    • Protein reagents are essential resources for several stages of drug discovery projects from structural biology and assay development through lead optimization. Depending on the aim of the project different amounts of pure protein are required. Small-scale expressions are initially used to determine the reachable levels of production and quality before scaling up protein reagent supply. Commonly, amounts of several hundreds of milligrams to grams are needed for different experiments, including structural investigations and activity evaluations, which require rather large cultivation volumes. This implies that cultivation of large volumes of either transiently transfected cells or stable pools/stable cell lines is needed. Hence, a production process that is scalable, speeds up the development projects, and increases the robustness of protein reagent quality throughout scales. Here we present a protein production pipeline with high scalability. We show that our protocols for protein production in Chinese hamster ovary cells allow for a seamless and efficient scale-up with robust product quality and high performance. The flexible scale of the production process, as shown here, allows for shorter lead times in drug discovery projects where there is a reagent demand for a specific protein or a set of target proteins.

  • High throughput generation of a resource of the human secretome in mammalian cells

    • The proteins secreted by human tissues and blood cells, the secretome, are important both for the basic understanding of human biology and for identification of potential targets for future diagnosis and therapy. Here, a high-throughput mammalian cell factory is presented that was established to create a resource of recombinant full-length proteins covering the majority of those annotated as 'secreted' in humans. The full-length DNA sequences of each of the predicted secreted proteins were generated by gene synthesis, the constructs were transfected into Chinese hamster ovary (CHO) cells and the recombinant proteins were produced, purified and analyzed. Almost 1,300 proteins were successfully generated and proteins predicted to be secreted into the blood were produced with a success rate of 65%, while the success rates for the other categories of secreted proteins were somewhat lower giving an overall one-pass success rate of ca. 58%. The proteins were used to generate targeted proteomics assays and several of the proteins were shown to be active in a phenotypic assay involving pancreatic beta-cell dedifferentiation. Many of the proteins that failed during production in CHO cells could be rescued in human embryonic kidney (HEK 293) cells suggesting that a cell factory of human origin can be an attractive alternative for production in mammalian cells. In conclusion, a high-throughput protein production and purification system has been successfully established to create a unique resource of the human secretome.

  • On the way to a smart factory for single-family wooden house builders in Sweden

    • To remain competitive, single-family wooden house builders might create smart wooden house factories. Smart factories and smart production are recent concepts used to denote ideas that support competitiveness in for example the automotive industry. In the housing sector, a concept introduced to increase competitiveness is industrialized house-building. The aim of the study presented here was to investigate whether industrialized house-building characteristics could be a step toward a smart factory for wooden house builders. In this paper, ideas of the smart wooden house factory are synthesized, and challenges for the single-family wooden house builder are identified. The study is founded on empirical data from one single-case study. A challenge identified was the maturity level of the culture and organization to commit to and utilize modern technology and modern working methods. The concept of the smart wooden house factory is still in its infancy and needs further research.

  • Revealing the content of Industry 4.0 : A review of literature

    • A fourth industrial revolution is prophesied, and there is a potential for the industrialized world to proactively adapt suitable practices. Despite the large interest from both industry and academia, a drawback with the vast literature on initiatives that tap into the fourth industrial revolution, Industry 4.0 and alike, is the fuzziness when it comes to terminology and content. The terms are mixed up, and sometimes used interchangeable and the constituent parts are not fully described. The purpose of this paper is to present the content of initiatives related to the fourth industrial revolution in a structured manner. This is expected to support understanding for the content of the fourth industrial revolution and thereby facilitate the transformation. The results presented in this paper is based on a traditional literature review. In total 13 relevant review papers were identified. The identified papers were analyzed, and a framework was developed including technologies and design principles. In total, eleven technologies and twelve design principles were identified for Industry 4.0. The most frequently occurring technologies were Cyber physical systems, Internet of Things, and Big data. The most frequently occurring design principles were Smart factory, Service orientation and Sustainability and resource efficiency. A categorization of the content into technologies and design principles clarify and structures the content of Industry 4.0. The developed framework can support academics in identifying, describing, and selecting Industry 4.0 scenarios for further investigations. For practitioners, the framework can give a basic understanding and some guidance in their implementation journey of Industry 4.0.

