    • Oxytocin treatment may permanently alter endocrine axes resulting in anti-stress and anabolic effects. However, the nutritional status influences the effects of oxytocin. The specific aims of this study were to investigate the effects of postnatal oxytocin treatment on reproductive performance in adult life, by studying maternal weight gain, adiposity, plasma levels of IGF-I as well as fetal and placental weights in the following groups of animals: (1) Ad libitum fed dams coming from ad libitum fed mothers. (2) Ad libitum fed dams coming from food-restricted mothers. (3) Food-restricted dams coming from ad libitum fed mothers. (4) Food-restricted dams coming from food-restricted mothers. Oxytocin treatment postnatally had long-term effects and increased adiposity in pregnant dams and stimulated placental and fetal growth relative to saline-treated dams. However, if the dams themselves had been exposed to food restriction during fetal life, the effect of postnatal oxytocin treatment changed. The oxytocin-treated mothers were still fatter but had smaller fetuses. In conclusion, postnatal oxytocin treatment influences reproductive performance in later life but is dependent on the mother’s previous and current nutritional experience.

  • Feeding and Manual Brushing Influence the Release of Oxytocin, ACTH and Cortisol Differently During Milking in Dairy Cows

    • AimThis study aimed to examine the effects of feeding or abdominal brushing on the release of the hormones oxytocin, ACTH and cortisol during milking in dairy cows. MethodsTwelve cows in early lactation were used (2 x 2 factorial experimental design), testing the effects of two types of sensory stimulation during milking over a 3 day period; feeding concentrate or manual abdominal brushing (1 stroke/s). Blood samples for hormone analyses were collected at time at -15, -1, 0 (onset of cluster), every min for 8 min, at 10, 12, 14, 16, 30, and 60 min. Hormone levels were assayed and AUC was calculated. ResultsMilking was associated with an immediate and significant rise of oxytocin. When milking was combined with feeding, significantly higher levels of oxytocin were observed at 2 and 4 mins (p < 0.05). No effect of brushing on oxytocin levels was observed. Milking alone was associated with a significant rise of ACTH levels. Feeding in connection with milking reduced the immediate rise of ACTH levels (p < 0.05) and AUC (p < 0.02), whereas no effects of brushing were found. Milking caused a progressive rise of cortisol levels. Concomitant feeding did not influence cortisol levels, whereas brushing significantly decreased cortisol levels at 1, 5 and 14 mins after onset of milking (p < 0.05). ConclusionFeeding increases oxytocin release in response to milking and decreases ACTH levels. Abdominal brushing did not influence these variables, but decreased cortisol levels. These data demonstrate that activation of afferent vagal nerve fibres and of cutaneous sensory nerves originating from the abdominal skin in front of the udder influence milking related hormone release differently.

    • Patients being treated for stress-related mental illness were observed during two summer programs in order to investigate the influence of multimodal nature-based therapy in a specially designed healing garden on patient recovery. The aim was to distinguish specific qualities, patterns and/or processes during the participants' stay in the healing garden. The study is a single-case study, using participatory observation. The data were coded following an inductive research process. The results showed that patients who underwent psychotherapy were more open and contact-seeking, and carried out extrovert recreational walks. By contrast, patients who underwent physiotherapy were introverted, emotionally withdrawn and performed introvert recreational walks. Interpretation of the data suggested that treatment combined with activities in certain parts of the rehabilitation garden induced and supported different psycho-physiological processes. The mechanisms and progress of these are discussed from the theory of situated cognition, and how these processes are stimulated and supported by characteristics in the garden. Oxytocin, a hypothalamic peptide which stimulates social interaction, induces anti-stress effects and stimulates growth and healing, may hypothetically be involved in these processes.

  • Oxytocin improves cytological and histological profiles of vaginal atrophy in postmenopausal women

