankton. Målsättningen med detta projekt var att utveckla en praktisk och robust DNA analys­metod som kan implementeras i svensk marin miljöövervakning. Vårt tillvägagångssätt var att följa med på ordinarie marina miljöövervakningsexpeditioner under ett års tid (2019–2020) och ta parallella havsvattenprov för så kallad DNA-streckkodning av växtplankton. Sammanlagt tog vi prov vid 19 övervakningsstationer som var spridda från Bottenviken i norr till Skagerrak i söder. Provtagningsfrekvensen var cirka 1 gång per månad. Praktiska metoder utarbetades för fältprovtagning, DNA-extraktion, sekvensering, bioinformatisk analys och taxo­nomisk annotering. Vi har även tagit fram system för datahantering hos nationell datavärd och gett förslag på en ny datatyp för nationellt datavärdskap för marin­biologi och oceanografi vid Svenskt Oceanografiskt Datacentrum (https://sharkweb. smhi.se/hamta-data/). En viktig del har varit att jämföra resultaten av DNA-streckkodning och mikroskopi. Resultaten visar att DNA-streckkodning ger ungefär dubbelt så högt biodiversitetsmått än mikroskopering, även om det skiljer sig åt mellan olika grupper av växtplankton. För att undersöka om DNA-streckkodning kan användas för kvantitativ analys tillsatte vi en intern standard till proverna bestående av syntetiskt DNA, men eftersom resultatet varierade så behöver man arbeta vidare med detta. Den relativa fördelningen av vanliga eukaryota växtplanktongrupper visade sig ha relativt bra överensstämmelse mellan DNA-streckkodning och mikroskopimåttet kolbiomassa, medan biovolym och abundans skiljde sig åt mer. DNA-streckkodning visade sig ge detaljerade utbredningsmönster av skadliga alger, till exempel för släktet Prymnesium bland häftalgerna (Coccolithophyceae). Vi har inom projektet kunnat utvärdera ekologiska drivkrafter för växtplankton­samhällets diversitet och artsammansättning, genom att miljöövervakningen mäter många fysikalisk-kemiska parametrar. Både salthalt och närsalter visade sig ha stor inverkan på växtplanktonsamhällets sammansättning och diversitet. Sammanfattningsvis ser vi att den framtagna DNA-streckkodningsmetoden skulle vara ett bra komplement till den etablerade växtplanktonövervakningen.

  • Can probiotics be an alternative to chlorhexidine for oral care in the mechanically ventilated patient? A multicentre, prospective, randomised controlled open trial

    • Background: Pathogenic enteric bacteria aspirated from the oropharynx are the main cause of ventilator-associated pneumonia (VAP). Using chlorhexidine (CHX) orally or selective decontamination has been shown to reduce VAP. In a pilot study we found that oral care with the probiotic bacterium Lactobacillus plantarum 299 (Lp299) was as effective as CHX in reducing enteric bacteria in the oropharynx. To confirm those results, in this expanded study with an identical protocol we increased the number of patients and participating centres.Methods: One hundred and fifty critically ill patients on mechanical ventilation were randomised to oral care with either standard 0.1% CHX solution (control group) or a procedure comprising final application of an emulsion of Lp299. Samples for microbiological analyses were taken from the oropharynx and trachea at inclusion and subsequently at defined intervals.Student’s t test was used for comparisons of parameters recorded daily and Fisher’s exact test was used to compare the results of microbiological cultures.Results: Potentially pathogenic enteric bacteria not present at inclusion were identified in oropharyngeal samples from 29 patients in the CHX group and in 31 samples in the probiotic group. Considering cultures of tracheal secretions, enteric bacteria were found in 17 and 19 samples, respectively. Risk ratios show a difference in favour of the Lp group for fungi in oropharyngeal cultures. VAP was diagnosed in seven patients in the Lp group and in 10 patients among the controls.Conclusions: In this multicentre study, we could not demonstrate any difference between Lp299 and CHX used in oral care procedures regarding their impact on colonisation with emerging potentially pathogenic enteric bacteria in the oropharynx and trachea.Trial registration: ClinicalTrials.gov, NCT01105819.   • A comprehensive dataset on spatiotemporal variation of microbial plankton communities in the Baltic Sea

    • The Baltic Sea is one of the largest brackish water environments on earth and is characterised by pronounced physicochemical gradients and seasonal dynamics. Although the Baltic Sea has a long history of microscopy-based plankton monitoring, DNA-based metabarcoding has so far mainly been limited to individual transect cruises or time-series of single stations. Here we report a dataset covering spatiotemporal variation in prokaryotic and eukaryotic microbial communities and physicochemical parameters. Within 13-months between January 2019 and February 2020, 341 water samples were collected at 22 stations during monthly cruises along the salinity gradient. Both salinity and seasonality are strongly reflected in the data. Since the dataset was generated with both metabarcoding and microscopy-based methods, it provides unique opportunities for both technical and ecological analyses, and is a valuable biodiversity reference for future studies, in the prospect of climate change.