  • Smart factories for single-family wooden houses : a practitioner’s perspective

    • Purpose: The meaning of Industry 4.0 has started to be outlined for the construction industry, but there is still limited knowledge on the implications for the single-family wooden house building industry. The purpose of this paper is to expand the understanding of what the fourth industrial revolution implies for the single-family wooden house industry. The paper contributes with practitioners’ view of the content and meaning of a smart single-family wooden house factory.Design/methodology/approach: An exploratory multiple case study was carried out at two Swedish single-family wooden house builders, combined with a traditional literature review.Findings: As a result of a multiple case studies, the content and meaning of a smart single-family wooden house factory was elaborated on. In total, 15 components of a smart single-family wooden house factory were identified, of which 8 corresponded to the components of Industry 4.0 as described in other sectors.Research limitations/implications: The study can be expanded to also include multi-family wooden house builders and other branches of the offsite wooden building industry.Practical implications: Managers in the house-building industry who want to improve and strive for a smart single-family wooden house factory can learn from this study, get an insight of what other companies consider as important and how it relates to Industry 4.0.Originality/value: To the best of the authors’ knowledge, this study is a first attempt to understand what Industry 4.0 mean and how it can be accomplished for the single-family wooden house offsite manufacturing industry.

    • To meet the demand of future building requirements, and to improve productivity and competitiveness, there is a need to modernize and revise the current practices in the wooden single-family house industry. In several other sectors, intensive work is being done to adapt to the anticipated fourth industrial revolution. The manufacturing industry has already begun its transformation with concepts such as smart manufacturing and Industry 4.0. So far, smart manufacturing has not been discussed to any significant extent for the wooden single-family house industry, even though it might be a way for this industry to improve productivity and competitiveness.The research presented in this thesis aims at increased knowledge about what smart manufacturing means for the wooden single-family house industry. This requires investigating what smart wooden house manufacturingis, what challenges that might be associated with it, and how smart wooden house manufacturing can be realized. At the core of this thesis is the conceptualization of smart wooden house manufacturing—when realized, it is expected to contribute to improve the competitiveness of the wooden single family house industry.The findings presented here are based on three Research Studies. Two studies were case studies within the wooden single-family house industry. The third study was a traditional literature review.The findings revealed two definitions and 26 components of smart wooden house manufacturing. At large, smart wooden house manufacturing emphasizes digital transformation with a focus on digital information flow, how to add information, information compilation, and information distribution between systems/programs and departments. Some of the challenges associated with smart wooden house manufacturing are, e.g. culture, competence and manual transfer of information between systems.The findings indicate similarities of smart wooden house manufacturing within certain components of industrialized house building and Industry 4.0, these components could enable the realization of smart wooden house manufacturing.

  • Supporting smart manufacturing in industrialized house building through product data management

    • Industrialized house building (IHB) needs to improve efficiency and reduce costs to be able to meet the demand of future building requirements. The discrete manufacturing industry has already begun its transformation with concepts such as Industry 4.0 and smart manufacturing. The focus of these concepts is on systems integration and utilizing different design principles and technologies to improve productivity and competitiveness. So far, smart manufacturing has not been discussed to any significant extent in the context of the IHB industry, even though it might be a way for this industry to improve efficiency and reduce costs.The purpose of this thesis is to support smart manufacturing realization in the IHB industry through improved systems integration capability. The objective is to develop a support system that helps to mitigate current product data management challenges in the IHB industry. The research presented here followed the design research methodology (DRM), and three research studies were conducted within the IHB industry, specifically within the context of the wooden single-family house industry.The findings emphasized that integrated information and communication technology (ICT) tools are a precondition for realizing smart manufacturing in IHB. A pilot product lifecycle management (PLM) system was developed as a support system to mitigate the challenges encountered in product data management. The support system was developed based on five requirements (integrate product data between ICT tools, extract product data to purchasing items, adaptable to the product realization process, organize product data in different views, and trace revisions on building information modeling (BIM) projects and BIM families) and six functions (extract and interpret product data from the BIM project, property mapping, revision management, different types of document structures, from document structure to item structure, and complete bill of materials). The pilot PLM system was perceived to have value and could, for the case company, mitigate their challenges with product data management and facilitate improved systems integration. Furthermore, 18 enablers for PLM implementation in IHB were identified and categorized into implementing organization, PLM implementation process, PLM system selection and development, and development of product data for PLM.