    • OBJECTIVE: To investigate if topical oxytocin can reverse vaginal atrophy, as assessed by cytological and histological examination of the vaginal mucosal epithelium, in postmenopausal women after 12 weeks of treatment as compared to placebo.STUDY DESIGN: Sixty-eight postmenopausal women diagnosed with vaginal atrophy were randomized for this multicenter, double-blinded, placebo-controlled trial. Thirty-three women received 600 IU vagitocin, an oxytocin containing gel, and 35 women received a placebo gel intravaginally. The dose was 600 IU daily for the first two weeks and thereafter 600 IU twice a week for 10 weeks. All participant women underwent four visits and a subgroup of 20 women had a further fifth visit. Vaginal smears for cytological evaluation were collected at all visits. Vaginal biopsies were taken in 20 women before and after 12 weeks of treatment for histological analysis. In these women a vaginal smear was also collected after 14 weeks.RESULTS: The increase in the percentage of superficial cells between 0 and 2 weeks was significantly greater after treatment with vagitocin in comparison with placebo (p = 0.04). The difference in the maturation value between 0 and 12 weeks was significantly higher in the vagitocin than in the placebo group (p = 0.01). The reduction in the scores of atrophy was according to the histological investigation significantly greater in the vagitocin group than in the placebo group at 12 weeks (p < 0.04).CONCLUSION: Daily intravaginal treatment with vagitocin 600 IU improves expressions of vaginal atrophy as recorded by cytological investigation of vaginal smears and histological analysis of vaginal biopsies. Treatment twice weekly seems to be less effective regarding the increase in superficial cells.

    • To explore the efficacy of local oxytocin for the treatment of post-menopausal vaginal atrophy. Design Double-blinded randomised controlled trial. Setting Healthy post-menopausal women in Stockholm, Sweden. Participants Sixty four post-menopausal women between February and June 2012 at the Karolinska University Hospital Huddinge/Sweden. Main outcome measures The efficacy of oxytocin for treatment of vaginal atrophy after seven weeks and cytological evaluation. Results The percentage of superficial cells in the vaginal smears and the maturation values were significantly increased after seven weeks of treatment with vagitocin 400 IU ( p = 0.0288 and p = 0.0002, respectively). The vaginal pH decreased significantly after seven weeks of treatment with vagitocin 100 IU ( p = 0.02). The scores of vaginal atrophy, according to the histological evaluation, were significantly reduced after administration of vagitocin 100 IU ( p = 0.03). The thickness of the endometrium did not differ between the treatment and placebo groups after seven weeks of treatment. The symptom experienced as the most bothersome was significantly reduced after seven weeks of treatment in the women receiving vagitocin 400 IU compared to women in the placebo group ( p = 0.0089). Conclusions Treatment with intravaginally applied oxytocin could be an alternative to local estrogen treatment in women with post-menopausal vaginal atrophy.

  • Behavioural expression of positive anticipation for food or opportunity to play in lambs

    • Anticipatory behaviours may serve a useful tool in studying positive emotional states in animals. This study aimed to investigate if lambs express anticipatory behaviours for opportunities to play or a food reward and if these behaviours would differ depending on the anticipated event. Forty-two male lambs were allocated into three treatment groups (control, play, food). Play and food lambs were conditioned to anticipate play or food in a holding pen for three minutes prior to accessing a reward arena containing toy objects or concentrate, respectively. Control lambs returned to their home pen following three minutes in the holding pen. Compared to the control lambs, both play and food lambs differed in several behaviours frequencies and durations, e.g. by an increased frequency of behavioural transitions and duration of walking. Following these observations, food lambs received the toy objects when entering the reward arena. The subsequent session in the holding pen resulted in a decrease in number of behavioural transitions, time spent walking and an increase in time standing still. In conclusion, anticipating a positive event resulted in differences in behaviour compared to a control group, and these behaviours are affected when the anticipated event does not fit with the lambs' expectations.   • The Effect of a Real Dog, Toy Dog and Friendly Person on Insecurely Attached Children During a Stressful Task : An Exploratory Study

    • The regulation of stress by an attachment figure is a key feature of attachment relationships. Previous research suggests that in some cases animal companionship may be regarded as an attachment relationship. This may be particularly important for persons with an insecure or disorganized attachment pattern who may find it more difficult than securely attached individuals to accept social support from humans. In our study, we investigated whether 31 boys (aged 7-12 years) with insecure/disorganized attachment would profit more from the presence of a dog (n = 11) than of a friendly human (n = 11) or a toy dog (n = 9) as support during a socially stressful situation (Trier Social Stress Test for Children, TSST-C). Stress levels were assessed via salivary cortisol recorded five times before, during, and after the TSST-C. The behavior of the children was coded from video recordings. Self-reported stress levels did not significantly differ between the groups before and after the TSST-C. Salivary cortisol, however, was significantly lower in the real dog condition than in the other two conditions (Kruskal-Wallis H test on area under the curve increase (AUCi): chi(2) = 15.17, df = 2, p = 0.001). Also, the more the children stroked the dog, the less pronounced was their stress reaction (r(s) = -0.818, p = 0.002). Our data suggest an important role of physical contact in the stress reducing effect. We conclude that the children investigated profited more from interacting with a friendly dog than with either a human or a toy dog in a stressful situation. We discuss the relevance of our findings for animal-assisted interventions.
