    • Intrinsic dynamics of DNA plays a crucial role in DNA protein interactions and has been emphasized as a possible key component for in vivo chromatin organization. We have prepared an entangled DNA microtube above the overlap concentration by exploiting the complementary cohesive ends of lambda-phage DNA, which is confirmed by atomic force microscopy and agarose gel electrophoresis. Photon correlation spectroscopy further confirmed that the entangled solutions are found to exhibit the classical hydrodynamics of a single chain segment on length scales smaller than the hydrodynamic length scale of single lambda-phage DNA molecule. We also observed that in 41.6% (gm water/gm DNA) hydrated state, lambda-phage DNA exhibits a dynamic transition temperature (T-dt) at 187 K and a crossover temperature (T-c) at 246 K. Computational insight reveals that the observed structure and dynamics of entangled lambda-phage DNA are distinctively different from the behavior of the corresponding unentangled DNA with open cohesive ends, which is reminiscent with our experimental observation.

  • A variable active galactic nucleus at z = 2.06 triply-imaged by the galaxy cluster MACS J0035.4−2015

    • We report the discovery of a triply imaged active galactic nucleus (AGN), lensed by the galaxy cluster MACS J0035.4−2015 (z d = 0.352). The object is detected in Hubble Space Telescope imaging taken for the RELICS program. It appears to have a quasi-stellar nucleus consistent with a point-source, with a de-magnified radius of re ≲ 100 pc. The object is spectroscopically confirmed to be an AGN at z spec = 2.063 ± 0.005 showing broad rest-frame UV emission lines, and detected in both X-ray observations with Chandra and in ALCS ALMA band 6 (1.2 mm) imaging. It has a relatively faint rest-frame UV luminosity for a quasar-like object, MUV, 1450 = −19.7 ± 0.2. The object adds to just a few quasars or other X-ray sources known to be multiply lensed by a galaxy cluster. Some diffuse emission from the host galaxy is faintly seen around the nucleus, and there is a faint object nearby sharing the same multiple-imaging symmetry and geometric redshift, possibly an interacting galaxy or a star-forming knot in the host. We present an accompanying lens model, calculate the magnifications and time delays, and infer the physical properties of the source. We find the rest-frame UV continuum and emission lines to be dominated by the AGN, and the optical emission to be dominated by the host galaxy of modest stellar mass M✶ ≃ 109.2 M⊙. We also observe some variation in the AGN emission with time, which may suggest that the AGN used to be more active. This object adds a low-redshift counterpart to several relatively faint AGN recently uncovered at high redshifts with HST and JWST.

  • Classification tree model of the personal economic burden of dementia care by related factors of both people with dementia and caregivers in Japan: a cross-sectional online survey

    • The purpose of this study was to clarify the microlevel determinants of the economic burden of dementia care at home in Japanese community settings by classifying them into subgroups of factors related to people with dementia and their caregivers.DesignA cross-sectional online survey.Participants4313 panels of Japanese research company who fulfilled the following criteria: (1) aged 30 years or older, (2) non-professional caregiver of someone with dementia, (3) caring for only one person with dementia and (4) having no conflicts of interest with advertising or marketing research entities.Primary outcome measuresInformal care costs and out-of-pocket payments for long-term care (LTC) services.ResultsFrom 4313 respondents, only 1383 caregivers in community-settings were included in this analysis. We conducted a χ² automatic interaction detection analysis to identify the factors related to each cost (informal care costs and out-of-pocket payments for LTC services) divided into subcategories. In the resultant classifications, informal care cost was mainly related to caregivers’ employment status. When caregivers acquired family care leave, informal care costs were the highest. On the other hand, out-of-pocket payments for LTC were related to care-need levels and family economic status. Activities of Daily Living and Instrumental Activities of Daily Living functions such as bathing, toileting and cleaning were related to all costs.ConclusionThis study clarified the difference in dementia care costs between classified subgroups by considering the combination of the situations of both people with dementia and their caregivers. Informal care costs were related to caregivers’ employment and cohabitation status rather to the situations of people with dementia. On the other hand, out-of-pocket payments for LTC services were related to care-need levels and family economic status. These classifications will be useful in understanding which situation represents a greater economic burden and helpful in improving the sustainability of the dementia care system in Japan.