  • Clinical Manifestations and Symptoms of Maxillary Sinusitis of Odontogenic Origin Demonstrated by Cone Beam Computed Tomography

    • Objectives: To compare the symptomatology of patients with maxillary sinusitis of dental origin (MSDO) with sinusitis due to upper respiratory tract infection (URTI) with a special focus on time to correct diagnosis. To define the accuracy of cone beam computed tomography (CBCT) in detecting the dental origin of the sinusitis.Methods: Retrospective review of the otolaryngology specialist care, primary health care and dental health care medical records of patients with maxillary sinusitis who has been referred for radiology. All patients were examined by CBCT, which has a better resolution regarding bony structures than low-dose computed tomography. To the best of our knowledge there is no previous study on this topic based on CBCT as diagnostic method.Results: Sixty-one patients were included in the study; of these, 25 had MSDO and 36 had URTI sinusitis. The MSDO patients more frequently reported foul odour and foul taste than patients with URTI sinusitis. The URTI sinusitis patients more frequently reported symptoms such as facial pain, facial congestion and cold-related symptoms. Both the time from the onset of symptoms to the first medical visit and the subsequent time to a correct diagnosis were significantly longer in the MSDO group. The accuracy of CBCT for detecting dental pathology as the underlying cause of sinusitis was 97%.Conclusion: The present study verifies that maxillary sinusitis of dental origin differs from viral-induced rhinosinusitis concerning symptomatology and clinical findings. Certain findings and symptoms could serve as valuable indicators of an underlying dental pathology, because although MSDO is well known, the present study shows that these patients are often misdiagnosed and the correct diagnose and treatment is often delayed several months. Consequently, better assessment is important. The suspicion of MSDO should be raised for patients with unilateral sinusitis presenting little pain, foul odour or foul taste and a long time course. These patients should be referred for radiology, preferably CBCT, to rule out odontogenic cause. CBCT is easy to perform for sinusitis examinations and has advances to common CT, especially regarding detection of pathology in bony structures as the periapical area. Because of this CBCT is a reliable tool in order to detect maxillary sinusitis of dental origin.

  • When Maxillary Sinusitis Does Not Heal : Findings on CBCT Scans of the Sinuses With a Particular Focus on the Occurrence of Odontogenic Causes of Maxillary Sinusitis

    • Objectives: This study sought to investigate the proportion of patients with suspected sinusitis referred for radiological examination who have radiologically verified sinusitis of odontogenic origin and to describe this type of sinusitis. Study Design: This investigation is a retrospective study. Methods: A total of 303 sinus examinations involving cone beam computed tomography (CBCT) performed at Sunderby Hospital, Lulea, Sweden in 2012 were independently reviewed by two radiologists. The number of cases of maxillary sinusitis and the correlation between maxillary sinusitis and odontogenic infections were determined. Results: Overall, 24% of the verified cases of sinusitis were odontogenic. An odontogenic origin was identified in 40% of unilateral maxillary sinusitis cases but only 6% of bilateral maxillary sinusitis cases (p=0.0015). Forty-nine out of 54 patients with periapical destruction had adjacent mucosal swelling in the maxillary sinus, but only 15 of these patients satisfied the criteria for sinusitis. Conclusion: The present study confirms the close relationship between odontogenic infections and unilateral maxillary sinusitis.
