  • Food-induced expression of orexin receptors in rat duodenal mucosa regulates the bicarbonate secretory response to orexin-A

    • Presence of appetite-regulating peptides orexin-A and orexin-B in mucosal endocrine cells suggests a role in physiological control of the intestine. Our aim was to characterize orexin-induced stimulation of duodenal bicarbonate secretion and modulation of secretory responses and mucosal orexin receptors by overnight food deprivation. Lewis x Dark Agouti rats were anesthetized and proximal duodenum cannulated in situ. Mucosal bicarbonate secretion (pH stat) and mean arterial blood pressure were continuously recorded. Orexin-A was administered intra-arterially close to the duodenum, intraluminally, or into the brain ventricles. Total RNA was extracted from mucosal specimens, reverse transcribed to cDNA and expression of orexin receptors 1 and 2 (OX1 and OX2) measured by quantitative real-time PCR. OX1 protein was measured by Western blot. Intra-arterial orexin-A (60–600 nmol·h–1·kg–1) increased (P < 0.01) the duodenal secretion in fed but not in fasted animals. The OX1 receptor antagonist SB-334867, which was also found to have a partial agonist action, abolished the orexin-induced secretory response but did not affect secretion induced by the muscarinic agonist bethanechol. Atropine, in contrast, inhibited bethanechol but not orexin-induced secretion. Orexin-A infused into the brain ventricles (2–20 nmol·kg–1·h–1) or added to luminal perfusate (1.0–100 nM) did not affect secretion, indicating that orexin-A acts peripherally and at basolateral receptors. Overnight fasting decreased mucosal OX1 and OX2 mRNA expression (P < 0.01) as well as OX1 protein expression (P < 0.05). We conclude that stimulation of secretion by orexin-A may involve both receptor types and is independent of cholinergic pathways. Intestinal OX receptors and secretory responses are markedly related to food intake.

    • Archäologisch relevante Forschungsdaten beste- hen aus einer Vielzahl verschiedener Arten von Informationen. Das Problem, dass Daten in ver- schiedenen Formaten und in örtlich verteilten Standorten vorliegen, wirkt sich auf eine effektive Verwaltung und die Verwendung des Vermögens aus. Es ist schwierig den Überblick zu behalten auf das, was Material ist, wo es sich und in welchem Zustand befindet und wie der allgemeine Zustand der Daten ist. In der Regel ist aus dem gleichen Grund das Material schwierig und zeitaufwendig zu übersetzen und zu verwenden. Es ist ein grund- sätzliches Problem bei archäologischen Datenar- chiven, die Forderung nach Integrität von Daten und Ressourcen mit der tiefen Kontextualität von archäologischen Daten und Interpretationen zu verbinden.Dieser Artikel beschreibt einen neuen Ansatz für die Integration von archäologischen Daten mit Hilfe eines semantischen Wiki-System, um die Integration von formalen Beschreibungen und flexiblen Unterlagen zu ermöglichen. Die sonsti- gen Vorzüge eines semantischen, Wiki-basierten Ansatzes sind die relativ einfache Bedienung, die Flexibilität der Bearbeitung, die Beschreibung und Strukturierung von Daten, die Unterstützung gr- undsätzlich aller denkbaren Arten von Daten und die Möglichkeit zur Zusammenarbeit und die Nutzung der Ressourcen in der Ferne.

  • Taking excavation to a virtual world : Importing archaeological spatial data to Second Life and OpenSim

    • The benefits of analysing and presenting archaeological spatial data in an interactive 3D environment have been discussed extensively in the literature. This paper reports of a R&D project that explored the possibilities of presenting archaeological information in virtual worlds with a specific focus on presenting and using actual documentation data captured by total stations and laser scanners directly in virtual worlds. Field trials were conducted in Second Life and OpenSim environments. The findings indicated that Second Life was a preferred environment because of the relatively large existing ecology of individual and institutional users. The proprietary nature of the environment and the consequent limitations to data transfer and the control of the world did, however, make the importing, linkage and manipulation of data problematic. OpenSim allowed a total control of the environment, but lacked certain technical features implemented in Second Life together with a comparable, large population of users.

    • Mobile technology has been seen as a major asset in on-site education for a decade, but until recently, many practi- cal issues have reduced the functionality and usability of applications. Different applications use varying approaches and platforms from mobile phones to larger tablet and laptop computers. The paper discusses the present evolution version of the Pulu application developed in LAMB project and how the pedagogic premises of the project have been realised in the technical development process. The client has new functionalities for designing routes and implemen- tation of a system question answering by moving in the environment. The system has also a new web based repository for sharing route data and other information.

    • The present paper discusses the results of an R&D project (fi PULU) in south western Finland to develop a general purpose mobile application framework for elementary and grammar schools with a special emphasis on distributed content creation and a usage model anchored in the convergence of pedagogics and location-based cultural heritage information. Since the beginning of 2001, the LAMB project and its predecessors have studied, developed and evaluated several different technological and content design approaches starting from stand-alone applications on palmtop computers, tablet PCs, and smart phones to modern small light-weight portable tablet computers. The technological focus of the effort has been to develop a simple and robust position-aware multiplatform software application based on a simple general purpose data model based on locations. The Java application designed for the LAMB project can be run on diverse portable computers and used by a group of pupils together. Interactions with the application are recorded on the device and the teacher can afterwards evaluate pupil input and device use data. The software is based on freely available and Open Source components. An important aspect of the system is that it can be updated and maintained on site by the local staff without extensive technical knowledge.
