  • Robust estimation of bacterial cell count from optical density

    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

  • Distribution of injected CO2 in a stratified saline reservoir accounting for coupled wellbore-reservoir flow

    • Geological storage in sedimentary basins is considered a viable technology in mitigating atmospheric CO2 emissions. Alternating high and low permeability strata are common in these basins. The distribution of injected CO2 among such layers affects e.g. CO2 storage efficiency, capacity and plume footprint. A numerical study on the distribution of injected CO2 into a multi-layered reservoir, accounting for coupled wellbore-reservoir flow, was carried out using the T2Well/ECO2N code. A site-specific case as well as a more general case were considered. Properties and processes governing the distribution of sequestrated CO2 were identified and the potential to operationally modify the distribution was investigated. The distribution of CO2 was seen to differ from that of injected water, i.e. it was not proportional to the transmissivity of the layers. The results indicate that caution should be taken when performing numerical simulations of CO2 injection into layered formations. Ignoring coupled wellbore-reservoir flow and instead adopting a simple boundary condition at the injection well, such as an inflow rate proportional to the transmissivity of each layer, may result in significant underestimation of the proportion of CO2 ending up in the shallower layers, as not all relevant processes are accounted for. This discrepancy has been thoroughly investigated and quantified for several CO2 sequestration scenarios.

  • Hydrologic issues associated with nuclear waste repositories

    • Significant progress in hydrology, especially in subsurface flow and solute transport, has been made over the last 35 years because of sustained interest in underground nuclear waste repositories. The present paper provides an overview of the key hydrologic issues involved, and to highlight advances in their understanding and treatment because of these efforts. The focus is not on the development of radioactive waste repositories and their safety assessment, but instead on the advances in hydrologic science that have emerged from such studies. Work and results associated with three rock types which are being considered to host the repositories, are reviewed, with a different emphasis for each rock type. The first rock type is fractured crystalline rock, for which the discussion will be mainly on flow and transport in saturated fractured rock. The second rock type is unsaturated tuff, for which the emphasis will be on flow from the shallow subsurface through the unsaturated zone to the repository. The third rock type is clay-rich formations, whose permeability is very low in an undisturbed state. In this case, the emphasis will be on hydrologic issues that arise from mechanical and thermal disturbances; i.e., on the relevant coupled thermo-hydro-mechanical processes. The extensive research results, especially those from multi-year large-scale underground research laboratory investigations, represent a rich body of information and data that can form the basis for further development in the related areas of hydrologic research.

  • Hydraulic Modeling of Induced and Propagated Fractures : Analysis of Flow and Pressure Data From Hydromechanical Experiments in the COSC-1 Deep Borehole in Crystalline Rock Near Åre, Sweden

    • To characterize the coupled hydromechanical behavior of rock fractures, the step-rate injection method for fracture in-situ properties (SIMFIP) was conducted with a specialized downhole probe developed by Guglielmi et al. (2014, https://doi.org/10.1007/s00603-013-0517-1). In June 2019, a field campaign was carried out near Åre, Sweden, where the SIMFIP probe was applied in the Collisional Orogeny in the Scandinavian Caledonides-1 scientific borehole to understand the dynamics of injection-induced fracture initiation, fracture opening, and shearing due to water injection-withdrawal in a borehole interval isolated by two packers. Three intervals were investigated at ∼500 m depth: (a) an unfractured section (intact rock), (b) a section with non-conductive fractures, and (c) a section with hydraulically conductive fractures. Pressure, injection flow rate, and borehole wall displacement were simultaneously measured during the tests. In the present study, the geometry of the induced fracture and deformation of existing fractures at different time stages of the tests are determined based on a hydrologic model by using the measured pressure and flow data during each time stage of the experiment. A numerical model for the fluid flow within the fracture and the packed-off borehole interval is implemented within COMSOL Multiphysics. By matching model simulations with observed data for all three sections, estimates of the induced and propagated fractures' radius and aperture at successive time stages have been obtained in each case. We could also determine the non-linear relationship between fracture aperture and pressure for values above fracture opening pressures. The model results provide insights for the understanding of pressure-induced fracture initiation and propagation in crystalline rock.

