    • Multivalency plays a major role in biological processes and particularly in the relationship between pathogenic microorganisms and their host that involves protein-glycan recognition. These interactions occur during the first steps of infection, for specific recognition between host and bacteria, but also at different stages of the immune response. The search for high-affinity ligands for studying such interactions involves the combination of carbohydrate head groups with different scaffolds and linkers generating multivalent glycocompounds with controlled spatial and topology parameters. By interfering with pathogen adhesion, such glycocompounds including glycopolymers, glycoclusters, glycodendrimers and glyconanoparticles have the potential to improve or replace antibiotic treatments that are now subverted by resistance. Multivalent glycoconjugates have also been used for stimulating the innate and adaptive immune systems, for example with carbohydrate-based vaccines. Bacteria present on their surfaces natural multivalent glycoconjugates such as lipopolysaccharides and S-layers that can also be exploited or targeted in anti-infectious strategies.

  • Early life microbial exposures shape the Crassostrea gigas immune system for lifelong and intergenerational disease protection

    • Background: The interaction of organisms with their surrounding microbial communities influences many biological processes, a notable example of which is the shaping of the immune system in early life. In the Pacific oyster, Crassostrea gigas, the role of the environmental microbial community on immune system maturation — and, importantly, protection from infectious disease — is still an open question.Results: Here, we demonstrate that early life microbial exposure durably improves oyster survival when challenged with the pathogen causing Pacific oyster mortality syndrome (POMS), both in the exposed generation and in the subsequent one. Combining microbiota, transcriptomic, genetic, and epigenetic analyses, we show that the microbial exposure induced changes in epigenetic marks and a reprogramming of immune gene expression leading to long-term and intergenerational immune protection against POMS.Conclusions: We anticipate that this protection likely extends to additional pathogens and may prove to be an important new strategy for safeguarding oyster aquaculture efforts from infectious disease

  • Genome-wide meta-analysis for Alzheimer's disease cerebrospinal fluid biomarkers.

    • Amyloid-beta 42 (Aβ42) and phosphorylated tau (pTau) levels in cerebrospinal fluid (CSF) reflect core features of the pathogenesis of Alzheimer's disease (AD) more directly than clinical diagnosis. Initiated by the European Alzheimer & Dementia Biobank (EADB), the largest collaborative effort on genetics underlying CSF biomarkers was established, including 31 cohorts with a total of 13,116 individuals (discovery n=8074; replication n=5042 individuals). Besides the APOE locus, novel associations with two other well-established AD risk loci were observed; CR1 was shown a locus for Aβ42 and BIN1 for pTau. GMNC and C16orf95 were further identified as loci for pTau, of which the latter is novel. Clustering methods exploring the influence of all known AD risk loci on the CSF protein levels, revealed 4 biological categories suggesting multiple Aβ42 and pTau related biological pathways involved in the etiology of AD. In functional follow-up analyses, GMNC and C16orf95 both associated with lateral ventricular volume, implying an overlap in genetic etiology for tau levels and brain ventricular volume.

    • We present ALMA band 7 data of the extreme OH/IR star, OH 26.5+0.6. In addition to lines of CO and its isotopologues, the circumstellar envelope also exhibits a number of emission lines due to metal-containing molecules, e.g., NaCl and KCl. A lack of C18O is expected, but a non-detection of C17O is puzzling given the strengths of H217O in Herschel spectra of the star. However, a line associated with Si17O is detected. We also report a tentative detection of a gas-phase emission line of MgS. The ALMA spectrum of this object reveals intriguing features which may be used to investigate chemical processes and dust formation during a high mass-loss phase.

  • Histopathological growth patterns of liver metastasis : updated consensus guidelines for pattern scoring, perspectives and recent mechanistic insights

    • The first consensus guidelines for scoring the histopathological growth patterns (HGPs) of liver metastases were established in 2017. Since then, numerous studies have applied these guidelines, have further substantiated the potential clinical value of the HGPs in patients with liver metastases from various tumour types and are starting to shed light on the biology of the distinct HGPs. In the present guidelines, we give an overview of these studies, discuss novel strategies for predicting the HGPs of liver metastases, such as deep-learning algorithms for whole-slide histopathology images and medical imaging, and highlight liver metastasis animal models that exhibit features of the different HGPs. Based on a pooled analysis of large cohorts of patients with liver-metastatic colorectal cancer, we propose a new cut-off to categorise patients according to the HGPs. An up-to-date standard method for HGP assessment within liver metastases is also presented with the aim of incorporating HGPs into the decision-making processes surrounding the treatment of patients with liver-metastatic cancer. Finally, we propose hypotheses on the cellular and molecular mechanisms that drive the biology of the different HGPs, opening some exciting preclinical and clinical research perspectives.