    • © 2019, The Author(s). Areas in western Antarctica are experiencing rapid climate change, where ocean warming results in more sea ice melt simultaneously as oceanic CO2 levels are increasing. In this study, we have tested how increased temperature (from −1.8to 3°C) and decreased salinity (from 35 to 20 and 10) synergistically affect the growth, photophysiology and biochemical composition of the Antarctic sea-ice diatom Nitzschia lecointei. In a separate experiment, we also addressed how ocean acidification (from 400 to 1000µatm partial pressure of CO2) affects these key physiological parameters. Both positive and negative changes in specific growth rate, particulate organic carbon to particulate organic nitrogen ratio, chl a fluorescence kinetics, lipid peroxidation, carbohydrate content, protein content, fatty acid content and composition were observed when cells were exposed to warming and desalination. However, when cells were subjected to increased pCO2, only Fv/Fm, non-photochemical quenching and lipid peroxidation increased (by 3, 16 and 14%, respectively), and no other of the abovementioned biochemical properties were affected. These results suggest that changes in temperature and salinity may have more effects on the biochemical composition of N. lecointei than ocean acidification. Sea-ice algae are important component of polar food webs, and their nutritional quality may be affected as a result of altered environmental conditions due to climate change and sea ice melt.

  • Gomphonemopsis ligowskii, a new diatom (Bacillariophyceae) from the marine Antarctic and a comparison to other Gomphonemopsis

    • A new marine diatom, Gomphonemopsis ligowskii, is described. Gomphonemopsis ligowskii was found as an epiphyte on the brown macrophyte, Desmarestia anceps, collected from King George Island, Antarctica in January 2016. This species is the first of the genus described from a polar region. It is a small and relatively rare taxon that is characterized by its unique shape and the presence of uniseriate striae, composed of round poroidal areolae on its valve face. The striae are arranged in parallel rows on either side of the axial area, and a row of rounded areolae on both sides of the valve mantle is confined to the wider part of the valve (headpole). A description of valve ultrastructure as well as a comparison with related taxa are provided.

    • - A new marine diatom species, Nitzschia biundulata is described. This species was collected and isolated from the ice cover in the Amundsen Sea, Antarctica during the Austral summer, 2011. The description is based on light and scanning electron microscopes examination as well as molecular analysis including 18S SSU rRNA and rbcL. Morphologically, N. biundulata is characterized by having two extensions of the valve margin in a form of two unequal undulations that are not seen in other taxa of the genus. Molecular sequence data indicated similarity with several Nitzschia species such as Nitzschia apiculata, N. palea, N. capitellata as well as Bacillaria paxillifer. Interestingly, large similarity was found with endosymbiont diatom associated with the dinophyte Peridinium balticum. A comparison with the most related species is provided. © 2019 J.     • Continuous observations of Antarctic benthic diatoms are necessary to detect changes in species composition and biodiversity that may result from environmental changes. The present work provides a systematic list of benthic diatoms from Potter Cove (62.03 degrees S 58.35 degrees W) collected during summer 2015. The new findings are compared with observations made 12 years ago (summer 2003). In total, 80 taxa were found, similar to that encountered earlier, but 17 species (21% of the total species number) were not observed in 2003, and 26 species (31%) of those reported in 2003 did not appear in 2015. The dominant species in 2003 and 2015 was the large epipelic and cosmopolitan Gyrosigma fasciola. Most of the species either newly observed in the present study, or absent in the present study but observed previously, occurred either rarely or very rarely. Gyrosigma arcuatum and Pleurosigma diversestriatum were newly observed in 2015 and not previously reported from polar regions. This study provides systematic and ecological information on all taxa encountered and is illustrated with light microscopy and scanning electron microscopy (SEM) images. We emphasize that ice-inhabiting and other benthic diatoms should be included when considering the biodiversity of polar diatoms. Thus, we hope that the present study will add a piece to the puzzle of climate change effects on the benthic diatoms in this vulnerable region and will complement earlier published species records in the area.

  • Application of Control Theory to a Commercial Mobile Service Support System

    • The Mobile Service Support system (MSS), which Ericsson AB develops, handles the setup of new subscribers and services into a mobile network. Experience from deployed systems show that traffic monitoring and control of the system will be crucial for handling overload situations that may occur at sudden traffic surges. In this paper we identify and explore some important control challenges for this type of systems. Further, we present analysis and experiments showing some advantages of proposed solutions. First, we develop a load-dependent server model for the system, which is validated in testbed experiments. Further, we propose a control design based on the model, and a method for estimation of response times and arrival rates. The main contribution of this paper is that we show how control theory methods and analysis can be used for commercial telecom systems. Parts of our results have been implemented in commercial products, validating the strength of our work.

This study employs numerical simulations to investigate the propensity of squat initiation in the vicinity of small, isolated rail surface irregularities. Time domain dynamic vehicle–track interaction analysis is used to obtain wheel–rail contact stress distributions, which are mapped onto a continuum finite element model that accounts for plastic deformation of the rail material. The evaluated stress and strain fields are quantified using two RCF impact measures: accumulated effective strain and the Jiang-Sehitoglu multiaxial low cycle fatigue parameter. It is shown that the RCF impact increases with increasing size of the surface irregularity and that clustering of irregularities might strongly promote RCF. The friction coefficient is identified as a very influential parameter. Further, the effect of variations in friction along the rail is evaluated. It is shown that a short rail section of low friction results in fairly high RCF impact.