  • 25 years of DECOVALEX - Scientific advances and lessons learned from an international research collaboration in coupled subsurface processes

    • This paper provides an overview of an international research collaboration for advancing the understanding and modeling of coupled thermo-hydro-mechanical-chemical (THMC) processes in geological systems. The creation of the international DECOVALEX Project, now running for over 25 years, was initially motivated by the recognition that prediction of these coupled effects is an essential part of the performance and safety assessment of geologic disposal systems for radioactive waste and spent nuclear fuel. Later it was realized that these processes also play a critical role in other subsurface engineering activities, such as storage of CO2, exploration of enhanced geothermal systems, and unconventional oil and gas production through hydraulic fracturing. Research teams from radioactive waste management organizations, national research institutes, regulatory agencies, universities, as well as industry and consulting groups have participated in the DECOVALEX Project, providing a wide range of perspectives and solutions to these complex problems. Analysis and comparative modeling of state-of-the-art field and laboratory experiments has been at the core of the collaborative work, with an increasing focus on characterizing uncertainty and blind prediction of experimental results. Over these 25 years, many of the major advances in this field of research have been made through DECOVALEX, as evidenced by three books, seven journal special issues, and a good number of seminal papers that have emerged from the DECOVALEX modeling work. Examples of specific research advances will be presented in this paper to illustrate the significant impact of DECOVALEX on the current state-of-the-art of understanding and modeling coupled THMC processes. These examples range from the modeling of large-scale in situ heater tests representing mock-ups of nuclear waste disposal tunnels, to studies of fluid flow and chemical-mechanical coupling in heterogeneous fractures, and to the numerical analysis of controlled-injection meso-scale fault slip experiments.

  • Hydrodynamic control of retention in heterogeneous aquifers and fractured rock

    • In this thesis, fluid flow and solute transport in heterogeneous aquifers and particularly in frac-tured rock have been investigated using Lagrangian Stochastic Advective-Reaction (LaSAR) framework. The heterogeneity of the aquifer structure or fracture configuration, as well as the various reaction/retention processes have been considered in the modelling approach. Advection and retention processes are considered to be the dominant transport processes. Monte-Carlo simulation results for transport of nonreactive tracers in 2D generic heterogeneous aquifers indicate that the travel time τ can be well approximated by a lognormal distribution up to a relative high degree of heterogeneity of the aquifers. Comparison between the Monte-Carlo simulation results and the results of first-order approximation reveals that the analytical solutions of the statistical moments of τ are valid only when the variability of the aquifer properties is small. For reactive tracers, Monte-Carlo simulations have been conducted by accounting for spatial variability of both hydraulic conductivity and one sorption parameter simultaneously. The simulation results indicate that the reaction flow path μ is a nonlinear function of distance for shorter distance, linear function for longer distance, and also that μ and τ are well correlated over the considered parameter range. The parameter β, which is purely determined by the flow condi-tions, quantifies the hydrodynamic control of retention processes for transport of tracers in frac-tures. Numerical simulations have been performed to study the statistical properties of the pa-rameter β, travel time τ and flow rate Q in a single heterogeneous fracture and in a sequence of fractures. The results of Monte-Carlo simulations indicate that the parameter β and τ are corre-lated with a power-law relationship β ∼ τm. The correlation between β and the flow rate Q have also been studied and an inverse power-law relationship β ∼ Q-m is proposed. The establishment of these relationships provides a link between the parameter β and measurable parameters τ (or Q). The LaSAR approach has been applied for prediction, evaluation and interpretation of the results of a number of tracer tests (TRUE-1, TRUE Block Scale and TRUE Block Scale Continuation) conducted by SKB at the Äspö site for tracer transport in fractures. The breakthrough curves may be predicted reasonably well, provided that the retention parameters, boundary conditions and hydraulic properties of the domain are given. The evaluation of TRUE tests indicates that the retention occurs mainly in the rim zone on site characterization time scales, while on the per-formance assessment time scale, diffusion and sorption in the unaltered rock matrix are likely to become dominant retention mechanisms.