  • Evidence for a chemically differentiated outflow in Mrk 231

    • Aims. Our goal is to study the chemical composition of the outflows of active galactic nuclei and starburst galaxies. Methods. We obtained high-resolution interferometric observations of HCN and HCO+ J = 1 -> 0 and J = 2 -> 1 of the ultraluminous infrared galaxy Mrk 231 with the IRAM Plateau de Bure Interferometer. We also use previously published observations of HCN and HCO+ J = 1 -> 0 and J = 3 -> 2, and HNC J = 1 -> 0 in the same source. Results. In the line wings of the HCN, HCO+, and HNC emission, we find that these three molecular species exhibit features at distinct velocities which differ between the species. The features are not consistent with emission lines of other molecular species. Through radiative transfer modelling of the HCN and HCO+ outflow emission we find an average abundance ratio X(HCN) = X(HCO+) greater than or similar to 1000. Assuming a clumpy outflow, modelling of the HCN and HCO+ emission produces strongly inconsistent outflow masses. Conclusions. Both the anti-correlated outflow features of HCN and HCO+ and the different outflow masses calculated from the radiative transfer models of the HCN and HCO+ emission suggest that the outflow is chemically differentiated. The separation between HCN and HCO+ could be an indicator of shock fronts present in the outflow, since the HCN/HCO+ ratio is expected to be elevated in shocked regions. Our result shows that studies of the chemistry in large-scale galactic outflows can be used to better understand the physical properties of these outflows and their effects on the interstellar medium in the galaxy.

  • A strong magnetic field in the jet base of a supermassive black hole

    • Active galactic nuclei (AGN) host some of the most energetic phenomena in the universe. AGN are thought to be powered by accretion of matter onto a rotating disk that surrounds a supermassive black hole. Jet streams can be boosted in energy near the event horizon of the black hole and then flow outward along the rotation axis of the disk. The mechanism that forms such a jet and guides it over scales from a few light-days up to millions of light-years remains uncertain, but magnetic fields are thought to play a critical role. Using the Atacama Large Millimeter/submillimeter Array (ALMA), we have detected a polarization signal (Faraday rotation) related to the strong magnetic field at the jet base of a distant AGN, PKS 1830-211. The amount of Faraday rotation (rotation measure) is proportional to the integral of the magnetic field strength along the line of sight times the density of electrons. The high rotation measures derived suggest magnetic fields of at least tens of Gauss (and possibly considerably higher) on scales of the order of light-days (0.01 parsec) from the black hole.

    • We report Atacama Large Millimeter Array (ALMA) Band 6 full-polarization observations of the lensed blazar PKS 1830-211 during its record-breaking radio and gamma-ray flare in the spring of 2019. The observations were taken close to the peak of the gamma activity and show a clear difference in polarization state between the two time-delayed images. The leading image has a fractional polarization about three times lower than the trailing image, implying that significant depolarization occurred during the flare. In addition, we observe clear intra-hour variability of the polarization properties between the two lensed images, with a quasi-linear increase in the differential electric-vector position angle at a rate of about two degrees per hour, associated with changes in the relative fractional polarization of ∼10%. This variability, combined with the lower polarization close to the peak of gamma activity, is in agreement with models of magnetic turbulence to explain polarization variability in blazar jets. Finally, the comparison of results from the full and differential polarization analysis confirms that the differential polarization technique (Martí-Vidal et al. 2016, A&A, 593, A61) can provide useful information on the polarization state of sources like gravitationally lensed radio-loud quasars.

    • Current mm / submm interferometers, like the Atacama Large mm / submm Array (ALMA), use receivers that register the sky signal in a linear polarization basis. In the case of observations performed in full-polarization mode (where the cross-correlations are computed among all the polarization channels) it is possible to reconstruct the full-polarization brightness distribution of the observed sources, as long as a proper calibration of delay off sets and leakage among polarization channels can be performed. Observations of calibrators, preferably with some linear polarization, with a good parallactic angle coverage are usually needed for such a calibration. In principle, dual-polarization observations only allow us to recover the Stokes I intensity distribution of the sources, regardless of the parallactic angle coverage of the observations. In this paper, we present a novel technique of dual differential polarimetry that makes it possible to obtain information related to the full-polarization brightness distribution of the observed sources from dual-polarization observations. This technique is inspired in the Earth-rotation polarization synthesis and can be applied even to sources with complex structures.
