    • The New York City Off-Hour Delivery (NYC OHD) program is the work of a private-public-academic partnership - A collaborative effort of leading private-sector groups and companies, public-sector agencies led by the New York City Department of Transportation, and research partners led by Rensselaer Polytechnic Institute. The efforts of this partnership have induced more than 400 commercial establishments in NYC to accept OHD without supervision. The economic benefits are considerable: The carriers have reduced operational costs and parking fines by 45 percent; the receivers enjoy more reliable deliveries, enabling them to reduce inventory levels; the truck drivers have less stress, shorter work hours, and easier deliveries and parking; the delivery trucks produce 55-67 percent less emissions than they would during regular-hour deliveries, for a net reduction of 2.5 million tons of CO2 per year; and citizens' quality of life increases as a result of reduced conflicts between delivery trucks, cars, bicycles, and pedestrians, and through the use of low-noise delivery practices and technologies that minimize the impacts of noise. The total economic benefits exceed $20 million per year. The success of the OHD program is due largely to the policy design at its core, made possible with the behavioral microsimulation. This unique optimization-simulation system incorporates the research conducted into an operations research/management science tool that assesses the effectiveness of alternative policy designs. This enabled the successful implementation of the project within the most complex urban environment in the United States.

  • Full-lifetime simulations of multiple unequal-mass planets across all phases of stellar evolution

    • We know that planetary systems are just as common around white dwarfs as around main-sequence stars. However, self-consistently linking a planetary system across these two phases of stellar evolution through the violent giant branch poses computational challenges, and previous studies restricted architectures to equal-mass planets. Here, we remove this constraint and perform over 450 numerical integrations over a Hubble time (14 Gyr) of packed planetary systems with unequal-mass planets. We characterize the resulting trends as a function of planet order and mass. We find that intrusive radial incursions in the vicinity of the white dwarf become less likely as the dispersion amongst planet masses increases. The orbital meandering which may sustain a sufficiently dynamic environment around a white dwarf to explain observations is more dependent on the presence of terrestrial-mass planets than any variation in planetary mass. Triggering unpacking or instability during the white dwarf phase is comparably easy for systems of unequal-mass planets and systems of equal-mass planets; instabilities during the giant branch phase remain rare and require fine-tuning of initial conditions. We list the key dynamical features of each simulation individually as a potential guide for upcoming discoveries.

  • Polygenic risk for anxiety influences anxiety comorbidity and suicidal behavior in bipolar disorder

    • Bipolar disorder is often comorbid with anxiety, which is itself associated with poorer clinical outcomes, including suicide. A better etiologic understanding of this comorbidity could inform diagnosis and treatment. The present study aims to test whether comorbid anxiety in bipolar disorder reflects shared genetic risk factors. We also sought to assess the contribution of genetic risk for anxiety to suicide attempts in bipolar disorder. Polygenic risk scores (PRS) were calculated from published genome-wide association studies of samples of controls and cases with anxiety (n = 83,566) or bipolar disorder (n = 51,710), then scored in independent target samples (totaln = 3369) of individuals with bipolar disorder who reported or denied lifetime anxiety disorders or suicidal attempts in research interviews. Participants were recruited from clinical and nonclinical settings and genotyped for common genetic variants. The results show that polygenic risk for anxiety was associated with comorbid anxiety disorders and suicide attempts in bipolar disorder, while polygenic risk for bipolar disorder was not associated with any of these variables. Our findings point out that comorbid anxiety disorders in bipolar disorder reflect a dual burden of bipolar and anxiety-related genes; the latter may also contribute to suicide attempts. Clinical care that recognizes and addresses this dual burden may help improve outcomes in people living with comorbid bipolar and anxiety disorders.

  • Anti-inflammatory Effects and Possible Mechanism of Action of Lupeol Acetate Isolated From Himatanthus Drasticus (Mart.) Plumel

    • Background: The species Himatanthus drasticus is popularly known in Northeast Brazil as "janaguba" and belongs to the family Apocynaceae. The latex collected from its stem bark is used for several purposes including anti-inflammatory properties and presents among its bioactive constituents the pentacyclic triterpene lupeol. The objective of the present work was to study in vivo and in vitro the lupeol acetate (LA) isolated from the plant latex, in several models of inflammation.Methods: Male Swiss mice (25-30 g, 6-24 animals per group) were administered with LA, 30 min before the test initiation. In the evaluation of analgesic activity the formalin test was used. The anti-inflammatory activity was evaluated by the following tests: paw edema induced by carrageenan and dextran, and the carrageenan-induced neutrophil migration into peritoneal cavities. Furthermore, the effect of LA on the myeloperoxidase release (MPO, an inflammation biomarker) from human neutrophils was also determined, as well as its antioxidant potential by the DPPH assay.Results: In the formalin test, LA (10, 25 and 50 mg/kg, i.p.) inhibited both the 1(st) (neurogenic, 0-5 min) and mainly the 2(nd) (inflammatory, 20-25 min) phase. Naloxone completely reversed the LA effect, indicating the participation of the opioid system. LA also significantly inhibited carrageenan-and dextran-induced paw edemas, as well as the neutrophil migration to the peritoneal cavity evaluated by the carrageenan-induced pleurisia. In this model, the effect of a very low dose of LA (0.1 mg/kg) was potentiated by the same dose of pentoxifylline (PTX), a known TNF-alpha inhibitor. LA (25 and 50 mu g/ml) was also very effective in inhibiting MPO released from stimulated human neutrophils, and significantly decreased the number of cells expressing iNOS activity in the paw of mice submitted to carrageenan-induced edema, suggesting a drug involvement with the NO system.Conclusions: The anti-inflammatory effect of LA probably involves the opioid system, asindicated by the complete blockade of the opioid antagonist naloxone. Furthermore, the LA effect was potentiated by PTX (a TNFalpha inhibitor). LA also decreased the number of iNOS cells, suggesting the participation of pro-inflammatory cytokines and the NO system in the drug action.

  • The unstable fate of the planet orbiting the A star in the HD 131399 triple stellar system

    • Validated planet candidates need not lie on long-term stable orbits, and instability triggered by post-main-sequence stellar evolution can generate architectures which transport rocky material to white dwarfs, hence polluting them. The giant planet HD 131399Ab orbits its parent A star at a projected separation of about 50-100 au. The host star, HD 131399A, is part of a hierarchical triple with HD 131399BC being a close binary separated by a few hundred au from the A star. Here, we determine the fate of this system, and find the following: (i) Stability along the main sequence is achieved only for a favourable choice of parameters within the errors. (ii) Even for this choice, in almost every instance, the planet is ejected during the transition between the giant branch and white dwarf phases of HD 131399A. This result provides an example of both how the free-floating planet population may be enhanced by similar systems and how instability can manifest in the polluted white dwarf progenitor population.
